目的:研究脑缺血后脑片[Ca2+]i变化。方法:采用新型Ca2+荧光指示剂Fura-2双波长法测定兔大脑中动脉阻塞(MCAo)局灶脑缺血后脑片细胞内游离钙([Ca2+]i)。结果:脑缺血后脑组织[Ca2+]i显著升高。结论:[Ca2+]i在脑缺血损害中起重要作用。  相似文献

目的研究S波段高功率微波(HPM)辐射后乳鼠心肌细胞的损伤效应及其机制。方法采用平均功率密度为5~30mW/cm^2的S波段HPM模拟源辐射原代培养的乳鼠心肌细胞,应用流式细胞术、原子力显微镜观察辐射后心肌细胞凋亡和坏死率、[Ca^2+]。及细胞膜形态。结果10—30mW/cm^2的S波段HPM辐射后6h,心肌细胞凋亡和坏死率增加(P〈0.05或P〈0.01),且随辐射剂量增大,坏死细胞增多;30mW/cm^2 HPM辐射后即刻[Ca^2+]i升高(P〈0.01),细胞膜发生穿孔。结论10—30mW/cm^2的S波段HPM辐射可造成心肌细胞凋亡和坏死,[Ca^2+];升高以及细胞膜穿孔,是S波段HPM辐射致心肌细胞损伤的重要机制。  相似文献

In this retrospective study 103 serum samples from 16 females with systemic lupus erythematosus (SLE), obtained during a mean follow-up time of 2 years, were investigated for the presence of anti-denatured [single-stranded (ss)] DNA antibodies of the IgG, IgM, and IgA classes. The anti-ssDNA antibodies were determined by an enzyme-linked immunosorbent assay (ELISA), and the results were expressed in three ways: as units derived from a single serum dilution and as two parameters,E andA, calculated from the dose-response curve,E being an estimate of the effective amount of antibodies andA a function of the reaction constant between the antigen and the antibody. The simultaneous occurrence of anti-ssDNA antibodies of all three immunoglobulin classes was seen most often in the patients with the shortest duration of the disease. Clinically active disease was found to correlate with high reaction constants of the IgA anti-ssDNA antibodies. There was also an association between the IgA anti-ssDNA antibody levels and the presence of nephritis. Great fluctuations in the amounts of effective antibodies of the IgG class were seen in seven patients, in six of whom changes in the disease activity also were seen. Changes in the disease activity were unaccompanied by fluctuations in the IgG anti-ssDNA levels in four patients; two of these patients were positive for antibodies against extractable nuclear antigens. We conclude that it is of value to express the results of the anti-ssDNA ELISA as a function of the dose-response curve when monitoring patients with SLE and that immunoglobulin class-specific determinations of anti-ssDNA antibodies may provide information about the disease activity in many patients with SLE.  相似文献

The Thy-1 antigens or rat brain and thymus have been isolated and chemically characterized, but those of mice have not been identified. Moreover, it is uncertain whether the antigens are glycolipids or glycoproteins. This study with highly purified preparations of gangliosides GM1, 1GD1a, GD1b and GT1b from bovine brain and several ganglioside fractions from mouse brain showed that Thy-1 activity does not reside in gangliosides, but rather in the chloroform-methanol-insoluble residue of brain remaining after extraction of gangliosides. The antigen could be solubilized from this residue with a non-ionic detergent. The antigenic activity of the solubilized preparation was heat-labile but resistant to periodate. The chemical properties of the Thy-1 antigen of mouse brain are discussed.  相似文献

Summary: Blends of high molecular weight poly(R‐3‐hydroxybutyrate) (PHB) ( = 352 000 g · mol?1), comprising of either low molecular weight poly(R‐3‐hydroxybutyrate) (D‐PHB) ( = 3 900 g · mol?1) or poly[(R‐3‐hydroxybutyrate)‐co‐(R‐3‐hydroxyvalerate)] (PHBV) ( = 238 000 g · mol?1) with 12 mol‐% hydroxyvalerate (HV) content as a second constituent, were investigated along with the thermal properties and morphologies. After isothermal crystallization, a lowering of the melting temperature of PHB can be observed with increasing content of the second component in the blends. This behavior points towards miscibility of the constituents both in the liquid and the solid state. Crystallization kinetics was studied under isothermal and non‐isothermal conditions. The overall kinetics of isothermal crystallization was analyzed in terms of the Avrami equation. Only one crystallization peak is observed in all cases for the PHB/D‐PHB and PHB/PHBV blends under the conditions studied. This demonstrates co‐crystallization of the constituents. The addition of D‐PHB or PHBV to PHB reduces the rate of crystallization of the blends compared to that of neat PHB. The corresponding activation energies of crystallization also decrease with an increasing concentration of the second constituent. Non‐isothermal crystallization, carried out with different cooling rates held constant, is discussed in terms of a quasi‐isothermal approach. The corresponding rate constants as functions of reciprocal undercooling show Arrhenius‐like behavior in a certain range of temperatures. At sufficiently high undercooling, the rate constants of crystallization for the isothermal process exceed those reflecting non‐isothermal conditions, whereas in the limit of low undercoolings, the rate constants become similar. Ring‐banded morphologies are observed when PHB is in excess. When the respective second component is the major component, fibrous textures of the spherulites develop.

