将HFRS病人Ab_1免疫BALB/c小鼠,取脾细胞与Sp2/0-Ag14小鼠骨髓瘤细胞融合,制备了HFRSV的McAb_2。初期融合试验中用ELISA间接法检测阳性克隆,阳性率为3.2~7.4%。其中5株杂交瘤经克隆化后阳性率达到100%。特异性鉴定表明,5种McAb_2只与HFRS病人Ab_1结合,不与正常人Ig结合。McAb_2所识别的独特型是人Ab_1与部分小鼠抗HFRSV的McAb上的交叉反应独特型。本文还应用McAb_2作为探针,对抗HFRSV的单克隆抗体的独特型进行了分析。 相似文献

本研究用陕西81-14A株HFRSV,免疫BALB/c小鼠,取该鼠牌细胞与SP2/0骨髓瘤细胞融合,获得分泌抗HFRSV抗体的C_4株杂交瘤细胞系。C_4株McAb与我国各地的28株及朝鲜的76~118株,Seoul株HFRSV均产生免疫反应,表明是HFRSV组特异性抗体,并适用于荧光素及辣根过氧化物酶标记、经初步应用效果满意。 相似文献

用McAb亲和层析法粗提HFRS病毒81-14A株感染BHK细胞培养上清液,效果较满意。病毒抗原获得率为48.07%。经McAb亲和层析法洗脱,病毒抗原,蛋白峰基本一致。特异性试验证明洗脱物为HFRS病毒抗原。SDS-RAGE经氨银染色主要有4条区带。 相似文献

Thirty-five strains of HFRS virus isolated from the patients andseveral host animals in various areas in China were analyzed by IFATwith 10 McAbs specific for HFRS virus.It has been proved that thereare antigenic differences among the strains.The HFRS virus strainsrevealed nine different reactions for the McAb,showing that there are atleast nine different antigenic determinants,including group-,type-andstrain-specific.The analytical result shows that antigenic differencesamong the HFRS virus strains are mainly related to difference in the hostanimals. 相似文献

应用常规方法建立了5株稳定分泌抗重组人α2a 型干扰素单克隆抗体的小鼠杂交瘤细胞系1B8、2B6、3F1、4G5和5G10,并对其免疫学特性进行了较系统的鉴定.5株单抗均为 IgG1,特异性强,与重组人α1 型、γ型干扰素以及受体菌菌体蛋白成分无交叉反应.间接 ELISA 测定小鼠腹水单抗效价1:320,000~1:10,240,000,其中3F1和5G10单抗对重组人α2a 型干扰素具中和活性(中和效价均为1:10,000)。竞争结合 ELISA 证实5株单抗针对3种不同表位。用3F1和5G10单抗建立的夹心 ELISA 能检测到60pg/ml的重组人α2a 和α2b 型干扰素。3F1单抗用于制备亲和层析柱纯化重组人α2a 型干扰素结果与进口单抗NK2相当。 相似文献

本文用HFRS病毒McAbs免疫家兔,制备了抗Id抗体(Ab_2),用小鼠γ球蛋白-Sepharose4B免疫亲和层析柱纯化了Ab_2,并以ELISA试验对其特异性作了鉴定。结果表明,所制备的Ab_2具有良好的Id特异性,并能与所用的小鼠抗HFRS病毒单抗、兔抗HFRS病毒抗体和HFRS病人血清结合。同时提示,这种Ab_2可能具有替代抗原的免疫潜能。 相似文献

Indirect fluorescent antibody test (IFAT) was employed to detectviral antigens in the peripheral blood lymphocytes from 77 patients withclinically diagnosed epidemic hemorrhagic fever (EHF). The positive ratewas 50.7%, and it was 64.3% (27/42) for early cases (2nd to 5th day ofdisease). It was found that the earlier and the more severe the cases were,the higher the positive rate was. Three McAbs, 4B9, 4E7 and 3H4 havebeen introduced to IFAT. Five reaction patterns of the three McAbs withviral antigen positive cases were observed. The McAbs showed theirdifferences in respect of positive rate. Combination of McAbs could raisethe positive rate. In the cases of early phase with positive EHFV antigensin the leucocytes, the positive rate of specific IgM in acute phase sera was96.3%. The positive rate and accuracy of EHF early diagnosis could beraised by combined application of detection of the viral antigens andspecific serum IgM in the acute phase. 相似文献
