目的 对比研究朝鲜淫羊藿酸性多糖酯化还原前后的理化特性,并探讨其改善油酸诱导的肝癌HepG2细胞脂质堆积活性的差异。方法 采用高效凝胶渗透色谱法测定朝鲜淫羊藿酸性多糖(EFPA)的均一性和分子量,高效液相色谱法测定EFPA和酯化还原后朝鲜淫羊藿酸性多糖(EFPA-R)的单糖组成;采用油酸(OA)处理HepG2细胞诱导建立脂质蓄积模型,不同浓度EFPA与EFPA-R(10、30、100、300 μg·mL-1)分别和OA共同作用于细胞24 h,采用CCK-8试剂盒测定细胞存活率,油红O染色观察细胞内脂滴蓄积情况,并采用试剂盒测定细胞内总胆固醇(TC)、甘油三酯(TG)含量。结果 EFPA为成分均一的多糖组分,分子量为125.8 kDa,由甘露糖、葡萄糖、半乳糖、葡萄糖醛酸和阿拉伯糖组成,摩尔比为1.7∶7.4∶1.4∶1.8∶1.0,葡萄糖占比最大,EFPA-R由甘露糖、葡萄糖、半乳糖和阿拉伯糖组成,摩尔比为0.8∶10.6∶2.1∶1.0;在10-300 μg·mL-1范围内,EFPA和EFPA-R对HepG2细胞的抑制作用较弱,作为给药浓度;与空白组相比,模型组细胞中TC、TG含量显著升高(P < 0.01),细胞内红色脂滴显著增多,与模型组相比,EFPA可显著降低细胞中TC、TG含量(P < 0.01),明显减少细胞内红色脂滴(P < 0.05或P < 0.01),EFPA-R干预后细胞则无明显变化。结论 EFPA可明显改善HepG2细胞脂质堆积情况,且呈现剂量依赖性,而半乳糖醛酸(GalA)的存在可能是其抑制HepG2细胞脂质蓄积的关键因素。  相似文献

ObjectiveEach pulmonary segment is an anatomical and functional unit. However, it is fundamentally difficult to precisely distinguish every pulmonary segment using the conventional pulmonary intersegmental planes from computed tomography images. Building arteriopulmonary segments is likely to be an effective way to identify pulmonary segments.MethodsThe thoracic computed tomography images of 40 patients were collected. The anatomic structures of interest were extracted in the transverse, sagittal, and coronal planes using the semi-automated segmentation tools provided by Amira software. The intrapulmonary vessels were subsequently segmented and reconstructed. The distributions of the pulmonary arteries, veins, and bronchi were observed. In patients with pulmonary masses, the mass was also reconstructed.ResultsThe three-dimensional reconstructed images showed the branches of the pulmonary artery ramified up to their eighth order covering the entire lung as well as evident intersegmental gaps without pulmonary arteries. The segmental artery was closely accompanied by the segmental bronchi in 486 pulmonary segments (90% of total number of segments). The size and spatial location of the pulmonary mass within a pulmonary segment were also clearly visible.ConclusionsDemarcation of arteriopulmonary segments can be used to precisely distinguish every pulmonary segment and provide its detailed anatomical structure before pulmonary segmentectomy.  相似文献

Inappropriate use of acetaminophen (APAP) can lead to morbidity and mortality secondary to hepatic necrosis. Ginsenoside Rg1 is a major active ingredient in processed Panax ginseng, which is proved to elicit biological effects. We hypothesized the beneficial effect of Rg1 on APAP-mediated hepatotoxicity was through Nrf2/ARE pathway. The study was conducted in cells and mice, comparing the actions of Rg1. Rg1 significantly improved cell survival rates and promoted the expression of antioxidant proteins. Meanwhile, Rg1 reduced the excessive ROS and the occurrence of cell apoptosis, which were related to Nrf2/ARE pathway. Expression of Nrf2 has a certain cell specificity.

