Smooth muscle is important for the contractility and elasticity of visceral organs. The zebrafish is an excellent model for understanding embryonic development, yet due to a lack of appropriate markers, visceral smooth muscle development remains poorly characterized. Here, we develop markers and trace the development of gut and swim bladder smooth muscle in embryonic and juvenile fish. The first smooth muscle marker we detect in the vicinity of the gut is the myoblast marker nonmuscle myosin heavy chain-b at 50 hours postfertilization (hpf), followed by the early smooth muscle markers SM22alpha-b, and alpha-smooth muscle actin at 56 and 60 hpf, respectively. Markers of more differentiated smooth muscle, smoothelin-b and cpi-17, appear by 3 days postfertilization (dpf). Tropomyosin, a relatively late marker, is first expressed at 4 dpf. We find that smooth muscle marker expression in the swim bladder follows the same sequence of marker expression as the gut, but markers have a temporal delay reflecting the later formation of swim bladder smooth muscle.  相似文献

Cotton is one of the most important raw materials for textile and clothing production. The main drawbacks of cotton fibers are their poor mechanical properties and high flammability. Compared with some synthetic polymer fibers, cotton fabrics treated with modern flame-retardant and reinforcement finishes often cannot meet rigid military specifications. Polypyrrole–magnesium oxide (ppy–MgO) and polypyrrole–magnesium oxide–carbon nanotube (ppy–MgO–CNT) composites were prepared with various weight ratios by in situ chemical polymerization method. 1,2,3,4-Butane tetracarboxylic acid (BTCA) was used as a cross-linking agent in the presence of sodium hypophosphite (SHP). The composite sol was coated on cotton fabric using the pad-dry-cure technique. The coated cotton fabrics were characterized by SEM, EDAX, XRD, UV-DRS and FT-IR analysis, and tested for flame retardant and UPF application. The flame-retardant study showed a maximum char length of 0.3 cm and the char yield was about 49% for the ppy–MgO–CNT composite. For that UPF application, a 30 UPF value was shown for the ppy–MgO–CNT composite. In the case of the antibacterial study, the zone of inhibition was observed for all of the test samples against MRSA and PAO1 bacteria. The zone of inhibition showed as 4.0, 3.0 mm for the ppy–MgO–CNT composite. Hence, the ppy–MgO–CNT composite was found to be efficient.

The Syrian hamster has proved useful in the evaluation of therapeutics and vaccines for severe acute respiratory syndrome-coronavirus-2 (SARS-CoV-2). To advance the model for preclinical studies, we conducted serial sacrifice of lungs, large pulmonary vessels, and hearts from male and female Syrian hamsters for days 1–4, and 8 post-infection (dpi) following infection with a high dose of SARS-CoV-2. Evaluation of microscopic lung histopathology scores suggests 4 and 8 dpi as prime indicators in the evaluation of moderate pathology with bronchial hyperplasia, alveolar involvement and bronchiolization being key assessments of lung disease and recovery, respectively. In addition, neutrophil levels, red blood cell count and hematocrit showed significant increases during early infection. We present histological evidence of severe damage to the pulmonary vasculature with extensive leukocyte transmigration and the loss of endothelial cells and tunica media. Our evidence of endothelial and inflammatory cell death in the pulmonary vessels suggests endothelialitis secondary to SARS-CoV-2 epithelial cell infection as a possible determinant of the pathological findings along with the host inflammatory response. Lastly, pathological examination of the heart revealed evidence for intracardiac platelet/fibrin aggregates in male and female hamsters on 8 dpi, which might be indicative of a hypercoagulative state in these animals.  相似文献
