Differential effects of long-term treatment with clozapine or haloperidol on GABAA receptor binding and GAD67 expression

One of the most consistent findings in postmortem studies of schizophrenia is increased GABAA receptor binding and reduced glutamic acid decarboxylase (GAD67) expression. Due to long-term antipsychotic treatment before death, these findings may reflect not only the consequences of schizophrenia but also medication effects. To differentiate between these options, we used an animal model and evaluated long-term effects of typical (haloperidol) and atypical (clozapine) antipsychotic drugs on the GABAergic system. A total of 33 adult male rats were treated in three cohorts over a period of 6 months. One cohort of 11 animals received clozapine (45 mg/kg/day), another one received haloperidol (1.5 mg/kg/day) and a third one received pH-adapted minimal concentrations of HCl in the drinking water. Receptor autoradiography of the GABAA receptor ([3H]-muscimol binding) and in situ hybridization in adjacent sections with 35S-labeled cRNA probes of the y-aminobutyric acid (GABA)-producing enzyme, GAD67, was performed. While haloperidol increased GABAA receptor binding in striatum and nucleus accumbens (NA), it suppressed GABAA receptor binding in temporal (TEMPC) and parietal (PARC) cortex. Clozapine induced GABAA receptor binding in infralimbic cortex (ILC) and similar like haloperidol in anterior cingulate cortex (ACC), two regions of the limbic cortex. In addition, either drug increased gene expression of GAD67. It is concluded that antipsychotic drugs differentially alter the GABAergic system, strongly suggesting that drug effects are partially responsible for the up-regulation of GABAA receptor binding in certain brain regions as observed in postmortem brains of schizophrenic patients. However, the reduced GAD67 expression seen in postmortem brains does not appear to reflect drug effects, since our animal model demonstrated increased gene expression.     

Subcortical and medial temporal MR-detectable metabolite abnormalities in unipolar major depression

The purpose of the present study was to determine whether MR-detectable alterations of choline-containing compounds in two key neural systems involved in major depression disorder namely the hippocampus and the basal ganglia can be detected. Multislice proton magnetic resonance spectroscopic imaging was applied in 11 patients with major depressive disorder (MDD) and ten matched healthy subjects. Voxels were selected from the left and right side of the hippocampus and the putamen. Significantly lower choline-containing compounds in the hippocampus and significantly higher choline-containing compounds in the putamen of patients with MDD compared to healthy subjects were found. No significant differences were found for the other metabolites in the two regions evaluated. Abnormal levels of choline-containing compounds most likely reflect altered membrane phospholipid metabolism. A reduced level in the hippocampus and an increased level in the putamen suggest regionally opponent membrane abnormalities. Abnormal amygdala activation profile in pedophilia

Despite considerable public interest research in neurobiological correlates of pedophilia is scarce. Since amygdala activation is central for emotional valuation, arousal, and salience, we investigated the activation profile of this structure in 10 male subjects with pedophilia (exclusively attracted to boys), all convicted sex-offenders and sentenced to forensic psychiatric treatment along with ten male heterosexual matched controls. We used a sexually non-explicit functional Magnetic Resonance Imaging (fMRI) paradigm with images of men, women, boys or girls randomly embedded in neutral target/non-target geometrical symbols. We applied statistical parametric mapping (SPM2) and SPSS 14 for image processing and analysis. While controls activated significantly less to pictures of children compared to adults, the activation profile was reversed in subjects with pedophilia, who exhibited significantly more activation to children than adults. The highest activation was observed for boys in the patient group, and for women in control participants. Our data show enhanced activation to children's pictures even in an incidental context and suggest the provocative hypothesis that a normally present mechanism for reduced emotional arousal for children relative to adults is reversed in pedophilia, suggesting a neural substrate associated with deviant sexual preference in this condition. More extensive research in this field would be of benefit for both the victims and the offenders.     

Antisperm antibodies and microorganisms in genital secretions—a clinically significant relationship?

Summary. In asymptomatic infertility patients, no significant relationship was found between the presence of antisperm antibodies (ASA) in serum and in semen samples (IgG and/or IgA ASA), differentiated with the mixed antiglobulin reaction (MAR), and the microbial colonization of ejaculates covering a broad spectrum of microorganisms. Likewise, there was no significant association of ASA with microbial findings in patients' female partners, who also presented without symptoms of genital tract infection and were screened at the same time. Furthermore, ASA in semen (IgG and IgA) were not significantly related to several potential markers of subclinical male sexual gland infection or inflammation (leukocytes, PMN elastase, albumin, C_3c) evaluated in aliquots of the same ejaculates used for immunological testing.     

Clinical and cytogenetic observations during a six-year period in an adult with Fanconi's anaemia

Summary A male adult patient suffering from Fanconi's anemia is described who was diagnosed 5 years before the onset of clinical symptoms by cytogenetic findings of chromosomeinstability in a lymphocyte culture. Repeated clinical, haematological and biochemical investigations of the untreated patient have been made during the observation period of six years. In the same period of time cytogenetic studies have been carried out which show no correlation in results compared with the clinical or physical findings. Four well defined lymphocyte clones have been discovered. The patient is still under observation of the clinic and the cytogenetic department.This work was supported by the Deutsche Forschungsgemeinschaft.     

