The expression of MHC isoforms in the skeletal muscles of nine patients with Duchenne muscular dystrophy (DMD) (from 2.5 to 15 yr of age) and three DMD carriers was studied using different specific anti-MHC MAbs. We also analyzed muscle fiber size and fiber reactivity with acridine orange and/or with a surface antigen marker. One-quarter of all fibers of DMD patients, or less with age, were of normal size and contained only adult slow MHC. Half of the muscle fibers contained adult and developmental MHCs. Only half of these fibers were representative of an active regenerative process. MHC co-expression also altered the proportion of normal fast or slow fibers. Adult fast MHCs were expressed as unique MHC only in small and very small fibers in the oldest DMD patients. In DMD carrier muscles, the greatest alterations in MHC expression were observed in patients with the most reduced dystrophin expression. However, MHC changes in dystrophin-positive fibers were similar to those observed in dystrophin-free fibers. In conclusion, disruptions or delays in the switching of all genes coding for adult fast and slow MHC and developmental MHC coincided with dystrophin deletion and with perturbations in its expression.  相似文献

目的:探讨心肌细胞内游离钙([Ca^2 ]i)和心肌ATP酶变化在心肌挫伤(MC)后心功能障碍发生机制中的意义。方法:随机选取兔36只,分为6组:正常对照、伤后2、4、8、12、24h,每组6只。经右颈总动脉插管至左心室测左室压力。采用BIM-II型生物撞击机致成MC模型。在上述时相点测定左室压力,并测定心肌细胞内[Ca^2 ]i和心肌匀浆组织及线粒体ATP酶活力。结果:心脏收缩/舒张功能受到损害,心肌细胞内[Ca^2 ]i升高,心肌匀浆组织及线粒体ATP酶下降,相关分析表明心功能障碍与心肌细胞内[Ca^2 ]i含量升高呈明显负相关,与ATP酶活性降低呈明显正相关。结论:MC后心脏功能下降,心肌细胞内[Ca^2 ]i含量升高和ATP酶活性下降可能是其原因之一。  相似文献

目的从电生理、酶学角度探讨微波照射对海马细胞的影响机理。方法用 2 4 5 0MHz连续波照射理疗机为照射源 ,以小白鼠为对象 ,观察微波照射强度为 1 0mW /cm2 时小鼠海马细胞膜Na+,K+ AT Pase、Ca2 +,Mg2 + ATPase活性、电压门控型Na+、K+、Ca2 +通道的变化情况 ,分别用组织化学染色法和膜片钳方法测定ATPase活性和离子通道功能。结果 1 )照射组海马Na+,K+ ATPase活性与对照组无显著差别 ,Ca2 +,Mg2 + ATPase活性比对照组显著降低 (P <0 .0 5 ) ;2 )照射组静息电位未发生显著变化 ,电压门控型Na+、K+、Ca2 +电流的诱导发生率显著地低于对照组 ,Na+电流峰值所在膜电位向去极化方向偏移 ,Na+电流衰减速率减慢 ,A电流的发生率显著低于对照。结论 2 4 5 0MHz微波照射小鼠 ,在 1 0mW /cm2 时 ,细胞的生存不会受影响 ,但海马细胞膜Ca2 +,Mg2 + ATPase活性受到抑制 ,电压门控型Na+、K+、Ca2 +离子通道受到损害 ,有可能影响学习记忆功能  相似文献

Ventricular-like and fast myosin heavy chains (VL-MHC and FMHC) are transiently expressed during slow skeletal muscle development. The influence of innervation on repression of these MHC isoforms is investigated over an 84-day time course in: (1) normal anterior latissimus dorsi (N-ALD) muscles, (2) regenerating ALD (R-ALD) muscles, (3) denervated ALD (D-ALD) muscles, and (4) regenerating and denervated ALD (RD-ALD) muscles. Western blotting demonstrates that the VL-MHC is expressed in R-, D-, and RD-ALD muscles, but not in N-ALD muscles. Expression of the VL-MHC is transient in R-ALD muscles. In contrast, VL-MHC expression persists in RD-ALD muscles, and appears with time in D-ALD muscles. FMHC was not detected in N-ALD muscles by Western blotting. Two FMHCs are seen in R-ALD and RD-ALD muscles, and in 13-day embryonic ALD muscles. The slower migrating FMHC (FMHCA) comigrates with developmentally regulated FMHCs in fast pectoralis muscle, while the faster migrating FMHC (FMHCB) comigrates with the faster migrating FMHC in embryonic ALD muscle (13 days in ovo). FMHCB decreases in amount over the time course in R-ALD muscles, while FMHCA persists. In contrast, substantial levels of both FMHCs persist in RD-ALD muscles, and appear with time in D-ALD muscles. The cellular distribution of MHCs is followed by immunocytochemistry. Regenerating cells expressing VL-MHC and FMHC are replaced by a mature population in R-ALD muscles. Some of the mature myofibers in R-ALD muscles express FMHC, but not VL-MHC. In RD-ALD and D-ALD muscles, both regenerating and mature muscle cells are seen which express VL-MHC and FMHC. Our results indicate that innervation is required for the repression of VL-MHC and FMHCB during regeneration of slow muscle.  相似文献

