During the initial stages of B lymphocyte differentiation heavy chain variable (VH), diversity (DH) and joining (JH) gene segments recombine to form a functional heavy chain variable region (VDJ) gene. Evidence for genetic polymorphism of the human JH gene segments has been obtained from mature rearranged VDJ sequences. We conducted an analysis of the published rearranged JH gene sequences and found that the JH alleles present in the two published germ-line JH region sequences were rare (approx. 2%) in the rearranged sequences. As an attempt to explain this discrepancy a 2.5-kb strech of DNA containing all the six heavy chain JH region genes and the most 3' DH gene segment, DHQ52, was amplified by the polymerase chain reaction from 39 individuals and analyzed for restriction fragment length polymorphism. Five new JH region haplotypes were found and sequenced. These new haplotypes contained the coding segment alleles that were frequent in antibody genes. Surprisingly, a high number of interallelic differencies in the non-coding sequence was found between the new and the two previously published haplotypes implying that the haplotypes had been separated early in evolution. Immunologic changes have been reported in the dementing illnesses of mid and late life. The results of two studies of drug-free males meeting research diagnostic criteria for primary neuronal degeneration of the Alzheimer's type suggest that serum IgG levels decrease with the progression of dementia. Serum IgG levels were inversely correlated with duration of illness and the levels of psychiatric symptomatology. 目的 运用噬菌体表面表达技术,获得基因工程抗腺病毒伴随病毒抗体Fab、IgG。方法 从酶联免疫吸附试验阳性的人外周血中分离淋巴细胞。提取总RNA,逆转录cDNA,聚合酶链反应扩增人IgG Fab轻、重链基因,利用pComb3载体构建噬菌体抗体库。用纯化的腺病毒伴随病毒为固相抗原筛选抗体Fab段,并在E.coli中进行分泌性表达。通过ELISA和间接免疫荧光试验、筛选和鉴定Fab抗体,并进行序列测定。然后将其重链和轻链基因克隆到全抗体表达载体pAC-L-Fc上。转染昆虫sf-9细胞,利用杆状病毒,昆虫细胞系统实现全抗体的分泌型表达,免疫沉淀试验鉴定它的抗蛋白。结果 分离到一株Fab克隆AAVs-31,腺病毒伴随病毒抗原和抗Fab抗体直接ELISA检测阳性,间接免疫荧光试验呈阳性,序列分析结果表明是一新的序列,所获得的基因为人源IgG Fab基因。由Gamma链和Kappa链组成。表达的全抗体ELISA反应、免疫荧光反应和间接免疫荧光试验均为阳性,免疫沉淀试验结果显示它结合病毒颗粒,不结合任一VP1、VP2、VP3单独衣鞘蛋白。结论 用噬菌体表面表达技术首次获得了抗腺病毒伴随病毒Ⅱ型单克隆抗体Fab段,并在真核系统中表达了其全抗体,它们识别的可能是衣鞘蛋白组装时形成的表位。 相似文献

本文应用微波辐射免疫比浊法加速测定免疫球蛋白(IgG、IgA、IgM).结果表明:用本法测定的线性范围较宽:IgG从3.24~51.92/L;IgA从0.49~7.92g/L;IgM从0.36~5.75/L.批内、批间精密度(CV):IgG从44~7.4%;IgA从4.1~9.5%;IgM从4.0~8.8%.本法与传统免疫比浊法和进口试剂自动免疫比浊法(Monarch 2000)测定比较,其结果相关良好,r从0.950至0.996,P>0.05.本法的特点是操作简便、快速,可明显缩短抗原和抗体反应达到终点的时间. 相似文献

Biological response modifiers (BRMs) are agents which can modify the immune response to cancer or invasion of the organism by infectious agents. An explosive appearance of new BRMs has resulted from the development of recombinant gene technology and the availability of monoclonal antibodies. Colony-stimulating factors first became available for the prevention of neutropenia but may also have a role in the treatment of infections. Interleukin-1 is being tested as a modular of hematopoiesis and may be useful as a helper factor for T- and B-cell function. Immunoglobulins are being used against viral and bacterial infections while interferons can prevent viral upper respiratory infections and suppress or irradicate some viral hepatitides. Amyloidosis is a complication of long-term hemodialysis treatment. The major histological feature of hemodialysis-associated
amyloidosis (HAA) is the deposition of amyloid fibrils in the affected lesions, due, in part, to elevated serum β2-microglobulin (β2M) levels. In vitro studies reveal that serum immunoglobulin light and heavy chains co-deposit with β2M
in tissues affected by HAA. Only one study of HAA has been performed in young dialysis patients. We therefore assessed risk
factors for HAA in a group (n=30) of young (18.7±0.9 years) patients receiving chronic, uninterrupted hemodialysis using cellulose acetate membranes. All
patients initiated dialysis before reaching 18 years of age. The pre-dialysis serum β2M level was 49.7±3.9 mg/l (normal 0–2.4
mg/l). Since serum albumin was normal (4.3±0.1 mg/dl) and serum protein/albumin was elevated (1.7±0.0, normal 1.2–1.5), indicating
increased circulating protein, we assayed immunoglobulins in the same patients. The serum immunoglobulin levels (expressed
as a percentage of the total level of serum proteins) were elevated (21.3±0.9%, normal 11.1%–21.0%). The Kt/v was 1.37±0.03,
suggesting that the high levels of serum β2M and immunoglobulins were not due to inadequate dialysis in these patients. Patients
with residual renal function (Kr) did display significantly lower serum levels of β2M (33.2±2.3, P=0.03). Furthermore, improved clearance of β2M correlated with higher values of Kr (r=0.914). In contrast, serum levels of immunoglobulin (22.6±3.7, P=0.5) were unaffected by Kr. In addition, there was no correlation between older age at onset of dialysis and serum levels
of either β2M (r=0.107) or immunoglobulins (r=0.321). Finally, the length of time on dialysis had no effect on serum levels of either β2M (r=0.105) or immunoglobulins (r=0.092). Taken together, these results indicate that young hemodialysis patients may be at risk for HAA.
