86对母婴血清标本用间接酶联免疫测定法(ELISA)测定了乙型脑炎抗体,118对用血凝抑制(血抑)试验测定了麻疹抗体。结果发现,脐血乙型脑炎抗体阳性率为79.1%,母血为74.4%,几何平均滴度在脐血显著高于母血(t=5.1857,P<0.01),比值为1.2∶1。脐血和母血的麻疹血抑抗体阳性率分别为86.4%及84.8%,几何均值也是前者高于后者(t=4.0349,P<0.01),比值为1.3∶1。 108份婴儿血清标本测定结果显示,0~3个月婴儿乙型脑炎抗体阳性率为35%,4~9个月仅为13%,10个月及以上的婴儿全部阴性。此结果与近年上海市小于1岁组乙型脑炎发病率最高相一致,提示应考虑提早乙型脑炎疫苗的接种年龄。  相似文献

目的 :探讨母血中胎儿有核红细胞数量与胎儿异常的关系 ,为将它用于无创性产前诊断提供实验依据。方法 :对孕 6~ 40周的 86例妇女 (包括胎儿正常组 68例、胎儿异常组 1 8例 )的外周血进行单密度梯度离心、瑞氏染色和细胞计数 ,分析母血中胎儿有核红细胞数量与孕周、胎儿异常及胎儿性别的关系。结果 :胎儿正常组 ,母血中胎儿有核红细胞平均数量为 1 4.2 3±1 2 .0 1 /ml,与妊娠周数呈直线相关 (R >R0 .0 5)。胎儿异常组中 ,母血中胎儿有核红细胞平均数量较正常胎儿组显著增加 ,约为 3 8.73± 2 4.97/ml,且与妊娠周数无明显直线相关性。母体外周血中胎儿有核红细胞数量与胎儿性别无统计学相关性。结论 :胎儿发生异常时母血中胎儿有核红细胞数量较正常妊娠时增加。母血中胎儿有核红细胞数量在胎儿正常时随孕周而增加 ,且不受胎儿性别的影响。母血中胎儿有核红细胞计数可以辅助用于产前筛查胎儿异常的发生。  相似文献

The majority of infectious bursal disease virus (IBDV) strains circulating in the broiler chicken industry in Canada are variant strains (varIBDV). Despite high levels of maternally derived antibodies (MtAb), the circulating varIBDVs can establish infection and cause severe immunosuppression in broiler chicks. The objective of this study was to evaluate circulating varIBDVs as broiler breeder vaccine candidates and investigate their protective efficacy against varIBDV challenge in their progeny chicks. Six groups of breeders (20 females/group) were vaccinated with varIBDV strains, SK09, SK10, SK11, SK12, and SK13 or saline at the age of 13 weeks and antibody response was determined by ELISA at 3–7-, and 20- weeks post-vaccination. We also included commercial chicks for the comparison. Results showed that SK-09 is the most antigenic strain, followed by SK-10, SK-12, and SK-13. In contrast, SK-11 showed the lowest antibody response, and over time, antibody titers steadily decreased. Eggs from breeders were collected at 21-week post-vaccination and incubated to produce their respective progenies. The serum antibody titer in day-old chicks showed a successful MtAb transfer. Progeny chicks (n = 40/group) were orally challenged with varIBDV-SK-09 strain at 6 days of age and serum antibody titer (19 d and 35 d of age), bursa to body weight ratio (19 d and 35 d of age), bursal viral load (9 d and 19 d of age) was examined to assess the protection against IBDV. Following the challenge, we found a significant increase in the antibody titers in MtAb-free and commercial vaccine groups than in the varIBDV groups, both at 19 d and 35 d of age. The BBW ratio and viral load data indicated a significant homologous and heterologous protection against varIBDV-SK-09 challenge by SK-09 and SK-10 MtAbs, respectively. Overall, this study demonstrated the feasibility of developing breeder vaccines using circulating varIBDV as candidate vaccine antigens.  相似文献

