Fascin蛋白在肿瘤中的表达及其临床应用价值研究进展

Fascin蛋白是一种肌动蛋白结合蛋白,主要表达于间叶组织和神经系统。近年来研究发现,很多人类肿瘤出现fascin蛋白的阳性表达,并且与肿瘤的侵袭转移以及患者的预后密切相关。而fascin基因在正常组织中却呈现低表达或不表达。本文讨论fascin基因编码的fascin蛋白表达上调对肿瘤细胞生物行为的意义并探讨其判断患者预后标记物和治疗靶点的前景。     

Fascin在乳腺浸润性导管癌中的表达及临床意义

目的检测Fascin在乳腺浸润性导管癌中的表达情况,并探讨其临床意义。方法用免疫组织化学法检测72例乳腺浸润性导管癌及配对癌旁组织中Fascin蛋白的表达和定位,以21例乳腺良性病变（10例乳腺纤维腺瘤、5例单纯性乳腺导管上皮增生和6例乳腺小叶增生）做为对照,收集病例临床病理资料,统计分析浸润性导管癌中Fascin的表达变化及与临床病理参数的相关性。结果Fascin蛋白在浸润性导管癌、癌旁乳腺组织、乳腺良性病变中的表达阳性率分别为26-39％（19／72）、0．14％（1／72）和0％（0／21）,浸润性导管癌中Fascin蛋白的表达明显高于癌旁乳腺及乳腺良性病变（P值均〈0．01）;浸润性导管癌中Fascin的表达与临床TNM分期、肿瘤病理分级和淋巴结转移相关（P值均〈0．05）;ER或PR阴性的浸润性导管癌病例Fascin表达阳性率更高（P值均〈0．05）。结论Fascin蛋白的检测对评价乳腺浸润性导管癌生物学行为和预后具有一定的参考价值,也有望成为一个乳腺癌的治疗靶点。     

Prognostic significance of fascin expression in extrahepatic bile duct carcinomas

Extrahepatic bile duct (EBD) carcinoma is an aggressive cancer with a poor prognosis. Fascin is an actin-bundling protein with roles in forming cell protrusions and mesenchymal and neuronal cell motility. Many human neoplasms up-regulate fascin. High fascin expression is thought to be a poor prognostic factor in some cancers; however, few data are available for the role of fascin in EBD carcinoma. We investigated fascin immunoreactivity in EBD carcinoma and tested it for correlations between fascin expression and clinicopathological parameters.Conventional tissue sections of 114 cases of EBD carcinomas were immunohistochemically analyzed for fascin expression.Fascin expression was tested by cytoplasmic staining. Negative, weak positive, and strong positive fascin staining was observed in 49 (43.0%), 32 (28.1%), and 33 cases (28.9%), respectively. Fascin expression in EBD carcinomas was significantly correlated with histological grade (P<0.0001), primary tumor (T) (P=0.002), TNM stage (P=0.036), lymphatic invasion (P=0.048), venous invasion (P=0.024), and adjacent organ invasion (P<0.0001). High fascin expression was a significant poor prognostic factor (P=0.0001) in EBD carcinoma. High fascin expression (P=0.004) and TNM stage (P=0.001) in EBD carcinoma were independent adverse prognostic factors.High fascin expression is significantly correlated with aggressive tumor phenotype in EBD carcinoma and is an independent poor prognostic factor in EBD carcinoma.     

利用组织芯片研究Fascin在食管癌中的表达

目的：本研究利用组织芯片技术高通量地研究Fascin蛋白在食管癌中的表达。方法：采用56例食管癌病人的病理蜡块制作组织芯片。应用免疫组化的方法检测Fascin在食管癌组织中的表达情况。同时对8例食管癌标本的冰冻切片显微切割“纯”癌和非癌组织，进行蛋白印迹研究。结果：在55例可提供信息的病例中，总体上72．7％的肿瘤中为阳性染色。其中高分化阳性率为95．2％，中分化为75％，低分化为35．7%，Fascin表达与食管癌分化有关（P〈0．001）。蛋白印迹结果也证实在分化较好的食管癌中Fascin为高表达。结论：利用组织芯片可以应用于回顾性Fascin表达的研究，研究中发现食管癌中Fascin的表达与组织分化有关。该试验方法简便，易于在临床推广。     

