The action of mefenamic acid, a nonsteroidal anti-inflammatory drug, on energy metabolism in the isolated perfused rat liver was investigated. Mefenamic acid in the range between 0.1 and 1.0 mM was infused to livers from well-fed rats and from 24-hr fasted rats. The former were perfused with substrate-free Krebs/Henseleit-bicarbonate buffer, allowing the measurement of glycogenolysis and glycolysis from endogenous glycogen. The livers from 24-hr fasted rats, on the other hand, were perfused with Krebs/Henseleit-bicarbonate buffer containing fructose, thus allowing the measurement of fructolysis and glucose synthesis. Oxygen consumption was measured in both cases. When present in the range between 0.1 and 0.5 mM, mefenamic acid increased glycolysis, oxygen uptake, glycogenolysis and fructolysis. Higher concentrations, depending on the perfusion conditions, were inhibitory. Glucose production from exogenous fructose, on the other hand, was inhibited at low mefenamic acid concentrations. In general terms, the effects of mefenamic acid on energy metabolism seemed to be the primary consequence of its uncoupling action on the respiratory chain. This conclusion is supported mainly by the opposite effects on glucose synthesis (inhibition) and oxygen consumption (activation). The intracellular concentration of mefenamic acid is much higher than the extracellular one, a phenomenon which may represent binding to intracellular membrane or proteins.  相似文献

This study explored the predictors and consequences of sexual assault occurring after the age of 16 years in a nonclinical sample of women. Child sexual abuse occurring before the age of 16 years was the only predictor of later sexual assault among comorbid risk factors. Peer sexual abuse, number of perpetrators, age at time of sexual abuse, and severity of sexual abuse did not increase the risk for later sexual assault. Adult sexual assault victims showed lower levels of mental health functioning than did survivors of child or peer sexual abuse. We discuss a specificity model of revictimization and the differential effects of child, peer, and adult sexual trauma on the developmental trajectory of sexual violence and psychosocial functioning.  相似文献

OBJECTIVE: Azadirachta indica, a Meliaceae family tree, has been used in India for many years in the treatment of several diseases in medicine and dentistry. Current research analyses the effects of the leaf aqueous extract from Azadirachta indica (Neem) on the adhesion, cell surface hydrophobicity and biofilm formation, which may affect the colonisation by Candida albicans. METHODS: Azadirachta indica extract was tested in vitro on strains of Candida albicans 12A and 156B. Changes in hydrophobicity were reported in assays of yeast adhesion to hydrocarbons, in biofilm formation with glucose and in the adhesion of the microorganisms on light cured composite resin. Assays involved enumeration of candidal colony-forming units together with scintillation counting of radiolabelled Candida and compared to a solution of chlorhexidine digluconate 0.125% widely used in dentistry. RESULTS: Yeast growth in Neem extract was not inhibited in concentrations ranging from 0.1mg/ml. A statistically significant increase (p<0.05) in cell surface hydrophobicity was evident for the two strain tested and there was also an associated increase in biofilm formation after contact with Neem extract in concentration 0.01 g/ml. Decrease in adhesion capacity of cells to composite resin was also recorded. CONCLUSION: An anti-adhesive mechanism of action by Azadirachta indica is proposed based on the results observed.  相似文献

In vitro trials were conducted to evaluate the effect of Azadirachtaindica (neem) extracts on mycelial growth, sporulation, morphology and ochratoxin A production by P. verrucosum and P. brevicompactum. The effect of neem oil extract from seeds and leaf was evaluated at 0.125; 0.25 and 0.5% and 6.25 and 12.5 mg/mL, respectively, in Yeast Extract Sucrose (YES) medium. Ochratoxin A production was evaluated by a thin-layer chromatography technique. Oil extracts exhibited significant (p ≤ 0.05) reduction of growth and sporulation of the fungi. No inhibition of ochratoxin A production was observed. Given its accessibility and low cost, neem oil could be implemented as part of a sustainable integrated pest management strategy for plant disease, as it has been shown to be fungitoxic by inhibition of growth and sporulation.  相似文献

