背景 致密性骨炎(OCI)和其他疾病有时难以鉴别,探讨血清骨转换生化标志物可为OCI的鉴别诊断提供依据。 目的 探索女性OCI患者的血清骨转换生化标志物的水平变化及临床意义。 方法 回顾性选取2013年6月至2022年2月在北京积水潭医院门诊及住院诊断为OCI的61例女性患者作为观察组,年龄15~50岁,平均(33.8±6.6)岁,病程2周~15年。选择同期61例女性体检健康者作为对照组,年龄15~48岁,平均(35.6±7.6)岁。比较两组一般临床资料和血清骨转换生化标志物水平,并对血清骨转换生化标志物与病情相关指标进行相关性分析。 结果 观察组血清白蛋白(45.4±2.9)g/L低于对照组(46.5±2.8)g/L(t=2.190,P<0.05)。血清骨转换生化标志物比较结果显示,观察组血清1型胶原羧基末端肽β特殊序列(β-CTX)〔0.28(0.23,0.37)μg/L〕、N-端骨钙素(OC)〔13.1(11.2,16.2)μg/L〕、25-羟维生素D3〔25-(OH)VD3〕〔(14.1±5.1)μg/L〕低于对照组〔0.36(0.29,0.48)μg/L,15.6(13.7,17.3)μg/L,(17.5±6.6)μg/L〕(Z=-2.983、-3.255,t=3.081,P<0.05)。长病程亚组OC水平〔14.6(12.4,18.5)μg/L〕高于短病程亚组〔11.7(10.2,14.0)μg/L〕(Z=-2.407,P<0.05)。多孕亚组β-CTX〔0.25(0.22,0.32)μg/L〕、OC水平〔12.2(10.3,15.0)μg/L〕低于非多孕亚组〔0.33(0.26,0.44)μg/L、13.4(12.0,18.8)μg/L〕(Z=-2.486、-1.897,P<0.05)。相关性分析显示,观察组血清1型前胶原氨基端延长肽(tP1NP)与妊娠次数、生产次数均呈负相关(rs=-0.276、-0.298,P<0.05),OC与体质指数(BMI)、视觉模拟评分法(VAS)评分、妊娠次数均呈负相关(rs=-0.284、-0.374、-0.360,P<0.05),25-(OH)VD3水平与BMI呈正相关(rs=0.275,P<0.05)。 结论 女性OCI患者血清OC、β-CTX水平明显降低,可为鉴别其他疾病提供依据;血清OC水平可以反映OCI患者的严重程度,同时OC水平与患者妊娠次数相关;tP1NP与妊娠次数、生产次数相关。  相似文献

Cell culture-based influenza vaccine manufacturing is of growing importance. Depending on virus strains, differences in infection dynamics, virus-induced apoptosis, cell lysis and virus yields are observed. Comparatively little is known concerning details of virus–host cell interaction on a cellular level and virus spreading in a population of cells in bioreactors. In this study, the infection of MDCK cells with different influenza A virus strains in lab-scale microcarrier culture was investigated by flow cytometry. Together with the infection status of cells, virus-induced apoptosis was monitored. A mathematical model has been formulated to describe changes in the concentration of uninfected and infected adherent cells, dynamics of virus particle release (infectious virions, hemagglutinin content), and the time course of the percentage composition of the cell population.  相似文献

Secretion of Igs and surface expression of HLA antigens was examined in lymphoid cells as a function of temp. Upon reducing the temp from 37 to 20 degrees C a progressive decrease in the secretion of Ig and surface expression of HLA antigens was noted. When the status of the oligosaccharides present on these glycoproteins was examined, conversion of high-mannose [endo-beta-N-acetylglucosaminidase-(Endo H) sensitive] to complex-type (Endo H resistant) oligosaccharides diminished with decreasing temp. At no time was an accumulation of Endo H resistant glycoproteins seen intracellularly. These results show that the phenomenon observed for synthesis and intracellular transport of viral glycoproteins in epithelial cells at reduced temp, namely intracellular accumulation of viral glycoproteins carrying complex sugar moieties, does not necessarily apply to glycoprotein transport in lymphoid cells. A difference in subcellular organization of epithelial and lymphoid cells may be responsible for this discrepancy.  相似文献

Magnocellular neurons in rat hypothalamic slices are known to exhibit dye coupling: the transfer of the fluorescent dye, Lucifer Yellow, from an intracellularly-injected neuron to one or more nearby neurons. The question of the hormonal identity of coupled cells and the possibility of dye coupling as an artefact led us to determine the immunoreactivity of dye-coupled magnocellular neurons in the paraventricular nucleus of the rat hypothalamus using antisera to oxytocin- and vasopressin-associated neurophysins. In 23 pairs, one triplet, and one quadruplet, immunoreactivity to one or the other antiserum was always exclusive, and dye coupling was always homotypic, that is, coupled neurons in each instance were reactive to the same antiserum. The quadruplet, triplet and 17 pairs were immunoreactive to vasopressin-associated neurophysin, and oxytoxin-associated neurophysin immunoreactivity was observed in the remaining pairs. Immunoreactivity to each antiserum was found for somasomatic and non somasomatic modes of coupling and for coupled neurons in the three magnocellular areas of the nucleus. A relationship between mode of coupling and hormone content was not detected. The data support the hypothesis that coupling is a real, functionally significant mechanism for coordinating neuronal activity in this nucleus, particularly under conditions of high hormone demand. They do not support the idea that coupling is artefact. The possibility of a relationship between hormone content and mode of coupling, and the projection pathway(s) of the coupled neurons of each type require further study.  相似文献

