目的:探讨黄芪-丹参药对通过调节微小核糖核酸-466b-5P(miR-466b-5p)改善高血压肾损害的机制。方法:基于微RNA(miRNA)测序寻找自发性高血压大鼠与Wistar-京都鼠(WKY)的差异基因,构建腺相关病毒转染小鼠,24只C57BL/6小鼠,随机选取12只尾静脉注射小鼠腺相关病毒miR-466b-5P(rAAV-miR-466b-5P)构建miR-466b-5p过表达组,余12只作为空白对照组注射腺相关病毒-9(AAV-9)空载体,转染6周后再各随机分为2组,每组6只,A组(黄芪-丹参+miR-466b-5P过表达组)和C组(黄芪-丹参+空白对照组)以黄芪配方颗粒:2.036 g/kg+丹参配方颗粒:0.255 g/kg灌胃;B组(miR-466b-5P过表达组)和D组(空白对照组)以相同体积生理盐水灌胃;灌胃28 d后观察各组小鼠尿β2-微球蛋白(β2-MG)、N-乙酰-β-D-葡萄糖苷酶(NAG)、微量白蛋白(mALB),血清胱抑素C(Cys-C)、血管紧张素Ⅱ(AngⅡ)、C反应蛋白(CRP)水平和肾脏病理,进行实时荧光定量(RT-qPCR)与蛋白质印迹法(Western Blotting)寻找其靶基因。结果:1)基于前期miRNA测序选定了miR-466b-5p;2)与D组比较,B组小鼠尿β2-MG、NAG、mALB、血清Cys-C、AngⅡ、CRP均显著升高(P<0.01);与B组比较,A组小鼠血清Cys-C、CRP明显升高(P<0.05),尿β2-MG、NAG、mALB、血清AngⅡ无明显改变(P>0.05);3)与D组比较,B组小鼠出现明显肾小球硬化、小管纤维化,A组较B组有明显改善;4)与D组比较,B组小鼠透明质酸酶2(HAS2)明显下调,而经黄芪-丹参干预后有所回调。结论:miR-466b-5p的过表达会导致高血压肾损害的发生,黄芪-丹参药对通过靶向HAS2下调miR-466b-5p改善高血压肾损害。  相似文献

MicroRNAs (miRNAs) and long noncoding RNAs (lncRNAs) are both types of noncoding RNA. They have been demonstrated to be involved in the regulation of various human inflammatory diseases and can be used as biomarkers for disease diagnosis and prognosis, and even be developed into new drugs. Gout is an arthritic disease caused by the deposition of monosodium urate crystal (MSU) in the joints, which can lead to acute inflammation and damage adjacent tissue. Recent studies have shown that miRNAs and lncRNAs mediate the progress of gout. Based on the pathogenesis of gout, including hyperuricemia, MSU deposition, acute gouty arthritis and gouty bone erosion, this paper reviewed the role of miRNAs and lncRNAs in the processes and the possible therapeutic targets of miRNAs and lncRNAs in gout.  相似文献

MiRNA(MicroRNA)是近年来在多种真核细胞及病毒中发现的一类内源性21-25nt短序列非编码单链RNA,在进化上具有高度的保守性,能够通过与靶mRNA特异性的碱基互补配对,引起靶mRNA的降解或者抑制其翻译,在基因调控中扮演重要的角色。本文试从MiRNA的概念特点作用机制及功能等方面对MiRNA的研究进展作一个简单回顾。  相似文献

MicroRNAs (miRNAs), small non-coding RNA molecules, are negative regulators of gene expression. Recent studies have indicated their role in various forms of cardiovascular disease. In spite of the number of miRNA microarray analyses performed, little is known about the genome-wide miRNA expression pattern in human myocardial infarction (MI). Using miRNA microarrays and bioinformatic analysis, miRNA expression was analyzed on human MI and foetal hearts compared to healthy adult hearts, to determine whether there is any similar expression pattern between MI and foetal hearts, and to identified miRNAs that have not previously been described as dysregulated in cardiovascular diseases. Of 719 miRNAs analyzed, ∼ 50% were expressed in human hearts, 77 miRNAs were absent from all tested tissues and 57 were confidently dysregulated in at least one tested group. Some expression patterns appeared to be similar in MI and foetal hearts. Bioinformatic analysis revealed 10 miRNAs as dysregulated in MI not yet related to cardiovascular disease, and 5 miRNAs previously described only in animal models of cardiovascular diseases. Finally, qRT-PCR analysis confirmed dysregulation of 7 miRNAs, miR-150, miR-186, miR-210, miR-451, and muscle-specific, miR-1 and miR-133a/b; all of these are believed to be involved in various physiological and pathological processes.  相似文献

Senecio spp. is one of the most frequent plant-related poisonings in cattle. Its ingestion generates the disease seneciosis, characterized by hepatic damages. Liver biopsies and serum markers dosage are tools used in diagnosis; however, many breeding cattle are undiagnosed. MicroRNAs are non-coding RNA, stable in biological fluids. Their difference in expression levels may indicate the presence of the poisoning. We analyzed the miRNA profiling to identify potential diagnostic biomarkers for Senecio brasiliensis poisoning. The expression of miR-21, miR-885, miR-122, miR-181b, miR-30a, miR-378, and let-7 f were evaluated in the serum of exposed cattle. At least one histological change was found in liver and lower quantity of albumin and high AST and ALP were also detected. MiRNAs miR-30a, miR-378, miR-21, miR-885, and miR-122 presented significantly higher expression in intoxicated animals than in healthy animals. Furthermore, miR-122, miR-885, and, especially, miR-21 signatures demonstrated high sensitivity and specificity, with potential application for detecting poisoning.  相似文献

MicroRNAs have become recognized as key players in the development of cancer. They are a family of small non-coding RNAs that can negatively regulate the expression of cancer-related genes by sequence-selective targeting of mRNAs, leading to either mRNA degradation or translational repression. Lung cancer is the leading cause of cancer-related death worldwide with a substantially low survival rate. MicroRNAs have been confirmed to play roles in lung cancer development, epithelial-mesenchymal transition and response to therapy. They are also being studied for their future use as diagnostic and prognostic biomarkers and as potential therapeutic targets. In this review we focus on the role of dysregulated microRNA expression in lung tumorigenesis. We also discuss the role of microRNAs in therapeutic resistance and as biomarkers. We further look into the progress made and challenges remaining in using microRNAs for therapy in lung cancer.  相似文献
