Liver transplantation, which serves as treatment of familial amyloidotic polyneuropathy (FAP), and domino liver transplantation, which utilizes resected livers from patients with FAP for treatment of liver diseases, may induce changes in transthyretin (TTR), a pathogenic FAP-related protein. To evaluate this possibility, we performed a 70% hepatectomy or administered tacrolimus to Dark Agouti (DA) rats for 7 days and then measured changes in liver TTR mRNA levels and changes in serum TTR concentrations. After hepatectomy, TTR mRNA levels decreased by 77%; at day 3, they returned to preoperative levels. Except for slightly elevated serum TTR concentrations 12 h after operation, serum TTR levels remained unchanged. Thus, partial hepatectomy did not influence serum TTR concentrations. After tacrolimus administration, TTR mRNA declined by 56% 12 h after the experiment started; however, after day 3, a rebound phenomenon occurred until day 7. Tacrolimus may facilitate serum TTR degradation, although production of TTR in the liver also increased. This finding -- that TTR, the source of FAP-inducing amyloid, did not increase after transplantation -- may help post-transplantation treatment of patients who have FAP and other liver diseases.  相似文献

In this review article, we demonstrate the mucin expression profile in normal tissue, invasive ductal carcinoma (IDC), two subtypes of intraductal papillary–mucinous neoplasm (IPMN dark cell type and IPMN clear cell type), pancreatic intraepithelial neoplasia (PanIN), and mucinous cystic neoplasm (MCN) of the pancreas. In MUC1, there are various glycoforms, such as poorly glycosylated MUC1, sialylated MUC1, and fully glycosylated MUC1. IDCs showed high expression of all the glycoforms of MUC1. IPMNs dark cell type showed no expression or low expression of all the glycoforms of MUC1. IPMNs clear cell type showed low expression of poorly glycosylated MUC1, but expression of sialylated MUC1 and fully glycosylated MUC1. Expression of MUC2 was negative in IDCs, high in IPMNs dark cell type and low in IPMNs clear cell type. MUC5AC was highly expressed in IDCs, IPMNs dark cell type, and IPMNs clear cell type. MUC6 expression was higher in IPMNs clear cell type than in IDCs and IPMNs dark cell type. Our recent study demonstrated that high expression of MUC4 in IDCs is correlated with a poor outcome for patients. In PanINs, expression of both MUC5AC and MUC6 are an early event, whereas up-regulation of MUC1 is a late event. MCNs do not look as if they will show a specific mucin expression profile according to the literature review.  相似文献

A series of the copper(II) complexes of the type CuL4X2 (L: ligand, X: anion) was prepared with the following substituted imidazoles as ligands 2-ethyl- ( 1a ), 2-ethyl-4-methyl- ( 1b ), 2-phenyl- ( 1c ), 2-undecyl- ( 1d ), and 2-heptadecylimidazole ( 1e ). The structures of these complexes were confirmed by elemental analysis, reflection spectra of the crystals as well as IR-spectroscopy. Polymerization of acrylonitrile was tested in the presence of each imidazole-copper(II) complex in dimethyl sulfoxide solution.  相似文献

Recently nutrition support team (NST) has been established for the purpose of prevention of complications which are caused by nutrition disorders and reduction of the medical expenses. Although physical examinations and blood biochemical data had been used as the indexes evaluating nutritional of patients, they were not suitable for the evaluation for the short-term in-patient. On the contrary, serum albumin (ALB) has been wildly used as a nutritional marker. However, it is impossible to evaluate nutrition state for the short-term in-patient and acute phase disease patient accurately, because the plasma half-life is 21 days and it takes long time to detect the change in nutritional state by its value. Rapid turnover proteins (RTP), whose plasma half-life is shorter, has paid attention to evaluate nutritional state for the short-term in-patients and acute phase disease patients. Although, prognostic inflammatory and nutritional index (PINI) was considered as a useful maker for evaluating inflammatory and nutritional states using the concentrations of transthyretin (TTR), a RTP, alpha1-acid glycoprotein (alpha1-AG), a chronic inflammation marker, C reactive protein (CRP), a acute inflammation marker, and ALB, However, it has several pitfalls. We newly made serum amyloid A (SAA) index using SAA instead of CRP. When we compared SAA index with PINI in many diseases, it turned out that SAA index became a more effective index which reflected the patient condition than did PINI. As for this index, it is expected to be used by NST while further alternation may be needed.  相似文献

