We report three patients with Feingold 2 syndrome with the novel features of growth hormone deficiency associated with adenohypophyseal compression, aortic dilation, phalangeal joint contractures, memory, and sleep problems in addition to the typical features of microcephaly, brachymesophalangy, toe syndactyly, short stature, and cardiac anomalies. Microdeletions of chromosome 13q that include the MIR17HG gene were found in all three. One of the patients was treated successfully with growth hormone. In addition to expanding the phenotype of Feingold 2 syndrome, we suggest management of patients with Feingold 2 syndrome include echocardiography at the time of diagnosis in all patients and consideration of evaluation for growth hormone deficiency in patients with short stature. 相似文献

Monoclonal antibodies to three distinct epitopes on human IgE: their use for determination of allergen-specific IgE 总被引:7,自引:0,他引:7
Three different monoclonal antibodies (MAb) against human immunoglobulin E have been obtained which specifically bind to human myeloma and polyclonal IgE. The antibodies showed high avidities for soluble IgE (0.7 X 10(9) to 3.3 X 10(9) M-1). These MAb defined three distinct epitopes on IgE. A mixture of these antibodies in combination with an 125I-labelled anti-mouse Kappa chain MAb has been used to measure allergen-specific IgE. This determination was performed by a solid-phase radioimmunoassay using allergen extracts coated to either chemically activated paper discs or to polyvinyl chloride wells. This method is 4-10 times more sensitive than other previously reported procedures. A similar technique has also been applied to detect individual allergens in immunoblots of allergen extracts. 相似文献

目的以反相高效液相色谱法(RP-HPLC)测定甘露药浴颗粒(外用)中盐酸麻黄碱的含量。方法色谱柱:WatersC18(150mm×4.6mm);流动相:水-乙烷-十二烷基硫酸钠-磷酸(650∶350∶5∶1);流速:1.0mL/min;柱温:26℃;检测波长:210nm。结果该方法的线性范围为0.127~0.130μg/μL(r=0.9996,n=5),平均回收率为99.72%,RSD=1.12%。结论本法简便、准确、重现性好,可用于该制剂的质量控制。 相似文献

目的观察妇科止血消痛颗粒对产后子宫复旧不全大鼠子宫组织基质金属蛋白-9(MMP-9)、组织金属蛋白酶抑制物-1(TIMP-1)表达的影响,探讨妇科止血消痛颗粒治疗产后子宫复旧不全的机制。方法建立大鼠产后子宫复旧不全病理模型,随机分为模型组、产妇康组及妇科止血消痛颗粒高、中、低剂量组。造模第1日开始各给药组分别灌胃相应药物,空白组、模型组予蒸馏水灌胃。均每日1次,连续7d。采用原位杂交法检测各组大鼠子宫组织中MMP-9、TIMP-1基因的表达水平。结果各组大鼠子宫内膜中均可检测到MMP-9和TIMP-1的杂交信号。模型组MMP-9表达强度明显超过空白组,TIMP-1表达弱于空白组。妇科止血消痛颗粒高、中、低剂量组及产妇康组,MMP-9 mRNA的表达明显降低(P〈0.05,P〈0.01);妇科止血消痛颗粒中、低剂量组与产妇康组MMP-9 mRNA的表达强度相似,但均低于妇科止血消痛颗粒高剂量组的表达强度(P〈0.01)。妇科止血消痛颗粒高剂量组及产妇康组TIMP-1 mRNA的表达明显升高(P〈0.05),且表达相似。结论妇科止血消痛颗粒通过抑制MMP-9的高表达,升高TIMP-1的低表达,从而发挥治疗产后子宫复旧不全的作用。 相似文献

目的建立清咽灵颗粒的定性定量方法。方法采用薄层色谱法鉴别麦冬、桔梗;高效液相法测定哈巴俄苷的含量,色谱柱:eclipse XDB-C18柱(150 mm×4.6 mm,5μm);流动相:乙腈-0.03%磷酸溶液(25∶75);检测波长:278 nm;流速:1.0 mL.min-1;柱温:30℃。结果薄层斑点清晰,阴性样品无干扰;哈巴俄苷含量测定在0.091 04~0.910 4μg与峰面积呈良好的线性关系。回归方程为Y=3.650×10-4X+3.147×10-3,r=0.999 9(n=5),平均回收率为101.8%,RSD=1.5%(n=9)。结论本方法快速、简便、准确,专属性强,重现性好,可作为清咽灵颗粒质量控制的方法。 相似文献

目的:建立氢化物原子荧光法测定溪黄草中硒含量的方法。方法:样品经1+1的硝酸-高氯酸消解完全、6.0mol/L的盐酸预还原后通过氢化物原子荧光法进行测定。结果:方法的检出限为0.02mg/kg,精密度为5.7%,回收率为86%-105%。结论:采用氢化物原子荧光法测定溪黄草中的硒,方法污染少、灵敏度高、准确度较好。 相似文献

目的:建立反相高效液相色谱法同时测定小儿抗病毒颗粒中黄芩苷和橙皮苷含量的方法。方法:色谱柱为Shim—packVP—ODSC18柱(150mm×4.6mm,5μm);流动相为乙腈-水-磷酸(21:79:0.1);流速1.0mE/min;检测波长:283nm。结果:橙皮苷在0.260~0.520μg范嗣内、黄芩苷在0.270—0.720μg范围内与峰面积呈良好的线性关系(橙皮苷r=0.99997、黄芩苷r=0.99996),平均回收率分别为99.37%和101.8%;RSD分别为1.52%和0.63%(n=6)。结论:该方法简便,结果准确,重现性好,可作为该制剂的质量控制方法。 相似文献

PurposeAcute kidney injury (AKI) is a serious complication of sepsis and is characterized by inflammatory response. MicroRNA-210 host gene (MIR210HG) is upregulated in human proximal tubular epithelial cells under treatment of inflammatory cytokines. This study aimed to explore the role of MIR210HG in sepsis-induced AKI.Materials and MethodsCell viability was detected by a cell counting kit 8 assay. The levels of proinflammatory cytokines were detected by enzyme-linked immunosorbent assay kits. The protein levels of p65, IκBα, and p-IκBα were examined by western blot analysis. The nuclear translocation of nuclear factor kappa B (NF-κB) was detected by immunofluorescence assay. The histological changes of kidneys were analyzed by hematoxylin and eosin staining assay.ResultsLipopolysaccharide (LPS) treatment significantly inhibited cell viability and increased productions of proinflammatory cytokines in proximal tubular epithelial cells (HKC-8). Additionally, MIR210HG levels in HKC-8 cells were increased by LPS treatment. MIR210HG silencing inhibited the LPS-induced cell inflammatory response. MIR210HG activated the NF-κB signaling pathway by promoting the phosphorylation of IκBα and nuclear translocation of p65. Rescue assays revealed that the MIR210HG-induced increase of cytokines levels and decline of cell viability were rescued by QNZ treatment. Knockdown of MIR210HG decreased blood urea nitrogen, serum creatinine, and proinflammatory cytokine levels in AKI rats. Moreover, the knockdown of MIR210HG protected against AKI-induced histological changes of kidneys in rats.ConclusionMIR210HG promotes sepsis-induced inflammatory response of HKC-8 cells by activating the NF-κB signaling pathway. This novel discovery may be helpful for the improvement of sepsis-induced AKI. 相似文献