目的 探讨母系表达基因3(maternally expressed gene 3,MEG3)对糖尿病视网膜病变(DR)Müller细胞的活化及炎症因子分泌的影响及机制。方法 高脂饮食联合链脲佐菌素腹腔注射构建小鼠DR体内模型。高糖刺激人视网膜Müller细胞株MIO-M1构建DR体外模型。免疫荧光化学染色及Western blot检测小鼠视网膜及Müller细胞中胶质纤维酸性蛋白(GFAP)的表达。Western blot及酶联免疫吸附实验(ELISA)检测小鼠视网膜及细胞培养基上清中血管内皮生长因子(VEGF)蛋白的表达。ELISA检测小鼠视网膜及细胞培养基上清中IL-1β蛋白的表达。分别或同时向Müller细胞中转染pcDNA-MEG3及miR-34a mimic对其表达进行干预。qRT-PCR检测MEG3 mRNA及miR-34a表达。结果 与正常对照组小鼠比较,DR组小鼠视网膜中GFAP、VEGF及IL-1β蛋白表达均增多(均为P<0.05),MEG3 mRNA表达降低(P<0.01)。与对照组比较,高糖组Müller细胞中MEG3 mRNA表达降低(P<0.01),而GFAP、VEGF及IL-1β蛋白表达均升高(均为P<0.05)。与高糖组比较,高糖+pcDNA-MEG3组中GFAP、VEGF及IL-1β蛋白表达均降低(均为P<0.05)。与正常对照组比较,DR组小鼠视网膜及高糖刺激的Müller细胞中miR-34a表达均升高(均为P<0.05)。与pcDNA组比较,pcDNA-MEG3组中miR-34a表达减少(P<0.01)。与pcDNA+NC mimic组比较,pcDNA+miR-34a mimic组中GFAP、VEGF及IL-1β蛋白表达均升高(均为P<0.05),而pcDNA-MEG3+NC mimic组中GFAP、VEGF及IL-1β蛋白表达均减少(均为P<0.05)。与pcDNA-MEG3+miR-34a mimic组比较,GFAP、VEGF及IL-1β蛋白表达水平均升高(均为P<0.05)。结论 MEG3在DR小鼠视网膜及高糖刺激的Müller细胞中表达均降低,其可通过负向调控miR-34a抑制Müller细胞活化及炎症因子的分泌。过表达MEG3可能成为DR治疗的新靶点。  相似文献

BACKGROUND: The pathophysiological mechanism of diabetes mellitus in the presence of the 3243 A-G tRNALEU(UR) mitochondrial DNA mutation is thought to result from deficient insulin secretion. However, few subjects with normal glucose tolerance have been studied to determine the sequence of events resulting in the development of diabetes mellitus. AIM: To determine whether abnormalities of insulin sensitivity, insulin secretion or glucose effectiveness are present in non-diabetic subjects with the 3243 A-G tRNALEU(UUR) mitochondrial DNA mutation. METHODS: Twelve non-diabetic subjects with the mutation were compared with 12 controls, matched for age and anthropometric parameters, using both oral and intravenous glucose tolerance tests, the latter with Minimal Model analysis. RESULTS: Following an oral glucose load we found significantly higher blood glucose levels at 90 min and 120 min and significantly higher insulin levels at 120 min and 180 min in non-diabetic subjects with the mutation but no difference in the insulinogenic indices at 30 min and 180 min. From the intravenous glucose tolerance test there was no difference in overall glucose tolerance, insulin sensitivity, first- or second-phase insulin secretion, proinsulin secretion or glucose effectiveness. Insulin-independent glucose disposal was increased in subjects with lower insulin sensitivity and declined with increasing age in subjects with the mutation but not in controls. CONCLUSIONS: While there appear to be subtle defects of glucose handling in non-diabetic subjects with the 3243 mutation, these could not be explained by differences in insulin sensitivity or secretion.  相似文献

目的探究长链非编码 RNA母系表达基因 3(LncRNA MEG3)、微小 RNA-21(miR-21)与冠状动脉钙化的相关性。方法选取 2018年 6月至 2019年 6月在邯郸市中心医院行冠状动脉造影及 CT检查确诊为冠状动脉钙化病人 189例作为钙化组,另选取同期在该院行冠状动脉造影及 CT检查显示无冠状动脉钙化者 183例作为未钙化组。采用实时荧光定量 PCR(qRT-PCR)法检测血清 LncRNA MEG3、miR-21表达水平,采用 Pearson法分析病人血清 LncRNA MEG3、miR-21水平及与糖化血红蛋白(HbA1c)、高敏 C反应蛋白( hs-CRP)、肿瘤坏死因子 -α(TNF-α)、白细胞介素 -1β(IL-1β)水平相关性,采用 logistic回归模型分析冠状动脉钙化发生的影响因素,采用 ROC曲线评估血清 LncRNA MEG3、miR-21表达水平对冠状动脉钙化的预测价值。结  相似文献

Successful implementation of marker vaccines against classical swine fever virus is dependent on a reliable accompanying diagnostic assay that allows differentiation of infected from vaccinated animals (DIVA ) as well as the development of a testing scheme during emergency vaccination. In this context, special attention needs to be paid to breeding farms, because the offspring of marker vaccinated sows possess maternally derived antibodies (MDA s). So far, limited information is available on the influence of MDA s on serological testing in the context of a DIVA strategy. Therefore, two commercially available Erns antibody ELISA s were compared, using serum samples of piglets with a high‐to‐moderate titre of MDA s against marker vaccine CP 7_E2alf. False‐positive results were detected by both Erns antibody ELISA s for serum samples of piglets with an age of up to 4 weeks. Interestingly, most samples tested false‐positive in the first Erns antibody ELISA were identified correctly by the other Erns antibody ELISA and vice versa. In conclusion, in case of emergency vaccination of sows, the specificity of both ELISA s in newborn piglets younger than 4 weeks may be relatively low. This could be addressed in a testing strategy by either not sampling piglets up to the age of 4 weeks or using both ELISA s in a screening‐confirmation set‐up.  相似文献
