Distinct genetic alterations and luminal molecular subtype in nested variant of urothelial carcinoma

对2014年1例初诊为河南省本地感染的间日疟病例进行实验室检测与流行病学溯源调查,以明确其感染来源。收集该病例的流行病学资料,分别采用厚薄血膜吉氏染色法、疟疾快速诊断试纸(RDT)法和巢式PCR法检查患者外周血液,并对其环子孢子蛋白(CSP)基因序列进行分析。流行病学调查显示,该患者曾于2013年5月至缅甸停留约1周,同年6月发病,确诊为间日疟,经青蒿琥酯治疗后,疟原虫转阴,症状消失。CSP基因序列分析显示,该患者前后两次发病时的血样扩增出的CSP序列的中央重复区完全一致,与缅甸分离株(Gen Bank登录号为ABS95455和ABS95456)的序列一致性分别为95.1%和100%,与2个河南分离株HN3和HN7(登录号为KP888996和KP889000)的序列一致性分别为88.8%和67.1%。推测该病例为输入性间日疟复发病例。 相似文献

应用卵圆孔定位仪治疗三叉神经痛 总被引:1,自引:0,他引:1
目的总结卵圆孔定位仪在选择性经皮温控射频热凝治疗三叉神经痛的应用方法和技巧。方法对三叉神经第Ⅱ、Ⅲ支疼痛者或第Ⅰ、Ⅱ、Ⅲ支同时疼痛者应用Hartel前入路半月神经节射频热凝治疗,其中260例患者应用三叉神经卵圆孔定位仪辅助定位,对39例患者术中应用X线或三维CT进行卵圆孔靶点验证。结果所有病例穿刺成功。术后即刻疗效:优良206例,良好42例,无疗效12例,总有效率95.4%,无严重并发症发生。结论卵圆孔定位仪可用于射频热凝治疗三叉神经痛的辅助定位,可以提高穿刺的成功率和安全性。 相似文献

目的 体外扩增间日疟原虫深圳株红内期小亚单位核糖体核糖核酸编码基因(SSUrDNA)片段,研究其结构与功能。方法 设计一对特异性引物,采用聚合酶链反应(PCR)从间日疟原虫患者血样中扩增出间日疟原虫SSUrDNA片段,以PUC19质粒T载体构建重组子导入大肠杆菌JM109;阳性克隆双酶切鉴定后,双脱氧末端终止法测定序列。结果 间日疟原虫SSUrDNA扩增片段大小为341bp;阳性克隆双酶切及PCR扩增均得到预期大小的片段;序列测定插入片段为341bp,与Sal I株顺序相比,仅在第151位处缺失一个碱基C。结论 成功克隆了间日疟原虫SSUrDNA片段.该序列在间日疟原虫虫株间高度保守。 相似文献

Febrifugine and isofebrifugine alkaloid mixtures extracted from the leaves and buds of Hydrangea macrophylla var. Otaksa, collected during different months, in Japan, were quantified using high-performance liquid chromatography. Leaves collected
during the flowering season, namely from June to August, contained 0.16–0.31 mg/g of the alkaloid mixture, whereas those collected
from September to December had less than 0.03 mg/g of the mixture. However, extracts of buds harvested from October to February
contained a consistently larger amount (more than 0.49 mg/g) of the alkaloids. Hot-water extracts from the leaves and buds
collected during different seasons were evaluated for antimalarial activity against Plasmodium yoelii 17XL in mice. The extract of leaves collected in August demonstrated high antimalarial activity, and all mice that received
the extract survived the infection. In contrast, the extract of leaves collected in December showed little activity. The extract
of buds collected in December cleared parasites, but with subsequent mortality to mouse. The present results show that the
amount of antimalarial agent—febrifugine and isofebrifugine mixture—in H. macrophylla var. Otaksa is both part- and season-dependent, suggesting that the choice of plant parts and their harvesting season are important factors