Über die Abhängigkeit des Gesundheitszustandes von plötzlichen Eruptionen auf der Sonne und die Existenz einer 27tägigen Periode in den Sterbefällen

Zusammenfassung Unter Zuhilfenahme der neuesten Forschungsergebnisse der Geo- und Astrophysik gelingt in vorliegender Arbeit der Nachweis, daß eine ganze Reihe von Störungen im normalen Ablauf der Lebensfunktionen, deren Auftreten man bisher gewissen Umschichtungen in der Atmosphäre zuschrieb, diesen nicht untergeordnet, sondern im wesentlichenbeigeordnet ist. Als gemeinsame Ursache für die Umwälzungen in der Troposphäre und die Auslösung der sog. Wetterfühligkeit, ferner vieler Krankheitsausbrüche und Todesfälle werdenheftige, kurzandauernde Eruptionen ermittelt, die von Zeit zu Zeit auf bestimmten Teilen der Sonnenoberfläche (den sog.M-Regionen) stattfinden. Die bei diesen solaren Vorgängen ausgestoßenen elektrischen Teilchen erreichen unter bestimmten Bedingungen die Erde und rufen dort sog.Elektroinvasionen hervor, deren Umfang am besten durch die Stärke der erdmagnetischen Feldstörungen charakterisiert wird. Mit Hilfe von zwei neuartigen, ausführlich beschriebenen Auswertungsmethoden, die jeden einzelnen Tag des bearbeiteten Zeitraumes von 5 Jahren erfassen, wird ein Material von rund36 000 Todesfällen in Kopenhagen und 4000 Todesfällen in Zürich individuell bearbeitet und zum ersten Male der Beweis erbracht, daß eine deutlich ausgeprägte27tägige Periode in der Häufigkeit der Sterbefälle vorhanden ist. Aus den gegebenen graphischen Darstellungen läßt sich ablesen, wieviel Tage im Mittel vergehen, bis die Anzahl der Todesfälle nach dieser oder jener Krankheitsgruppe auf einen Anstieg der solaren Tätigkeit reagiert. An Hand einiger Tabellen wird ferner gezeigt, daß die Schwankungen der Sterbeziffern auch der Größenordnung nach in einem sehr annehmbaren Verhältnis zu den entsprechenden Schwankungen der übergeordneten kosmischen Phänomene stehen. Eine geradezu erstaunliche Übereinstimmung zeigen Kurven, in denen die Sterbefälle bestimmter Krankheitsgruppen von zwei rund 1000 km voneinander entfernten Großstädten (Kopenhagen und Zürich) dargestellt sind. Um zu beweisen, daß die solaren Eruptionen, irdischen Elektroinvasionen und gewisse, mit ihnen verbundene andere Vorgänge nur denZeitpunkt für die Auslösung der Todesfälle bestimmen, nicht aber die Anzahl der ausgelösten Fälle, wird ein besonderes Kapitel denbereitschaftserhöhenden Faktoren gewidmet und an Hand mehrerer graphischer Darstellungen auf deren besondere Bedeutung hingewiesen. Gegen Ende der Arbeit wird auf das immer noch ziemlich rätselhafteAgens eingegangen, das die Wirkungen der solaren Eruptionen auf den Menschen vermittelt. Nach Ausschluß rein meteorologischer und luftelektrischer Elemente wird festgestellt, daß nach dem derzeitigen Stande der Wissenschaftdrei Strahlenarten in Frage kommen, von denen eine jede für sich imstande wäre, die beobachteten Veränderungen im menschlichen Organismus hervorzurufen. Praktisch dürfte allerdings das Zusammenwirken dieser Strahlungsgattungen maßgebend sein, für derenEntstehung in der freien Natur und biologische Wirksamkeit eine große Anzahl von Tatsachen angeführt wird. Zum Schluß wird ein kurzer Hinweis auf diepraktische Nutzanwendung der bisherigen Erkenntnisse gegeben.Mit 12 Abbildungen im Text.

Zusammenfassung Abschließend läßt sich sagen, daß Tumormetastasen im Recessus opticus das Chiasma weitgehend zerstören können und so zu dem klinischen Symptom des Zentralskotoms führen. Wenn diese Einwucherung vor Einsetzen der allgemeinen Hirnschwellung erfolgt, kann es zum Bild der primären Opticusatrophie ohne Stauungspapille kommen.Ferner kann bei hochgradigem Hydrocephalus int. der Recessus opticus des 3. Ventrikels das Chiasma derartig komprimieren, daß es zur Druckschädigung in Form von Markscheidenzerfall kommt, so daß das zunächst befremdliche Bild eines Zentralskotoms bei Stauungspapille entsteht.
