绿茶对小鼠Lewis肺癌生长的抑制及免疫调节   总被引:3,自引:0,他引:3  
为了研究茶叶防癌作用,以接种Lewis肺癌细胞的C57/BL6J小鼠为实验模型,研究绿茶对带瘤小鼠肿瘤生长的抑制及免疫调节作用。结果表明,接种Lewis肺癌细胞后,C57/BL6J小鼠胸腺重量及其指数减轻,T淋巴细胞CD4亚群阳性百分数及CD+4/CD+8比值下降,外周血白细胞本底化学发光降低,受酵母多糖刺激产生的化学发光增高以及脾IgM抗体生成细胞数减少。绿茶对小鼠体内Lewis肺癌的生长有明显抑制作用,并对上述各项指标的不良改变有不同程度的保护效果  相似文献

目的探讨Fas配体阳性睾丸细胞对共移植的胰岛细胞存活起全身和/或局部的免疫豁免作用。方法将同种大鼠胰岛细胞与不同数量的睾丸细胞同侧或两侧共移植于糖尿病受体肾包膜下,观察移植物功能和存活情况,以及移植物内淋巴细胞凋亡。结果单纯移植胰岛组(对照组)平均存活期为(4.6±1.1)天。睾丸细胞和胰岛细胞同侧共移植时,当睾丸细胞数为1×106(A组)时,胰岛细胞平均存活期为(23.8±4.6)天;睾丸细胞数增至1×107时(B组),胰岛细胞存活期大于(57.5±4.0)天,均较对照组明显延长(P<0.01)。两侧共移植时,当睾丸细胞数为1×105(C组),胰岛细胞平均存活期为(6.0±1.4)天,与对照组相似(P>0.05);睾丸细胞数增至1×106(D组),胰岛细胞存活期(11.5±3.1)天较对照组或C组延长(P<0.01);睾丸细胞数增至1×107(E组)时,胰岛细胞存活期(31.2±15.9)天延长更加明显(P<0.01)。相同剂量的睾丸细胞与胰岛细胞分别同侧和两侧共移植时发生移植排斥反应各自的病理表现是不同的,前者移植物内有淋巴细胞浸润,部分胰岛玻璃样变,抗胰岛素抗体染色仅见少量阳性细胞,而后者胰岛细胞移植物内有大量的淋巴细胞和少量的中性粒细胞浸润,抗胰岛素抗体阳性细胞较少,未见胰岛玻璃样变。原位细胞凋亡检测发现同侧共移植后的移植物与肾实质交界处有淋巴细胞凋亡。电镜检查发现胰岛移植物存活超过60d的标本可见大量存活的胰岛细胞团和睾丸Sertoli细胞,有散在的凋亡淋巴细胞。结论睾丸细胞与同种胰岛细胞移植后不仅可诱导局部免疫豁免,而且也具有一定的全身免疫保护作用,但较局部免疫豁免作用弱;这些作用可延长移植物的存活期,并具有剂量依赖性。  相似文献

Background The purpose of this study was to explore neuroretinal transplantation in a large animal model of severe retinitis pigmentosa and to establish graft development, long-term survival, graft-host integration, and effects on the host retina. Methods Rhodopsin transgenic pigs, aged 6 months, received in one eye a fetal full-thickness neuroretinal sheet in the subretinal space by means of vitrectomy and retinotomy. Six months postoperatively, eyes were studied in the light microscope and with immunohistochemical markers. Full-field electroretinography (ERG) was performed at 4 and 6 months. Results Laminated grafts with well-organized photoreceptors, rod bipolar cells, and Müller cells were found in five of six eyes. Neuronal connections between graft and host retina were not seen. In the five eyes containing a graft, the number of surviving rods in the host retina was significantly higher compared with unoperated eyes. The ERG did not reveal any significant difference in b-wave amplitude between operated and control eyes, but the cone-derived response in operated eyes increased significantly from 4 to 6 months while the rod response in control eyes decreased significantly. Conclusions Fetal full-thickness neuroretina can be transplanted safely to an eye with severe retinal degeneration. In their major part, the transplants develop a normal laminated morphology and survive for at least 6 months. Graft and host retinal neurons do not form connections. Retinal function in the host is reduced initially by the surgical trauma, but the presence of a well-laminated graft counteracts this effect and rescues rods from degeneration. 钴60放射法制备免疫功能抑制大鼠模型   总被引:1,自引:0,他引:1  
目的通过系统的钴60放射计划,对大鼠免疫功能抑制的合适剂量进行评估,为干细胞移植研究提供合适的的动物模型。方法SD大鼠随机分为12.5Gy、10Gy、7.5Gy、5Gy4个剂量组,10只/组。计算放射后2个月的死亡率。检测白细胞数量,评价各剂量组动物的白细胞参考值范围。结合死亡率和白细胞参考值范围,确定合适的放射剂量。通过对外周血中性粒细胞(PMN)吞噬实验和外周血白细胞移行抑制实验(MIT),对细胞免疫功能进行检测。结果行12.5Gy照射的大鼠,3d内全部死亡;10Gy放射后的大鼠在1个月内全部死亡;7.5Gy照射的大鼠2个月内死亡1只;5Gy照射的大鼠没有死亡情况发生。设定95%作为可信区间,计算白细胞数量参考值范围(109个/L),未放疗组:16.978+1.96×6.46;5Gy放疗组:4.93+1.96×0.72;7.5Gy放疗组:2.313+1.96×0.782;10Gy放疗组:1.03+1.96×0.507。t检验表明,随着放疗剂量的增加,各实验组的白细胞数量显著减少。对于细胞免疫功能的检测,外周血中性粒细胞(PMN)吞噬实验结果表明,机体全身免疫功能降低;外周血白细胞移行抑制实验(MIT)表明,机体T细胞免疫功能降低。结论7.5Gy可以一定程度上抑制大鼠的免疫功能,为合适的放疗剂量。  相似文献

