静电法制备小微囊包裹成年猪胰岛的实验研究   总被引:4,自引:0,他引:4  
目的 探索用静电法制备小微囊包裹成年猪胰岛细胞。方法 用自制的静电微囊发生装置、制备海藻酸钠 -多聚赖氨酸 -海藻酸钠 (APA)微囊 (微囊直径 <35 0 μm) ,包裹成年猪胰岛 ,体外检测APA小微囊猪胰岛生物活性及微囊膜的通透性 .结果 静电法制备的APA微囊直径 30 0~ 35 0 μm ,大小相对均一 .小微囊包裹成年猪胰岛后 ,每个微囊内可见 1~ 2个胰岛团 ,表面光滑 .囊内胰岛组织学结构完整 ,体外培养微囊化胰岛素分泌良好 ,葡萄糖刺激释放明显 ,显示了良好的细胞活力及微囊膜通透性 .结论 用我们自制的静电微囊发生装置能制备APA微囊包裹成年猪胰岛细胞 ,微囊直径 30 0~ 35 0 μm ,表面光滑 ,囊内猪胰岛生物活性良好  相似文献

本文提出并研究了一种新型的电磁热疗系统-H面号角环形相控阵系统。推导出了适应性广的理论公式。以肺部CT图为模型,用矩量法计算得到了胸腔横断面内电场分布及吸收功率分布的计算机数值结果。结果表明,在200MHz的电磁波辐射下,适当调节阵列各单元的相位,振幅和位置,肺部肿瘤可得到优化的选择性加热,从而达到最佳的热疗效果。  相似文献

目的:观察海藻酸钠-多聚赖氨酸-海藻酸钠(APA)微囊包裹的牙髓干细胞移植在Scid小鼠腹腔内后成牙本质的作用,探讨APA作为牙齿组织工程支架材料的可行性。方法:用APA微囊包裹第二代人牙髓干细胞,植入Scid鼠腹腔内,植入6周后取材,通过检测被APA微囊包裹的牙髓干细胞是否表达牙本质基质特异性蛋白,观察牙本质形成情况。结果:APA微囊包裹的牙髓干细胞在植入6周后可以检测到特异性牙本质基质蛋白-牙本质涎磷蛋白的形成。结论:用APA作为支架材料,采用组织工程方法包裹的牙髓干细胞可以形成牙本质基质,牙髓干细胞和APA用于组织工程牙本质的构建具有广阔前景。  相似文献

Objectives

To compare the effects of conventional core stabilization and dynamic neuromuscular stabilization (DNS) on anticipatory postural adjustment (APA) time, balance performance, and fear of falls in chronic hemiparetic stroke.

Design

Two-group randomized controlled trial with pretest-posttest design.

Setting

Hospital rehabilitation center.

Participants

Adults with chronic hemiparetic stroke (N=28).

Interventions

Participants were randomly divided into either conventional core stabilization (n=14) or DNS (n=14) groups. Both groups received a total of 20 sessions of conventional core stabilization or DNS training for 30 minutes per session 5 times a week during the 4-week period.

Main Outcome Measures

Electromyography was used to measure the APA time for bilateral external oblique (EO), transverse abdominis (TrA)/internal oblique (IO), and erector spinae (ES) activation during rapid shoulder flexion. Trunk Impairment Scale (TIS), Berg Balance Scale (BBS), and Falls Efficacy Scale (FES) were used to measure trunk movement control, balance performance, and fear of falling.

Results

Baseline APA times were delayed and fear of falling was moderately high in both the conventional core stabilization and DNS groups. After the interventions, the APA times for EO, TrA/IO, and ES were shorter in the DNS group than in the conventional core stabilization group (P<.008). The BBS and TIS scores (P<.008) and the FES score (P<.003) were improved compared with baseline in both groups, but FES remained stable through the 2-year follow-up period only in the DNS group (P<.003).

Conclusions

This is the first clinical evidence highlighting the importance of core stabilization exercises for improving APA control, balance, and fear of falls in individuals with hemiparetic stroke.  相似文献

Purpose: The purpose of this study is to quantitatively assess the effect of 6 months of supervised adapted physical activity (APA i.e. physical activity designed for people with special needs) on spatio-temporal and kinematic parameters of gait in persons with Multiple Sclerosis (pwMS).

Methods: Twenty-two pwMS with Expanded Disability Status Scale scores ranging from 1.5 to 5.5 were randomly assigned either to the intervention group (APA, n?=?11) or the control group (CG, n?=?11). The former underwent 6 months of APA consisting of 3 weekly 60-min sessions of aerobic and strength training, while CG participants were engaged in no structured PA program. Gait patterns were analyzed before and after the training using three-dimensional gait analysis by calculating spatio-temporal parameters and concise indexes of gait kinematics (Gait Profile Score – GPS and Gait Variable Score – GVS) as well as dynamic Range of Motion (ROM) of hip, knee, and ankle joints.

