Cell growth and terminal differentiation are controlled by complex signaling cascades that regulate the expression of specific subsets of genes implicated in cell fate and morphogenic processes. We have recently cloned and characterized a novel Ste20-like kinase termed SLK that is associated with adhesion structures during cell adhesion and spreading. However, the specific function of SLK is poorly understood. To gain further insight into the role of SLK, we have characterized its activity, expression, and distribution in skeletal muscle and during the in vitro differentiation of C2C12 myoblasts. Although SLK is expressed ubiquitously in adult tissues, our results show that it is predominantly expressed in muscle masses during development. Furthermore, SLK activity is upregulated during the differentiation of C2C12 myoblasts. In addition, we have found that SLK localizes presynaptically at neuromuscular junctions and that it is preferentially expressed in types I and IIA myofibers at major myofibrillar striations. Supporting a role in myoblast function and differentiation, SLK expression is induced in Myf5- and Pax7-positive activated satellite cells during regeneration and expression of dominant negative SLK in C2C12 cultures impairs myoblast fusion, suggesting a role for SLK in muscle cell differentiation.  相似文献

神乐康(SLK)(7,3.5,1.75g/kg,po)能显著增加大鼠脑内边缘区的去甲肾上腺素(NA)、多巴胺(DA)及DA和5-羟色胺(5-HT)的代谢产物3,4二羧苯乙酸(DOPAC)、高香草酸(HVA)、5-羟吲哚乙酸(5-HIAA)的含量;SLK能增加大鼠脑内纹状体的DA及DA代谢产物HVA的含量,亦能升高DOPAC和5-HIAA的水平。  相似文献

Thirty-five patients with primary or radiationinduced hyperparathyroidism underwent autotransplantation with fresh diseased parathyroid tissue. Due to previous neck surgery (34 percent), concomitant thyroid surgery (66 percent), and a high proportion of multiglandular parathyroid disease (43 percent), all patients ran a high risk of becoming hypoparathyroid. At follow-up, a mean of 28 months after operation, 32 patients had normal parathyroid function, 2 had hyper- and 1 had hypoparathyroid function. The amount of tissue used for transplantation was individually chosen by relating the serum calcium level to the total parathyroid mass. In patients with water-clear cell hyperplasia and thus low endocrine activity on a weight basis, large grafts were needed, whereas less than 120 mg was used in most other cases. In two of our three failures we did not follow these tactics. Our results show that autografting fresh diseased parathyroid tissue can minimize the occurrence of postoperative hypoparathyroidism in patients at high risk of this complication. In no case has recurrent or progressive hypercalcemia developed.  相似文献

目的探讨溃疡性结肠炎的有效治疗方法。方法将40例溃疡性结肠炎(UC)患者随机分为2组,柳氮磺吡啶(SASP)加思连康(四联活菌片)治疗20例为治疗组,单用柳氮璜吡啶治疗20例为对照组。结果治疗组治愈率40%,有效率80%;对照组分别为10%和50%。治愈率与有效率治疗组均优于对照组。结论柳氮璜吡啶联合思连康治疗溃疡性结肠炎疗效明显。  相似文献

We simultaneously transplanted a liver and kidney (SLK) into a 55‐year‐old woman with end‐stage liver disease secondary to recurrent primary biliary cirrhosis. The patient was blood group O, the donor was A2 (A, non‐A1) and the patient's A isoagglutinin titer was 512. Good renal function was evident by normalization of her creatinine values following transplantation. Recovery was unremarkable and she was discharged on post op day 9. The patient has not experienced an episode of rejection in either organ during the 6 months of follow‐up. This case is important because high A IgG isoagglutinin levels (8 or higher) in kidney alone A2→ O transplantation are detrimental to outcome but do not affect outcome in liver alone A2→ O transplants; however, no such anti‐A titer data have been published for A2→ O (or B) SLK transplantation.  相似文献
