The fat mass and obesity-associated gene (FTO) rs9939609 A-allele is linked to obesity and dyslipidemia, yet the independent influence of this polymorphism on blood lipids remains equivocal. We examined the influence of the FTO rs9939609 polymorphism on fasting and postprandial blood lipids in individuals homozygous for the risk A-allele or T-allele with similar anthropometric and demographic characteristics. 12 AA and 12 TT males consumed a standardized meal after fasting overnight. Blood samples were collected at baseline (−1.5 h), before the meal (0 h), and for five hours postprandially to measure lipid, glucose, and insulin concentrations. Time-averaged total area under the curve (TAUC) values (0–5 h) were calculated and compared between genotypes. Fasting triacylglycerol (TG), high-density lipoprotein cholesterol, low-density lipoprotein cholesterol, total cholesterol, non-esterified fatty acid (NEFA), glucose, and insulin concentrations were similar between groups (p ≥ 0.293). TAUC for TG was similar in AAs and TTs (95% confidence interval (CI) −0.52 to 0.31 mmol/L/h; p = 0.606). Likewise, TAUC values were similar for NEFA (95% CI −0.04 to 0.03 mmol/L/h; p = 0.734), glucose (95% CI −0.41 to 0.44 mmol/L/h; p = 0.951), and insulin (95% CI −6.87 to 2.83 pmol/L/h; p = 0.395). Blood lipids are not influenced by the FTO rs9939609 polymorphism, suggesting the FTO-dyslipidemia link is mediated by adiposity and weight management is important in preventing FTO-related lipid variations.  相似文献

Metabolic pathways leading to lipid biosynthesis in four different developmental stages of Schistosoma mansoni were explored and quantified by incubation in the presence of labeled precursors in a chemically defined medium. At the schistosomulum stage and in male, female, or paired worms, glycerol and oleate incorporation into neutral lipids, mainly in the form of triacylglycerols, was greater than into phospholipids, whereas in 11-and 15-day-old worms, synthesis mainly led to phospholipids. Incorporation into phospholipids was recovered largely in phosphatidylcholine, and distribution into other phospholipids depended on the developmental stage. Incorporation of choline and ethanolamine into their respective phospholipids represented up to 15% of the parasitic phospholipid content. The formation of phosphatidylcholine by phosphatidylethanolamine methylation occurred mainly in the immature parasitic stages. Inositol incorporation was also measurable, whereas [14C]serine incorporation was low or undetectable. Addition of 1-palmitoyl-2-[14C]oleyl phosphatidylcholine revealed a very high uptake of this phospholipid by the immature stages but further metabolism was not detectable. In contrast, adult S. mansoni were completely unable to take up or absorb this exogenous phospholipid. The most striking aspect of this study was the relatively high metabolic activity in 11-day-old worms and the lower but sustained activity on day 15 and at the schistosomulum stage. By comparison, biosynthetic activity in adult S. mansoni, on which research studies have been focused until now, was very low. We also discuss the participation of lipid metabolism in the constant renewal of the membrane complex which is essential to parasitism by S. mansoni.  相似文献

