The regional distribution of inflammation during acute lung injury (ALI) is not well known. In an ovine ALI model we studied regional alveolar inflammation, surfactant composition, and CT-derived regional specific volume change (sVol) and specific compliance (sC). 18 ventilated adult sheep received IV lipopolysaccharide (LPS) until severe ALI was achieved. Blood and bronchoalveolar lavage (BAL) samples from apical and basal lung regions were obtained at baseline and injury time points, for analysis of cytokines (IL-6, IL-1β), BAL protein and surfactant composition. Whole lung CT images were obtained in 4 additional sheep. BAL protein and IL-1β were significantly higher in injured apical vs. basal regions. No significant regional surfactant composition changes were observed. Baseline sVol and sC were lower in apex vs. base; ALI enhanced this cranio-caudal difference, reaching statistical significance only for sC. This study suggests that apical lung regions show greater inflammation than basal ones during IV LPS-induced ALI which may relate to differences in regional mechanical events.  相似文献

Sympathoadrenal (SA) cell lineage encompasses neural crest derivatives such as sympathetic neurons, small intensely fluorescent (SIF) cells of sympathetic ganglia and adrenal medulla, and chromaffin cells of adrenal medulla and extra-adrenal paraganglia. SA autografts have been used for transplantation in Parkinson's disease (PD) for three reasons: (i) as autologous donor tissue avoids graft rejection and the need for immunosuppressant therapy, (ii) SA cells express dopaminotrophic factors such as GNDF and TGFβs, and (iii) although most of SA cells release noradrenaline, some of them are able to produce and release dopamine. Adrenal chromaffin cells were the first SA transplanted cells in both animal models of PD and PD patients. However, these autografts have met limited success because long-term cell survival is very poor, and this approach is no longer pursued clinically. Sympathetic neurons from the superior cervical ganglion have been also grafted in PD animal models and PD patients. Poor survival into brain parenchyma of grafted tissue is a serious disadvantage for its clinical application. However, cultured sympathetic cell grafts present a better survival rate, and they reduce the need for levodopa medication in PD patients by facilitating the conversion of exogenous levodopa. SA extra-adrenal chromaffin cells are located on paraganglia (i.e., the Zuckerkandl's organ), and have been used for grafting in a rodent model of PD. Preliminary results indicate that long-term survival of these cells is better than for other SA cells, exerting a more prolonged restorative neurotrophic action on denervated host striatum. The ability of SA extra-adrenal cells to respond to hypoxia, differently to SA sympathetic neurons or adrenal medulla cells, could explain their good survival rate after brain transplantation.  相似文献

Vagal paraganglia resemble the carotid body and are chemosensitive to reduction in the partial pressure of oxygen (PO2PO2) (O’Leary et al., 2004). We hypothesised that they may also mediate communication between the immune system and the central nervous system and more specifically respond to the pro-inflammatory cytokines: interleukin-1 beta (IL-1β) and tumour necrosis factor-α (TNF-α). We recorded axonal firing rate of isolated superfused rat glomus cells – located at the bifurcation of the superior laryngeal nerve – to IL-1β or TNF-α at concentrations of 0.5 ng/ml, 5 ng/ml and 50 ng/ml. Twenty-three successful single fibre recordings were obtained from 10 animals. IL-1β and TNF-α had no statistically significant effect on the frequency of action potentials observed (p = 0.39 and 0.42, respectively, repeated measures ANOVA). The activity of both cytokines was tested by observing translocation of P65-NFκB from cytoplasm to nucleus in cultured HELA cells. In conclusion, an immune role for SLN paraganglia has not been established.  相似文献

Paragangliomas are tumors arising in the paraganglia and rarely occur in the spinal canal. In the literature, 11 such cases have been reported. We present 7 additional cases, 3 of which were epidural in location in contrast to previously described intradural cases. The problems of histological diagnosis of these tumors are discussed. The value of determination of biogenic amines in the tumor tissue or the urine has been pointed out.  相似文献

Summary The present study provides a LM and EM inventory of the fibers of the rat abdominal vagus, including dorsal and ventral trunks and the five primary branches. Whole mounts (n = 15) were prepared to characterize the branching patterns. A set of EM samples consisting of both trunks and all branches (i.e. dorsal and ventral gastric, dorsal and accessory celiac, and hepatic) were then obtained from each of six additional animals. A complete cross-sectional montage (x 10000) was prepared from each sample. All axons were counted, and >10% of them were evaluated morphometrically.The means of unmyelinated axon diameters for each of the five branches were similar (0.75–0.83 m). However, the shapes of the fiber size distributions, as summarized by their skew coefficients, revealed that the two gastric branches differed significantly from the two celiac branches; furthermore, the hepatic size distribution differed from all others. Most of the myelinated fibers (85%) in all branches were <2.6 m in diameter and had sheath widths between 0.1 and 0.5 m. The gastric branches, however, also contained a few larger myelinated fibers with sheath widths as great as 0.85 m. Whole mounts revealed fibers which were not of supradiaphragmatic orgin within all five vagal branches; these adventitial bundles were traced along the perineurium between adjacent branches. The sum of the fibers in the five branches (26930) was 21% more than the number counted in the parent trunks (22272); this excess probably reflects the adventitial fiber content. The whole mounts also showed that a large and regularly positioned paraganglion was associated with the dorsal branches.The structural profiles observed (i.e. unmyelinated and myelinated fibers size distributions, presence of extrinsic fascicles, glomus tissue content, etc.) differentiate the vagal branches into three morphologically distinct sets: a gastric pair, a celiac pair, and a hepatic branch. The fiber counts, when considered with observations of the numbers of efferents and adventitial fibers in the nerve, suggest that the percentage of efferent fibers is much higher than in all the widely accepted estimates found in the literature: efferent fibers may represent over a quarter of the total number of fibers.  相似文献