The Thy-1 antigens or rat brain and thymus have been isolated and chemically characterized, but those of mice have not been identified. Moreover, it is uncertain whether the antigens are glycolipids or glycoproteins. This study with highly purified preparations of gangliosides GM1, 1GD1a, GD1b and GT1b from bovine brain and several ganglioside fractions from mouse brain showed that Thy-1 activity does not reside in gangliosides, but rather in the chloroform-methanol-insoluble residue of brain remaining after extraction of gangliosides. The antigen could be solubilized from this residue with a non-ionic detergent. The antigenic activity of the solubilized preparation was heat-labile but resistant to periodate. The chemical properties of the Thy-1 antigen of mouse brain are discussed.  相似文献

ABSTRACT

Avian influenza virus (AIV) can cause serious zoonotic disease, thereby threatening the poultry industry and human health. An efficient and rapid detection approach is crucial to prevent and control the spread of avian influenza. In this study, a novel protein microarray was developed. Haemagglutinin proteins of H5 and H7 subtypes and nucleoprotein (NP) were purified and spotted onto the initiator-integrated poly-(dimethylsiloxane) as antigens. Monoclonal antibodies with inhibition effect were screened and utilized for the synchronous detection of three avian influenza antibodies in different species. In the protein microarray, the cut-off values were 40%, 50% and 30% inhibition for H5 antibody detection; 50%, 50% and 20% for NP antibody detection; 40%, 50% and 40% for H7 antibody detection in chicken, peacock and duck sera, respectively. The 95 serum samples were detected by microarray, and results were compared with the findings of AIV antibody test enzyme-linked immunosorbent assay (ELISA) or haemagglutination inhibition (HI) test. NP antibody detection in the microarray showed 100% (55/55) agreement ratio in chicken using ELISA. Compared with HI, H5 antibody detection in the microarray showed 100% (95/95) agreement ratio in chicken, peacock and duck, whilst those of H7 displayed 98.18% (54/55) agreement in chicken, 100% (20/20) in peacock and 90% (18/20) in duck. In conclusion, this novel protein microarray is a high-throughput and specific method for the detection of AIV antibodies and simultaneous distinction of antibodies against H5 and H7 subtypes. It can be applied to the serological diagnosis and epidemiological investigation of AIV.

目的研究新发现的人内源性逆转录病毒 NP9基因在系统性红斑狼疮(systemic lupus erythematosus, SLE)患者体内表达及其蛋白功能预测.方法逆转录PCR、T-A克隆技术和DNA序列测定分离、克隆和分析 NP9基因, 检测40例SLE患者和48名正常对照的 NP9基因表达, 借助NCBI BLAST系列分析工具及相应的基因分析软件预测其蛋白功能.结果 SLE患者组逆转录病毒 NP9基因表达阳性率为77.5%(31/40),明显高于正常对照的8.3%(4/48), P<0.01. NP9基因编码产物由74个氨基酸组成,含有多个细胞核内分布信号,其等电点(pI)为9.59,与SLE患者体内高表达的某些细胞因子具有较高同源性.结论 SLE患者存在逆转录病毒 NP9基因特异性转录, NP9表达可能与SLE发生发展相关.  相似文献

The preparation steps usually necessary for obtaining ultrathin frozen sections of biological material (chemical prefixation, enclosing, cryoprotective treatment, freezing, sectioning, and post-staining the sections for transmission electron microscopy) are submitted to a critical analysis. The application of cryo-ultramicrotomy, in particularly for cytochemical purposes, is reviewed. Fundamental considerations of chemical prefixation and poststaining are supported by examples from yeast cytology. Furthermore, the efficiency of the cryo-ultramicrotomy (electron optical resolution of ultrastructural details) is demonstrated on yeast cells and protoplasts.  相似文献

 目的 观察NVB加PDD治疗晚期非小细胞肺癌疗效。方法  1999年 7月~ 2 0 0 1年 8月我们应用进口长春瑞滨 (法国皮尔法伯制药公司生产 ) ,NVB 2 5~ 30mg/m2 ,d1,d8,溶于 0 .9%生理盐水15 0ml中快速静滴 ,再以 2 5 0ml0 .9%生理盐水快速静滴 ,DDP 4 0~ 5 0mg/d1,2 ,3,静脉滴注 ,同时给予水化。 2 1天为一周期。结果 本联合化疗方案治疗晚期非小细胞肺癌 4 0例 ,取得了较好疗效 ,其中CR 4例、PR 2 6例 ,总有效率 75 %。结论 NP联合方案治疗晚期肺癌是合理、方便、安全、有效的一种可行的方案 ,可以作为一线方案推荐临床推广。  相似文献