Aim: Hormone‐sensitive lipase (HSL) has been proposed to regulate triacylglycerol (TG) breakdown in skeletal muscle. In muscles with different fibre type compositions the influence on HSL of two major stimuli causing TG mobilization was studied. Methods: Incubated soleus and extensor digitorum longus (EDL) muscles from 70 g rats were stimulated by adrenaline (5.5 μm , 6 min) or contractions (200 ms tetani, 1 Hz, 1 min) in maximally effective doses or by both adrenaline and contractions. Results: Hormone‐sensitive lipase activity was increased significantly by adrenaline as well as contractions, and the highest activity (P < 0.05) was seen with combined stimulation [Soleus: 0.40 ± 0.03 (SE) m‐unit mg protein?1 (basal), 0.65 ± 0.02 (adrenaline), 0.65 ± 0.03 (contractions), 0.78 ± 0.03 (adrenaline and contractions); EDL: 0.18 ± 0.01, 0.30 ± 0.02, 0.26 ± 0.02, 0.32 ± 0.01]. Glycogen phosphorylase activity was always increased more by adrenaline compared with contractions [Soleus: 60 ± 4 (a/a + b)% vs. 46 ± 3 (P < 0.05); EDL: 60 ± 5 vs. 39 ± 6 (P < 0.05)]. After combined stimulation glycogen phosphorylase activity in soleus [59 ± 3 (a/a + b)%] was identical to and in EDL [45 ± 4 (a/a + b)%] smaller (P < 0.05) than the activity after adrenaline only. Conclusions: In slow‐twitch oxidative as well as in fast‐twitch glycolytic muscle HSL is activated by both adrenaline and contractions. These stimuli are partially additive indicating at least partly different mechanisms of action. Contractions may impair the enhancing effect of adrenaline on glycogen phosphorylase activity in muscle.  相似文献

目的分析在高脂血症患者治疗中应用阿托伐他汀和辛伐他汀的降脂效果。方法将2017年1月—2020年1月在我院医治的90例高脂血症患者随机设为两个组别,给予对照组(45例)辛伐他汀治疗,给予试验组(45例)阿托伐他汀治疗。观察两组患者治疗前后血脂水平和生化指标变化情况。结果试验组治疗后总胆固醇、三酰甘油、低密度脂蛋白胆固醇低于对照组,高密度脂蛋白胆固醇高于对照组,两组对比差异具有统计学意义(P<0.05)。两组患者治疗后肌酐、尿素氮指标差异无统计学意义(P>0.05)。结论阿托伐他汀和辛伐他汀应用于高脂血症患者治疗中均能起到不同程度的降脂效果,但阿托伐他汀的效果更好,安全可靠。  相似文献

Influence of large molecular polymeric pigments (LMPP) isolated from fermented Zijuan tea on the activity and mRNA expression of key enzymes involved in lipid metabolism in rat was explored. The results show that intragastric infusion of high-dose LMPP (1.215g/kg body weight) effectively suppressed the elevation in TC and LDL-C (p<0.05), and prevented the reduction in HDL-C (p<0.05), compared with the hyperlipidemia model group. LMPP significantly enhanced the activity of HL and HSL, and increased the HSL mRNA expression in the liver tissue and adipose tissue. High-LMPP treatment significantly reduced the HMG-CoA reductase expression by 56.5% in the liver compared with hyperlipidemia model group. In contrast, LDL-R expression was increased by 120% in the presence of high-LMPP treatment. These results suggest that LMPP have the hypolipidemic effect to some extent and significantly enhance HSL mRNA expression in the liver and adipose tissue, thereby increasing HSL activity in rat.  相似文献

目的:观察清脑汤治疗H型高血压的临床疗效及对同型半胱氨酸、血脂的影响。方法:选择2016年10月至2018年12月在本院就诊的168例H型高血压患者为研究对象,按照随机数字表法分为对照组和治疗组,每组各84例,对照组采用非洛地平缓释片治疗,每次5 mg,每日清晨口服,治疗组采用中药清脑汤治疗,日1剂,水煎服,分2次温服。两组均以7 d为1个疗程,均连续治疗4个疗程。观察两组患者治疗前后血压、血脂水平和心功能。结果:治疗后,治疗组日间收缩压、夜间收缩压、日间舒张压、夜间舒张压较对照组明显降低,差异有统计学意义(P<0.05);治疗后,治疗组同型半胱氨酸(homocystein,Hcy)、总胆固醇(total cholesterol,TC)、高密度脂蛋白(high density lipoprotein-C,HDL-C)及三酰甘油(triacylglycerol,TG)水平与对照组比较明显降低,差异有统计学意义(P<0.05);对照组不良反应发生率为16.67%,治疗组不良反应发生率为5.95%,两组不良反应发生率比较,差异具有统计学意义(P<0.05);治疗组每搏输出量(stroke volume,SV)、左心室射血分数(left ventricular ejection fraction,LVEF)高于对照组(P<0.05),左心室舒张末期内径(left ventricular end-systolic dimension,LVEDD)低于对照组(P<0.05);对照组有效率为73.81%,治疗组有效率为90.48%,两组有效率比较,差异具有统计学意义(P<0.05)。结论:清脑汤可以提高H型高血压患者的临床疗效,降低同型半胱氨酸及血脂的浓度。  相似文献