Triple label immunohistochemistry was used to study the coexistence of the catecholamine-synthesising enzymes dopamine beta-hydroxylase (DBH) and tyrosine hydroxylase (TH) and several neuropeptides including neuropeptide Y (NPY), vasoactive intestinal polypeptide (VIP), substance P (SP), calcitonin gene-related peptide (CGRP), somatostatin (SOM) and galanin (GAL) as well as nitric oxide synthase (NOS) in developing pelvic paraganglion cells in a series of human male fetal, neonatal and infant specimens ranging in age from 13 wk of gestation to 3 y postnatal. 13–20 wk old fetal specimens possessed large clusters of paraganglion cells lying lateral to the urinary bladder and prostate gland which were intensely DBH-immunoreactive (-IR) but lacked TH, NOS and the neuropeptides investigated. With increasing fetal age small clusters of paraganglion cells were observed in the muscle coat of the urinary bladder. At 23 wk of gestation occasional paraganglion cells were NOS or NPY-IR while at 26 wk of gestation the majority of paraganglion cells were TH-IR and a few were SOM or GAL-IR. Some postnatal paraganglia within the bladder musculature contained cells which were all VIP, SP or CGRP-IR while others displayed coexistence of NOS and NPY, SP and CGRP, or NPY and VIP. The presence of NOS in certain paraganglion cells indicates their capacity to generate nitric oxide (NO). These results show that human paraganglion cells develop different phenotypes possibly dependent upon their location within the bladder wall. A delicate plexus of branching varicose nerves was observed in the fetal paraganglia which increased in density with increasing gestational age. The majority of these nerves were VIP-IR while others were CGRP, SP, NPY, NOS or GAL-IR. The presence of nerve terminals adjacent to the paraganglion cells implies a neural influence on the functional activity of the paraganglia. Some paraganglia in the late fetal and early postnatal specimens contained Timofeew's sensory corpuscles, resembling pacinian corpuscles in their morphology. The central nerve fibre of these corpuscles displayed immunoreactivity for SP, CGRP and NOS, the latter indicating a possible role for NO in afferent transmission from the urinary bladder. In addition, a few corpuscles were penetrated by a noradrenergic nerve fibre immunoreactive for NPY and TH, which may have a modulatory role on the sensory receptor.  相似文献

We report a rare case of a solitary primary paraganglioma arising in the mesentery, found in a 72-year-old woman who presented with abdominal pain and a palpable abdominal mass. This extra-abdominal paraganglioma developed from paraganglionic cells that travelled by vertebral migration from the root of the superior mesenteric artery. Extra-adrenal paraganglia extend anywhere from the neck down to the base of the pelvis. Ultrasonography, computed tomography (CT), and angiography showed a solid and cystic heterogeneously enhanced mass, which was fed by the superior mesenteric artery, without distant metastasis. Exploratory laparotomy revealed a large, dark, brownish-red mass in the mesentery of the ileum, which was distinct from the ileum. The mass consisted of peripherally solid areas with central hemorrhage and cystic degeneration. It was diagnosed as a paraganglioma histologically. The patient is free from recurrence of paraganglioma after 1 year of follow up. To our knowledge, this represents only the seventh case of a paraganglioma arising in the mesentery.  相似文献

Summary The rat's vagal hepatic branch and associated tissues were studied using light and electron microscopy. Whole mounts, serial sections, and vascular endocasts were used to characterize the tissue from the anterior vagal trunk to the porta hepatis. Fiber number and caliber as well as intraneural organization were analyzed from complete cross-sectional electron micrographic montages of the hepatic branch sampled at its point of separation from the anterior vagal trunk.The hepatic branch ramified from the anterior vagus in one (in 47% of the specimens), two (in 37%) or three (in 16%) bundles. The single bundled hepatic branch contained 2887±287 unmyelinated fibers, and their size distribution, with a mean diameter of 0.66±0.02 m, was Gaussian. Myelinated fibers numbered only 21±4 per branch and had a complex size distribution ranging from 0.5 to 1.8 m with a mean of 1.2±0.03 m. Forty four ±6% of the myelinated fibers were found in a single subfascicle in the dorso-medial pole of the nerve. Whole mounts at this level revealed that a distinct bundle, here designated an extrinsic hepato-gastric bundle, occurred within the hepatic branch and linked the omental hepatic branch and the distal anterior gastric branch, apparently without central vagal connections. In the lesser omentum, between the esophagus and the hepatic artery proper, the hepatic branch formed a plexus which was characterized by numerous nerve divisions, anastomoses and large paraganglia (196–463 glomus cells per paraganglion). This plexiform segment ended with the recombining of the hepatic branch into 5–7 bundles which variously ascended in the porta, descended on the hepatic artery proper, or traversed the portal vein. Through its omental course, the hepatic branch traveled in close apposition to the hepato-esophageal artery and the corresponding vein as well as a prominent lymphatic vessel with associated hemolymph nodes.  相似文献