萘哌地尔衍生物(BWYJ)对血管平滑肌细胞游离钙浓度的影响   总被引:1,自引:0,他引:1  
目的:观察萘哌地尔衍生物(BWYJ)对家兔血管平滑肌细胞游离钙浓度的影响,对其血管活性进行机理探讨,进一步明确其作用机制。方法:采用钙荧光指示剂Fura-2/AM负载的培养家兔胸主动脉平滑肌细胞,观察该药对NA、5-HT和高钾所致的[Ca2 ]i的升高的影响。结果:Fura-2/AM负载血管平滑肌细胞的实验中,静息时各浓度的BWYJ对[Ca2 ]i无明显影响,但其对NA和5-HT所引起的血管平滑肌[Ca2 ]i增加均有明显的抑制作用,而不影响高钾所致血管平滑肌[Ca2 ]i的增加。结论:BWYJ通过阻断细胞膜上的α1受体或5-HT2A受体,抑制这些受体中介的钙内流,从而抑制细胞内Ca2 的释放,使血管平滑肌[Ca2 ]i降低。  相似文献

目的:观察杞菊胶囊对自发性高血压大鼠左室肥厚的防治作用,并从心肌细胞内钙离子浓度([Ca^2+]i)与钙调神经磷酸酶(CaN)角度探讨其作用机制.方法:以自发性高血压大鼠为左室肥厚模型,观察杞菊胶囊对左室重量、心肌[Ca^2+]i与 CaN活性的影响.结果:杞菊胶囊可防治自发性高血压大鼠左室肥厚并降低心肌细胞内钙离子浓度与钙调神经磷酸酶活性.结论:杞菊胶囊可防治心肌肥厚,其机制与降低心肌钙浓度及钙调神经磷酸酶活性有关.  相似文献

目的:观察毛茛提取液含药血清对培养家兔主动脉平滑肌细胞内游离钙浓度的影响.方法:体外培养家兔主动脉平滑肌细胞(VSMC),用荧光指示剂INDO-AM负载细胞,灌胃给予不同浓度药物,观察不同稀释浓度含药血清及不同时相药物血清对由NE诱导细胞内游离钙浓度([Ca2 ]i)增加的影响.结果:毛茛提取液各含药血清组均能降低由NE诱发的细胞内[Ca2 ]i的升高.结论:毛茛提取液含药血清能减少VSMC细胞内[Ca2 ]i.  相似文献

目的  观察复方利多卡因乳膏(EMLA)应用于宫腔镜手术的麻醉效果。方法  择期将60例宫腔镜手术患者随机均分为对照组(Ⅰ组)和复方利多卡因组(Ⅱ组);手术前7 min,于宫颈口和内腔各涂抹2.5 g超声导膏和EMLA;术者消毒时续贯推注芬太尼1.0μg/kg和丙泊酚1.5~2.5 mg/kg至睫毛反射消失,泵注丙泊酚4~6 mg/(kg·h)维持麻醉。观察T0、T1、T2及T3时MAP、HR、SpO2、BIS和丙泊酚总用量、术后VAS评分、手术时间、苏醒时间以及术中体动。结果  ①血流动力学:T2及T3时,Ⅰ组较Ⅱ组高(P <0.05);麻醉开始后,Ⅰ组内T1最低(P <0.05)。②BIS:两组间比较差异无统计学意义;麻醉后,两组BIS都降低,但Ⅱ组中,T3较T2及T1高(P <0.05)。③其他:两组丙泊酚总用量、VAS、苏醒时间以及术中体动比较差异有统计学意义(P <0.05)。结论  在宫腔镜手术静脉麻醉中,局部应用EMLA能增强麻醉效果,有利于保持生命体征平稳及苏醒。