Results: The training originated significant improvements in stride length, gait speed and cadence in the APA group, while GPS and GVS scores remained practically unchanged. A trend of improvement was also observed as regard the dynamic ROM of hip, knee, and ankle joints. No significant changes were observed in the CG for any of the parameters considered.

Conclusions: The quantitative analysis of gait supplied mixed evidence about the actual impact of 6 months of APA on pwMS. Although some improvements have been observed, the substantial constancy of kinematic patterns of gait suggests that the full transferability of the administered training on the ambulation function may require more specific exercises.

Background

l-glutamate is a major excitatory neurotransmitter in the mammalian brain. Glutamate receptors have been reported in the rat adrenal cortex and in human aldosterone-producing adenomas (APA). However, details regarding the expression levels and functions of these receptors in human adrenocortical tissues remain unknown.

Methods

The mRNA levels of glutamate receptors were evaluated by qPCR in: 12 normal adrenal cortex (NAC), 11 APA, and 12 cortisol-producing adenoma (CPA) tissues. Protein localization was evaluated by immunohistochemistry for 15 NAC, 5 idiopathic hyperaldosteronism cases, 15 APA and 15 CPA. H295R cells were treated with angiotensin-II or forskolin alone or combined with the GRM2/3 agonist LY354740.

Results

The level of GRM3 mRNA was higher in APA than in CPA (P = 0.0086) or NAC (P = 0.0022). GRM1, IGLUR2, and IGLUR3 were also detected in adrenocortical tissues. When added to angiotensin-II/forskolin treatments, LY354740 decreased aldosterone and cortisol production in H295R cells.

Conclusions

GRM3 is considered to regulate steroidogenesis in adrenocortical tissues.  相似文献

BackgroundIn view of freezing of gait's circumstances of occurrence in Parkinson's disease, attentional resources appear to be involved in step initiation failure. Anticipatory postural adjustments (APAs) are essential because they allow unloading of the stepping leg and so create the conditions required for progression.Our main objective was to establish whether or not a change in attentional load during step initiation modulates APAs differently in patients with vs. without freezing of gait.MethodsThree groups of 15 subjects were recruited: elderly people and parkinsonian patients with or without freezing of gait. Attention was modulated before step execution by means of an auditory oddball discrimination task with event-related potential recording. The primary endpoint was the occurrence of inappropriate APAs following the attentional task, i.e. APAs not followed by a step after an intercurrent auditory stimulus.ResultsIn parkinsonian patients with freezing of gait, inappropriate APAs were recorded in 63% of the trials and were observed more frequently than in patients without freezing of gait (51%) and elderly controls (48%). Furthermore, inappropriate APAs in freezers were longer and more ample than in parkinsonian non-freezers and controls. Lastly, postural preparation was impaired in the parkinsonian patients.ConclusionOur results indicate that allocation of attentional resources during step preparation influences the release of APAs differently in freezers and non-freezers. Modulating attentional load is partly responsible for triggering an inappropriate motor program. This difficulty in focusing attention or resisting interference may contribute (at least in part) to the gait initiation failure observed in parkinsonian freezers.  相似文献

目的:探讨阿帕替尼(apatinib,APA)联合顺铂(cisplatin,DDP)对胃癌(gastric carcinoma,GC)细胞增殖、侵袭和迁移 能力的影响及其分子机制。方法:收集2016年1月到2019年6月武威市人民医院手术切除的50例GC患者的癌及癌旁组织标 本,以及GC细胞系MGC803和SGC7901, 用qPCR检测组织中HMGA2和细胞中增殖、迁移及侵袭相关mRNA的表达水平。采 用脂质体转染技术, 将pcHMGA2转染MGC803和SGC7901细胞,经分别用不同浓度的DDP和APA处理,分为NC、pcHMGA2、 pcHMGA2+DDP及pcHMGA2+DDP+APA组, 用Western blotting检测GC细胞中HMGA2蛋白的表达水平,MTT、Transwell小室 法分别检测细胞的增殖、迁移和侵袭能力。结果:HMGA2 mRNA在GC组织中表达水平高于癌旁组织(P<0.05),且高表达组 GC患者的生存率显著降低(P<0.01)。DDP 显著抑制 MGC803 和 SGC7901 细胞的增殖、侵袭和迁移能力(均P<0.01); DDP+APA组MGC803和SGC7901细胞的增殖、侵袭和迁移能力显著低于DDP组(均P<0.01);APA显著增强DDP对GC细胞 HMGA2表达的抑制作用(均P<0.01);APA通过下调HMGA2表达增强DDP对GC的抗肿瘤性。结论:APA能促进DDP对GC的 抗瘤作用,其分子机制可能是增强DDP对HMGA2表达的抑制作用有关。  相似文献
