目的 探讨不同年龄条件下的α1 抗糜蛋白酶 (AACT)信号肽基因多态、α2 巨球蛋白(A2M )基因外显子 2 4缬氨酸 /异亮氨酸 (V/I)多态与帕金森病 (PD)发病风险间的关系。 方法 采用聚合酶链式反应限制性片断长度多态性方法 (PCR RFLP) ,在 66例PD患者、12 0例健康对照者中观察AACT信号肽基因多态、A2M基因V/I多态的分布 ,并通过比值比 (OR)对两种多态与PD作相关分析。 结果 PD患者与健康老人间不存在AACT A/T等位基因多态分布的差异 (P >0 0 5 ) ,但PD患者AACT A/A纯合子基因型与PD正关联 (OR =2 44 ,P <0 0 5 ) ,而且这种正相关主要存在于 <60岁的PD患者中 (OR =12 5 7,P <0 0 5 ) ;PD患者中A2M I等位基因的频率明显低于对照组(OR =0 3 9,P <0 0 5 ) ,且PD患者A2M I/I纯合子基因型与PD负关联 (OR =0 3 2 ,P <0 0 5 ) ,这种负关联主要存在于≥ 60岁的PD患者中 (OR =0 3 1,P <0 0 5 )。 结论 AACT A/A纯合子基因型可能是PD发病的风险因子 ,而A2M I等位基因则有可能是PD发病的保护因子。  相似文献

目的了解α-2-巨球蛋白(A2M)对人胚肺二倍体成纤维细胞(2BS细胞)衰老过程的影响. 方法将正、反义真核表达载体,经脂质体(lipofectin)介导,转染年轻2BS细胞,分别获得正、反义转染细胞.经Southern及Northern印迹验证后,测定正、反义转染细胞的衰老相关指标,再用Western印迹法检测转染细胞中p16、p21的变化. 结果转染的外源性A2M cDNA片段已完全整合到基因组中.与对照2BS/neo(空载体)细胞和正常细胞相比,正义A2M表达载体转染2BS细胞得到的稳定细胞(2BS/A2Ms细胞)生长更为迅速,S期和M期的比例略高于对照2BS/neo细胞和正常细胞组.衰老相关的β-半乳糖苷酶(SA-β-Gal)染色阳性率约为55.4%,而且细胞寿限较正常细胞和对照细胞组略有延长,但无统计学意义(P>0.05).与之相反,反义A2M表达载体转染2BS细胞得到的稳定细胞(2BS/A2Ma细胞)较对照和正常细胞组生长更为缓慢,S期和M期的比例略低,SA-β-Gal染色阳性率约为72.3%,细胞形态扁平、增大,表现为衰老细胞的特征,细胞寿限略短于对照和正常细胞组,但也无统计学意义(P>0.05).此外发现2BS/A2Ms细胞的生长速率、S期和M期的比例、细胞寿限均明显高于2BS/A2Ma细胞(P<0.01).比较正、反义转染细胞最大传代次数差异有统计学意义(P<0.05).转染细胞中p16、p21的表达变化并无统计学意义. 结论α-2-巨球蛋白延缓2BS细胞衰老作用甚微,且不经过p16、p21基因起作用.  相似文献

To investigate the effect of modification of biological environmental conditions, one of the factors influencing the healing of anterior cruciate ligament rupture, we performed experimental anterior cruciate ligament ruptures on New Zealand rabbits. After experimental rupture, intra-articular alpha-2 macroglobulin was injected into the knees of the rabbits in the experiment group to prevent structural changes resulting from the enzymatic reactions in the ruptured anterior cruciate ligament. At the end of 10th day of the experiment, we observed that the anterior cruciate ligaments in the experiment group had retained their prerupture brightness and volume when compared with the control group in which intra-articular alpha-2 macroglobulin had not been injected. We also noted that the anterior cruciate ligaments in the experiment group had not retracted or swollen, the incision sites were regular and clean, and they did not show any signs of degeneration. In the histological examination, the anterior cruciate ligaments in the control groups showed disruption of the collagen network and a significant diminution in number of fibroblasts and fibrocytes (p <. 001). At the end of this study, we concluded that the necessary conditions for the healing and repair of ruptured anterior cruciate ligament could exist if the enzymatic and biological environments were under control.  相似文献

Objective

To explore influence of electroacupuncture (EA) therapy of tonifying the kidney and regulating governor vessel on amyloid beta (Aβ) related degradation enzymes in the hippocampus of a rat model of Alzheimer's disease (AD) induced by Aβ1-42.

Methods

Forty Wistar male rats were randomly divided into 4 groups: a normal group, a sham operation group, a model group and an EA group, 10 rats in each one. The rats in normal group were normally fed. The rats in sham operation group were bilaterally injected in the hippocampus with 5 µL of saline and they were normally fed after the injection. The rats in the model group and the EA group were bilaterally injected in the hippocampus with 5 µL of Aβ1-42 on each side. Rats in the EA group received EA of 5?Hz continuous wave at the “B?ihuì (百会 GV20)” and bilateral “Shènshū (肾俞 BL23)” for a duration of 15 min per time every day and continuously for 15 days. After 15 days, the hippocampal expression levels of insulin degrading enzyme (IDE), lipoprotein (LPL), transthyretin (TTR), apolipoprotein E (APoE), α2 macroglobulin (α2M) and Aβ1-42 of the 4 groups were tested by Western blot.

Results

Compared with the sham operation group, the expression levels of IDE, LPL, TTR, APoE and α2M in the hippocampus were significantly lower (P?<?0.05, P?<?0.01) and the expression of Aβ1-42 was significantly higher(P?<?0.01) in the model group. Compared with the model group, the expression levels of IDE, LPL, TTR, APoE and α2M in the hippocampus of these rats were significantly lower (P?<?0.05, P?<?0.01), the expression of Aβ1-42 was significantly higher(P?<?0.01) in the EA group.

Conclusion

EA therapy of tonifying the kidney and regulating governor vessel can enhance the expression of IDE, LPL, TTR, APoE, and α2M in the hippocampus of AD rats injected by Aβ1-42, and may consequently promote the degradation of aβ1-42 to help improve the pathological manifestations of AD and therefore delay its progression.  相似文献