Background Pityrosporum ovale is a common saprophyte on the skin capable of inducing IgE antibody production in atopic dermatitis (AD) patients. Allergens ofP. ovale have been examined in several studies, but consensus on them is lacking. Objective This study was carried out to obtain more information about the IgE antibody response against P. ovale. including niunnun. Methods Sera from 64 AD patients and 10 healthy controls were analysed with immuno-blotting and the nitrocellulose radio allergosorbent test (RAST) method specifically developed to detect antimannan P. ovale IgE antibodies. Results In immunoblotting a total of 39 different IgE stained protein bands were seen. A high molecular weight staining was also seen especially in patients who displayed elevated mannan P. ovale RAST values. The most commonly stained protein bands in immunoblotting were 9 and 96 kD bands with antibodies in 73 and 65% of AD patients who had been positive in commercial P. orbiculare RAST with total serum IgE less than 4000 kU/I. Mannan RAST appeared positive in 77% of them. Positive immunoblotting to either of these bands was seen in 90% and, if added with staining with ihe 20 kD band, in 100% of these AD patients. A comhination of 9 kD IgE staining and mannan P. ovale RAST was positive in 92% of the patients and % kD and mannan P. ovale RAST in 85% of the patients. Conclusion It is evident that P. ovale has several allergens, the 9. 96 and 20 kD regions being the most important. According to our results mannan is also an important allegen of P. ovale 相似文献

用Protein A-Sepharose亲和层析法提纯McAb-IgG,皮下多点和腹腔免疫雄性Wistar大鼠,间隔两周共3次,融合前3天采用腹腔、尾静脉和脾内注射等三种加强免疫方法,以间接ELISA法和抗体竞争ELISA法筛选Anti-Id阳性克隆。结果显示:融合前大鼠血清抗小鼠IgG或抗免疫原McAb-IgG滴度,静脉组和脾内组比腹腔组高一个滴度;Anti-Id阳性克隆出现频率,腹腔组为0.9%和0.3%,静脉组为11.0%和13.3%,脾内组为16.4%和12.9%,静脉组和脾内组明显高于腹腔组(P<0.01);上述三种加强免疫方法对抗小鼠IgG阳性克隆出现频率无明显影响(P>0.05);与腹腔加强免疫组相比,静脉和脾内加强免疫组也可提高细胞融合效果(P<0.01).本实验结果提示,腹腔和皮下多点多次长程基础免疫结合静脉或脾内加强免疫是制备Anti-Id McAb比较好的免疫方案。 相似文献

对包括慢性肝炎、肝炎肝硬变和原发性肝癌在内的100例慢性HBV感染者作了HBsAg亚型分析。结果显示,在这组病例中adr亚型占61%,adw占33%,ayr占2%,adwr占4%。adr亚型有随年龄增长更占优势的趋向。adr和adwr亚型感染者HBsAg滴度较adw组显著增高,且e抗原阳性率亦较高。上述发现提示,具有adr和adwr亚型的HBV株可能具有较强的复制活性,不易被清除,从而导致受感染者 相似文献

In order to explore which amino acids or which blocks of amino acids in the 29 amino acid neuropeptide galanin are important for recognition of the endogenous ligand by galanin receptor subtypes present in the jejunum and in the hypothalamus, respectively, we have carried out L-Ala substitutions of individual amino acids or of blocks of amino acids in the rat galanin sequence and examined the binding of the obtained analogs to the rat hypothalamic and jejunal galanin receptor subtypes. This study reveals that the galanin sequence YLLGPH9–14 is essential for recognition of galanin by both the rat hypothalamic and jejunal galanin receptor subtypes. Substitution of the N-terminal amino acids, GWTL1–4, leads to total loss of affinity of galanin for both hypothalamic and jejunal galanin receptors. The α-helical C-terminal amino acid (25–29) part of galanin has no greater influence on the affinity of galanin to the hypothalamic galanin receptor subtype. L-Ala substitution of the C-terminal amino acids of galanin KHGLT25–29 shows, however, that this C-terminal motif is essential for the recognition by the jejunal galanin receptor subtype, whereas amino acids in the middle portion of galanin NSAG5–8 are of importance for binding to the hypothalamic but not to the jejunal receptor. [Ala5–8] Galanin thus has a more than 100-fold higher affinity to jejunal receptor than to the hypothalamic receptor, while [Ala25–29] galanin has a more than 100-fold higher affinity for the hypothalamic than for jejunal galanin receptor subtypes. pH dependence of the galanin binding to these receptor subtypes is also different. © Munksgaard 1997. 相似文献