To probe mechanisms of cadmium (Cd) damage to the lung extracellular matrix (ECM), we developed Cd-resistant (CdR) rat lung fibroblasts (RFL6) by incubation with graded concentrations of Cd. CdR cells downregulated lysyl oxidase (LO), a copper (Cu)-dependent enzyme essential for crosslinking of collagen and elastin in the ECM, in conjunction with upregulation of other Cu-binding proteins including Cu,Zn-superoxide dismutase (SOD1), copper chaperone for SOD1 (CCS1), metallothionein (MT), and Menkes P-type ATPase (ATP7A), a Cu transporter in the membrane of the Golgi apparatus, as well as gamma-glutamylcysteine synthetase (gamma-GCS), an enzyme for glutathione biosynthesis. Reduction and loss of cytoplasmic distribution of LO in CdR cells were accompanied by its dislocation with the Menkes P-type ATPase and the endoplasmic reticulum marker. CdR cells displayed a defect in LO catalytic activity but an enhancement in Cu,Zn-SOD catalytic activity consistent with the protein expression levels of these enzymes. Although long-term Cd exposure of cells enhanced the Menkes P-type ATPase protein expression, actually, it reduced Cu-dependent catalytic activity of this enzyme in parallel with the deficiency of LO. The low level of 64Cu bound to the LO fraction and the high level of 64Cu bound to the MT fraction provide direct evidence for limitation of Cu bioavailability for LO existing in the CdR cells. These results suggest that downregulation of LO is linked with upregulation of other Cu-binding proteins and with alteration in Cu homeostasis in the CdR phenotype.  相似文献

体外测定青蒿琥酯钠(SA)对大鼠红细胞膜Na~( )-K~( )-交换ATP酶活性的影响.方法:在反应系统中分别加入不同浓度的SA(0,0.5,1,5和10 μmol·L~(-1)),通过测定反应系统中释放的无机磷含量,计算酶活性.结果:随着SA浓度(O,0.5,1,5和10 μmol·L~(-1))的增高,对Na~( )-K~( )-交换ATP酶活性的抑制作用也随之增强,抑制率分别为15%,29%,46%和75%.将底物ATP浓度增加为125,250,375和500 μmol·L~(-1),进行了酶的动力学测定.用直线回归分析作Eadie-Hofstee动力学曲线,结果表明,SA对该酶的抑制作用为竞争性抑制.结论:提示SA可影响宿主红细胞膜的离子转运及膜的功能.  相似文献

Clinically, phenol is used often as a neurolytic agent to treat pain and spasticity. The purpose of this study was to examine the time course of denervation and recovery in several hindlimb muscles following application of a 5% aqueous solution of phenol to the sciatic nerve. Phenol was applied to the sciatic nerve of adult female rats either by intraneural or perineural injection. Axonal degeneration was evident within the sciatic nerve 2 days following phenol application, although variable amounts of damage were observed. By 2 weeks, the soleus and tibialis anterior had atrophied to 63% and 51% of control. Reinnervation of hindlimb muscles occurred between 2 and 4 weeks following the nerve block. Following denervation, the soleus became slower in that all of the fibers expressed the slow myosin heavy chain (MHC). At 5 months, maximum tension of the soleus was 74% of control and the muscle consisted of more fast fibers on average, some of which expressed IIx MHC. These data suggest that 5% phenol causes an injury to the nerve that is more severe than a crush injury, and that reinnervation of denervated muscles may be by motoneurons other than those that originally innervated the muscles. © 1996 John Wiley & Sons, Inc.  相似文献

采用一种改进的方法,从100g猪心室肌中提取心肌肌球蛋白370mg,并进一步分离和纯化其亚基成分,分别得到肌球蛋白轻链24mg和重链210mg,核酸含量<1%,不含有肌动蛋白、原肌球蛋白、肌钙蛋白及其它杂蛋白;SDSPAGE显示,猪心肌球蛋白重链为200~220KD一条带,肌球蛋白轻链则为27kd、21.4kd、19kd三条带;纯化猪心肌球蛋白的Ca2+激活ATP酶活性为0.24μmolPi/(mg·min),但其分离后的亚基则无此活性。  相似文献