The initiation mechanism of vinyl polymerization by a copper(II) chelate of poly(vinyl alcohol) was studied in aqueous solution in the presence of carbon tetrachloride, in order to determine the free radical species formed during the initiation step. Trichloromethyl radicals were ascertained to be the main initiation species, by spin trapping and gelation experiments. Differences in polymerization activity between methyl methacrylate and acrylonitrile are discussed.  相似文献

In order to clarify the mechanism of catalysis of polyvinylamine-copper(II) chelates on the initiation of vinyl polymerizations, stability constants of these chelates were measured by applying a modification of BJERRUM'S method, and each concentration of the chemical species was estimated on the basis of the chelate theory. From these results, the activity of the polyvinylamine-copper(II) chelates for the polymerization of acrylonitrile and methyl methacrylate at different pH values was discussed.  相似文献

The polymerization of acrylonitrile (AN) and of methyl methacrylate (MMA) initiated by various copper(II) chelates was studied in the presence of carbon tetrachloride. Several vinylamine (VAm)/vinyl alcohol (VA) copolymers with different contents of NH2 and OH bearing monomeric units were chosen as polymer ligands as well as polyvinylamine (PVAm)/poly(vinyl alcohol) (PVA) mixtures (of equal NH2 and OH group contents as the corresponding copolymers), PVAm, and PVA. The activity of the used polymer chelates to initiate the AN and the MMA polymerization was compared at various pH values of the reaction systems. For the polymerization of MMA, the VAm/VA copolymer systems showed a higher activity than the PVAm chelate containing the same amount of NH2 groups, contrary to the case of the polymerization of AN, where the polymerization activity showed a reverse behavior. Chelate formation of vinylamine/vinyl alcohol copolymers with copper(II) was studied both by the spectrophotometric and by titrimetric methods. For comparison, polyvinylamine/poly(vinyl alcohol) mixtures were also taken up for measurements. In the case of the copolymer systems, copper(II) ions were found to be coordinated exclusively by nitrogen atoms of the copolymer ligand. The complexation constants of the chelates were determined by applying the modified Bjerrum method, in neglect of the acid dissociation of hydroxy groups in the copolymers. It was found that the copolymer/Cu(II) chelates tend to be unstable with decreasing contents of NH2 groups in the copolymers. In the homopolymer mixture systems, copper(II) ions are not coordinated by the oxygen atoms of poly(vinyl alcohol), but only by the nitrogen atoms of polyvinylamine.  相似文献

In this symposium, ways in which medical technologists can contribute as supporting members in Japan's health care system will be discussed. Improvement in the job specifications of health care professionals has been identified as a major pillar of Japanese health care reform. As members of the medical support team, along with university professors and medical laboratory researchers, medical technologists will be expected to improve their knowledge and skill. Medical technologists should therefore be prepared to make great efforts to progress according to expectations, and these things must be kept in mind as a future guideline for medical technologists in Japan.  相似文献

Four well-trained combination skiers were studied through pre- and post-training for the effects of short-term intermittent training during hypoxia on muscle energetics during submaximal exercise as measured by Phosphorus-31 nuclear magnetic resonance and maximal aerobic power ( O2max). The hypoxia and training in the cold was conducted in a hypobaric chamber and comprised 60-min aerobic exercise (at an intensity equivalent to the blood lactate threshold), using a cycle ergometer or a treadmill twice a day for 4, consecutive days at 5°C, in conditions equivalent to an altitude of 2000 m (593 mm Hg). No change in O2max was observed over the training period, while in the muscle energetics during submaximal exercise, the values of phosphocreatine/(phosphocreatine + inorganic phosphate) and intracellular pH were found to be significantly increased by training during hypoxia. During recovery, the time constant of phosphocreatine was found to have been significantly reduced [pre, 27.9 (SD 6.7) s; post, 22.5 (SD 4.7) s, P < 0.01]. The observed inhibition of phosphocreatine as well as that of intracellular pH changes after training during hypoxia and quicker recovery of phosphocreatine in submaximal exercise tests, may indicate improved oxidative capacity (i.e. a high adenosine 5-triphosphate formation rate) despite the short-term hypoxia training.