Summary:  The eye is the prototypic immune-privileged organ. Its antigens were once believed to be expressed exclusively in the eye, which resides behind an efficient blood–organ barrier, and were believed to be unknown to the immune system. Self-tolerance to ocular components was therefore believed to be based not on immune tolerance but on immune ignorance. It is now known that the relationship between the immune system and the eye is much more complex. On the one hand, immune privilege is now known to involve not only sequestration but also active mechanisms that (i) inhibit innate and adaptive immune processes within the eye and (ii) shape the response that develops systemically to antigens released from the eye. On the other hand, retinal antigens are found in the thymus and have been shown to shape the eye-specific T-cell repertoire. However, thymic elimination of self-reactive T cells is incomplete, and such 'escapee' T cells are tolerized in the periphery as they recirculate through the body by encounter with self-antigen in healthy tissues. Due to the relative inaccessibility of the healthy eye to the immune system, peripheral tolerance mechanisms may not operate efficiently for ocular antigens, leaving a weak link in the homeostasis of tolerance. The case shall be made that although immune privilege protects vision by keeping the immune system at bay, a potential for developing destructive anti-retinal autoimmunity may be the price for the day-to-day protection afforded by immune privilege against inflammatory insults.  相似文献

Ffteen cases of peripheral T cell lymphoma were studied to evaluate the respective properties of various histologic types using enzyme histochemical and ultrastructural examinations in addition to immunological methods. Eleven cases in an ATLA negative group manifested various histologic patterns such as IBL like, pleomorphic and Lennert's lymphomas in comparison with the relatively monomorphic proliferation of neoplastic lymphoid cells in the 4 ATLA positive cases. The presence of neoplastic clear cells is characteristic of peripheral T-cell malignancies, and is likely to be found in CD4 lymphomas. There is an occasional reaction of epithelioid histiocytes and plasma cells with eosinophils, the former being designated Lennert's lymphoma and the latter IBL like T-cell lymphoma. Immunological examination revealed four immunophenotypic patterns: (1) CD2+3+4+8+, (2) CD2+ 34+8, (3) CD2+3+48+, and (4) CD2+3+4+8+, but did not provide information concerning the intimate relationship between histologic types and immuno phenotyes. β-Glucuronidase reactivity, however, contributed to the distinction between helper and suppressor T cell malignancies, suggesting its usefulness for distinguishing these two cell types and their malignant counterparts.  相似文献

Summary Recurrent and severe infections and absence of thymic shadow in X-ray examination were observed in children with the transposition of the great arteries (TGA). Among 45 children (29 boys and 16 girls) with TGA whose age ranged from 3 days to 16 years and who were hospitalized during 1 year, infectious diarrhea was observed in 77.7% cases, urinary tract infections in 44.5%, respiratory tract infections in 42.2%, sepsis in 17.5%, and meningitis in 8.8%. Nine of the children died, sepsis was a cause of death in seven children, and there were postsurgical complications in two children. Immunologic abnormalities in children with TGA included a decreased level of T-lymphocytes and T29° subpopulation, impaired mitogen-induced lymphoproliferation in vitro, and increased nitro blue tetrazolium (NBT) reduction activity of monocytes. Impaired parameters of cellular immunity correlated with worst clinical status. No disorders of humoral immunity were observed. These observations may be important for forming opinion about proper therapy and the cause of death in children with TGA.All results have been included in T. 目的:为了探讨细胞免疫功能及细胞因子在隐型孢子虫感染中的作用.方法:本文采用免疫分析技术对29例隐型孢子虫病患者外周血T细胞亚群,红细胞免疫功能及血清中IL-2(白细胞介素-2)、IL-6(白细胞介素-6)、sIL-2R(可溶性IL-2受体)、TNF-α(肿瘤坏死因子-α)及sICAM-1(可溶性细胞间粘附分子-1)等细胞因子水平进行测定.结果:患者外周血CD 4T细胞、CD4/CD8比值和红细胞表面C3b受体花环率(C3bRR)显著低于正常对照组(P<0.01),血清中IL-2、sIL-2R、IL-6、TNF-α及sICAM-1水平则显著高于正常对照(P<0.01).结论:隐孢子虫感染可致机体细胞免疫功能降低,细胞因子网络失衡.  相似文献