目的:探讨电视胸腔镜(VATS)解剖性肺段切除术与肺叶切除术治疗Ia 期非小细胞肺癌(NSCLC)患者的手术情况及对患者肺功能的影响。方法:选取我院手术治疗的Ⅰa期NSCLC患者,收集时间2014年1月至2016年12月,根据术式不同分为两组,均采用VATS手术治疗,A组(54例)患者采用解剖性肺段切除术、B组(60例)采用肺叶切除术治疗,对比两组患者的手术效果及术后肺功能变化。结果:A组患者的手术时间、清扫淋巴结数目与B组比较差异无统计学意义(P>0.05);A组患者的手术出血量、术后胸腔引流量、术后拔管时间、术后住院时间均显著的低于B组患者(P<0.05);术前,A组和B组患者的FEV1%、FVC%、MVV%测定值差异无统计学意义(P>0.05),术后3个月复查,A组患者的FEV1%、FVC%、MVV%测定值均显著高于B组患者(P<0.05);手术后,A组患者的并发症发生率(7.41%)低于B组患者(13.33%),但是差异无统计学意义(P>0.05)。结论:VATS解剖性肺段切除术治疗Ⅰa期NSCLC患者具有手术创伤小、术后恢复快、对患者肺功能影响更小的优势。  相似文献

目的:观察非霍奇金淋巴瘤(non-Hodgkin's lymphoma,NHL)患者自体造血干细胞移植(autologous hematopoietic stem cell transplantation,ASCT)术后应用重组人α-2b干扰素(α-2b IFN)进行早期干预治疗的临床疗效。方法:选取18例行ASCT的NHL患者为研究对象,移植前疾病评估均未达到完全缓解(complete remission,CR),试验组血象恢复后给予IFN 3 000 000 U次/隔日干预治疗,3个月后停用;对照组未行干扰素干预治疗,分析总体疗效及两组对比的生存情况。结果:随访中位时间为34(10~50)个月,患者中位生存时间为37(31~45)个月,3年总体无进展生存(progressive free survival,PFS)、总生存(overall survivial,OS)分别为54.7%、66.8%。ASCT后试验组1年内无疾病复发,2年内复发率为12.5%;对照组1年内复发率为20%,2年内复发率为30%。结论:NHL患者在ASCT后给予重组人α-2b IFN早期干预治疗,患者耐受性好,可能降低移植后早期复发率。  相似文献

BackgroundDual antiplatelet therapy (DAPT) is the cornerstone treatment of acute myocardial infarction (AMI).ObjectiveThe present study aimed to investigate the efficacy and safety of triple antiplatelet therapy (TAPT) in elderly female patients with diabetes and ST segment elevation myocardial infarction (STEMI), who had undergone percutaneous coronary intervention (PCI).MethodsWe designed a randomized, single-blind study. Control group A (97 elderly male patients with diabetes and STEMI, whose CRUSADE scores were < 30) received aspirin, ticagrelor, and tirofiban. A total of 162 elderly female patients with diabetes and STEMI were randomly divided into two groups according to CRUSADE score. Group B (69 patients with CRUSADE score > 31) received aspirin and ticagrelor. Group C (93 patients with CRUSADE score < 30) received aspirin, ticagrelor and tirofiban. P values < 0.05 were considered statistically significant.ResultsCompared to the findings in group A, post-PCI Thrombolysis in Myocardial Infarction (TIMI) grade 3 blood flow and TIMI myocardial perfusion grade 3 were significantly less prevalent in group B (p < 0.05). When compared to groups A and C, the incidence of major adverse complications was significantly higher in group B (p < 0.05).ConclusionTAPT could effectively reduce the incidence of major complications in elderly female patients with diabetes and STEMI. This study investigated the hypocholesterolaemic effects of bitter melon aqueous extracts (BMAE) in vitro, the inhibitory effects of BMAE on pancreatic cholesterol esterase (CEase) and incorporation of cholesterol into micelles were investigated. BMAE decreased the in vitro micellar solubility of cholesterol in a dose-dependent manner. The conformation of CEase was investigated by means of circular dichroism (CD) and fluorescence. The result revealed the decrease of α-helix contents, increase of β-sheet and exposure of aromatic amino acid residuals. The incorporation of cholesterol into micelles was inhibited by BMAE. A complex was observed by transmission electron microscopy (TEM), which indicated interaction between cholesterol and BMAE. The result revealed that BMAE can play a role in decreased intestinal cholesterol absorption via inhibition of CEase, and of micelle formation.  相似文献