Efficiency of solid and liquid inocula and their use for spawn production were compared so that improved cultivation conditions for the edible mushroom Pleurotus ostreatoroseus could be tested. Solid and liquid inocula were prepared respectively with Potato Dextrose Agar (PDA) and Liquid Potato Dextrose (LPD). Wheat grains and cotton residues were used as substrates for spawn preparation. Inoculum types did not affect the development of P. ostreatoroseus, and LPD spawns were cheaper, more homogenous, less contaminated. Decomposition activity of mushroom growth, as a percentage of organic matter loss (OML), was higher in the wheat grain spawn and was not influenced by the inoculum type. Advantages in the use of cotton residue for spawn production were longer storage time, lower contamination and reduced costs. The cotton residue substrate may be also used for the production of mushroom fruiting bodies.  相似文献

The species of the genus Pleurotus produce large amounts of biomass and exopolysaccharide (EPS) in submerged cultures, which may be used for biotechnological purposes. In the present work two Brazilian autochthonous strains of edible Pleurotus (P. ostratoroseus Sing. and P. ostreatus "florida") were used. The fungi grown in liquid Potato Dextrose medium (PD) were used as inocula to cultivate those microorganisms in the POL culture medium. After a 9-day incubation, the optimal growth time for biomass production, P. ostreatus "florida" presented higher biomass production (22.8 g d.w./l) than P. ostreatoroseus (16.8 g d.w./l). After a 7-day incubation, the optimal growth time for EPS production, P. ostreatoroseus produced higher amounts of crude EPS (5.8 g d.w./l) than P. ostreatus "florida" (1.4 g d.w./l). Relative carbohydrate composition for P. ostreatoroseus and P. ostreatus "florida" EPS were: glucose (95.5-87.7), galactose (traces - 4.9), mannose (traces - 3.1), xylose (1.3-2.5), and arabinose (3.2-1.8). Lower ammonium sulfate concentration in the POL culture medium increased the exopolysaccharides production by P. ostreatoroseus.  相似文献

Aims/Background: In the sinusoidal bed the distribution of water is flow‐limited, but it becomes partly barrier‐limited when adenosine triphosphate (ATP) is introduced. This effect could be exerted either directly by ATP or by substances released from presinusoidal regions. Furthermore, portally infused ATP seems to be able to diffuse in the direction of the arterial bed. It is not known if this diffusion route is specific. Answers to these questions can be obtained from indicator‐dilution experiments in retrograde perfusion. Methods: Indicator‐dilution experiments, using [14C]sucrose and [3H]water, were conducted. Rat livers were perfused in the retrograde mode (hepatic vein+hepatic artery→portal vein). Results: When ATP was infused into the hepatic vein, the distribution of [3H]water remained essentially flow‐limited. The infusion of ATP into the hepatic artery increased the sucrose and extra‐sucrose spaces of the arterial bed, but infusion into the hepatic vein was without effect. Conclusions: The results indicate that the induction of barrier‐limited distribution of [3H]water is not a direct effect of ATP. Furthermore, if the transhepatic diffusion of ATP can occur from presinusoidal regions to the arterial bed, as shown by previous work, a similar diffusion does not occur from postsinusoidal regions.  相似文献

Fifty-two isolates of Fusarium species and one of Gibberella fujikuroi were tested for galactose oxidase (GO) production. Five Fusarium isolates contained GO activity in the culture filtrate: three F. graminearum and one each F. moniliforme f. sp. subglutinans and F. acuminatum. This is the first time F. acuminatum is reported to be a producer of GO enzyme. GO enzyme activity produced by isolates was assayed through a time course. Moreover, GO protein was partially purified from the most productive four isolates to show that the activity measured in the culture filtrates was due to the presence of GO protein.  相似文献