运动相关蛋白Fascin在胰腺癌的表达及其临床意义

目的 探讨运动相关蛋白Fascin在胰腺癌的表达及其与临床病理特征的关系.方法 采用RT-PCR法检测SW1990、Patu8988、BxPC3、CfPAC1 4株胰腺癌细胞株Fascin mRNA表达;采用免疫组化方法 检测54例胰腺癌及42例相应癌旁胰腺组织Fascin蛋白表达.结果 胰腺癌细胞株SW1990、Patu8988、CfPAC1有Fascin mRNA表达,BxPC3无表达;54例胰腺癌组织Fascin蛋白表达阳性35例,占64.8%,42例癌旁胰腺组织中均无阳性表达.Fascin蛋白表达与肿瘤分化程度(P<0.01)和淋巴转移(P<0.05)呈显著正相关,但与肿瘤大小及远处转移无相关性(P>0.05).结论 Fascin蛋白在胰腺癌组织中有较高的阳性表达率,检测其表达有助于胰腺癌的诊断,并有助于判断胰腺癌恶性程度.     

Fascin蛋白在妇科恶性肿瘤中的研究进展

Fascin蛋白作为一种肌动蛋白结合蛋白,通过增加细胞膜突起、参与细胞信号传导通路以及改变细胞与细胞外基质的黏附等途径促进肿瘤细胞侵袭转移。与妇科恶性肿瘤的发生、组织分化、肿瘤分级和淋巴结转移等因素有关。Fascin蛋白有望成为判断妇科肿瘤患者预后的有效指标,以Fascin蛋白为靶点的治疗可能为妇科恶性肿瘤的治疗提供新的思路。     

胃癌及相应癌旁组织中Fascin的表达及临床意义

目的：探讨F asc in在胃癌及相应癌旁组织中的表达及其临床病理意义。方法：应用免疫组织化学SP法检测52例胃癌及相应癌旁组织中F asc in的表达。结果：胃癌组织中F asc in阳性表达率高于癌旁组织,分别为71%和9%（P〈0.05）,F asc in阳性表达与癌组织病理分级、外膜侵犯、淋巴结转移密切相关（P〈0.05）。结论：胃癌组织中存在F asc in细胞,F asc in的表达与胃癌的分化程度、浸润深度及淋巴结转移有关,提示检测胃癌组织中F asc in的表达有助于评估肿瘤的恶性程度。     

Fascin promotes the motility and invasiveness of pancreatic cancer cells

AIM:To explore the role of actin-bundling protein, fascin during the progression of pancreatic cancer. METHODS:The plasmid expressing human fascin-1 was stably transfected into the pancreatic cancer cell line MIA PaCa-2. The proliferation, cell cycle, motility, scattering, invasiveness and organization of the actin filament system in fascin-transfected MIA PaCa-2 cells and control non-transfected cells were determined. Immunolocalization of Fascin, an Actin-Bundling Protein and Glial Fibrillary Acidic Protein in Human Astrocytoma Cells

Fascin is a 55-kDa globular protein that functions to organize filamentous-actin into parallel bundles. A role for fascin in cell migration has led to its study in many tumor types. In this report, we investigate fascin in astrocytomas. We show that fascin is expressed in astrocytes and in a panel of human astrocytoma cell lines. Immunofluorescence analysis demonstrates that fascin and the intermediate filament protein, glial fibrillary acidic protein (GFAP), are both expressed in the perinuclear region and within cytoplasmic processes of astrocytes and astrocytoma cells. Amino acid residues within the NH2 terminus of GFAP can undergo phosphorylation; these modifications regulate intermediate filament disassembly and occur during cytokinesis. We show that fascin and specific phosphorylated species of GFAP colocalize within dividing cells. Finally, we demonstrate that fascin co-immunoprecipitates with GFAP and that immunocomplex formation is preferential for GFAP phosphorylated at serine residues 8 and 13. These data show that fascin and GFAP are immunolocalized regionally within cells and tumors of astrocytic origin and suggest that their binding may occur during dynamic reorganization of intermediate filaments.