A water-soluble extract of a traditional Chinese black tea (Pu-Ehr) has been shown to precipitate mixed bile salt micelles in foods. In addition, long-term ingestion of this black tea extract (BTE) significantly reduces blood cholesterol levels in rats. We investigated the effects of BTE tablets (a formula designed to enhance compliance) as a dietary supplement in a 3-month double-blind randomized group comparison study in borderline hypercholesterolemic human subjects (n = 47). All subjects ingested BTE tablets (333 mg) or placebo 3 times daily before meals for 3 months. In the BTE-treated group, the initial mean blood total (6.14 ± 0.14 mol/L) and low-density lipoprotein (LDL) cholesterol (4.32 ± 0.14 mol/L) levels decreased with time and were significantly (P < .01) lower (total cholesterol, 5.62 ± 0.11; LDL cholesterol, 3.81 ± 0.13 mol/L) after 3 months of ingestion. Furthermore, the mean body weights (P < .05) and triacylglycerol levels (P < .01) were also significantly reduced after 3 months of BTE intake compared with the baseline levels. Significant improvements in the mean LDL cholesterol, body weight, and triacylglycerol values were not accompanied with undesirable changes in other biochemical parameters measured in the subjects. None of the subjects complained of any adverse effects (eg, abdominal distension). The results indicate that BTE intake elicited a significant antihypercholesterolemic effect and might be useful for improving blood cholesterol levels in subjects at risk for heart disease or obesity.  相似文献

Hyperlipidemia and hyperhomocysteinemia are regarded as major risk factors for cardiovascular disease. Medical nutrition therapy (MNT), which involves a combination of nutritional diagnosis, therapy, and counseling for at-risk patients, has been proposed as a protocol to control these risk factors. The purpose of this study was to investigate the effect of an intensive MNT, specifically tailored to Korean hyperlipidemic patients, on levels of serum lipids and plasma homocysteine. Forty hyperlipidemic patients with blood levels of total cholesterol ≥200 mg/dL or triacylglycerol ≥150 mg/dL were recruited from the Kyung Hee University Medical Center (Seoul, Korea). They were randomly divided into control (no treatment) or experimental (MNT treatment) groups. A registered dietitian performed MNT, emphasizing dietary folate intake based on the guidelines for Korean hyperlipidemic patients; this was performed 5 times during the 12 weeks of the experiment. After the MNT period, we observed that body mass index (P < .001) and fat percentage (P < .05) was significantly decreased in the experimental group but not in the control group. Daily energy consumption, as well as carbohydrate, fat, and cholesterol intakes, all decreased significantly (P < .05) in the MNT group, whereas folate intake increased (P < .05). Blood levels of triacylglycerol (−30.8%), total cholesterol (−20.9%), low-density lipoprotein cholesterol (−14.5%), and ratio of low-density lipoprotein/high-density lipoprotein (−17.1%) were significantly lower in the experimental group after MNT (P < .001) compared with the control group. Plasma homocysteine levels were unchanged (P = .98); however, the levels of folate (P < .01), vitamin B6 (P < .01), and vitamin B12 (P < .05) increased significantly in the experimental group. These results indicate that MNT, with an emphasis on folate intake, can be effective at reducing blood levels of lipids and improving the blood levels of folate, vitamin B6, and vitamin B12 in Korean hyperlipidemic patients.  相似文献
