Adenophora polysaccharides (AP), is an active principle extracted from the root of Adenophorae Potaninii Korsh originated in Gansu Province and isolated with boiling water(1). AP is isolated and purified from the crude drug by DEAE-cellulose and Sephadex G-200 column, with a white powder and mean molecular weight of 8.3 × 104 , and [α]D20of AP is 68(1). AP is only composed of glucose judging from the analysis of it with patina chromatography (PC) and gas chromatography-mass spectrometer (GC-MS) methods.The methylation analysis showed that AP is composed of (1→6) linked glucose residues. The measure of nuclear magnetic resonance imaging (NMR) 1H NMR and 14C NMR techniques further proved that AP is α(l→6) linked by Dglucose. 目的:研究异黄酮genistein 对肿瘤转移的治疗作用。方法:B16-BL6 细胞由尾静脉注入C57BL/6 小鼠体内,于d 15 观察动物肺转移灶。genistein 混悬于2% 卵磷脂中,从接种瘤细胞前1 天开始ip,剂量为100 和200mg·kg-1·d-1 。结果:genistein 可抑制肺转移灶形成,200 mg·kg-1 用药组小鼠转移灶数目明显减少;作为对照,腹腔给予环磷酰胺100 mg·kg-1 后,形成的转移灶数目无明显变化,但形成的转移瘤灶体积减小。当genistein 和环磷酰胺合用时,转移小鼠的存活时间明显延长。结论:genistein 可抑制肿瘤转移,其作用与细胞毒药物不同, 与细胞毒药物合用可提高其抗肿瘤转移的作用。  相似文献

目的 探讨青年急性进展型脑梗死患者进展与治疗前后血中D-二聚体(D-D)、高敏 C-反应蛋白(hs-CRP)含量变化与脑梗死发展的关系.方法 选择急性脑梗死 A 组(未发生进展)30例、B组(发生进展)20例和正常对照组50例分别测定其hs-CRP、D-D进展及治疗前后含量并加以分析.结果 青年急性进展型脑梗死组D-D和 hs-CRP含量明显高于对照组和非进展型脑梗死组(P<0.01); D-D和 hs-CRP含量经统计学处理呈正相关(r=0.64,P<0.01);而且脑梗死在治疗和有效控制后D-D和 hs-CRP含量明显下降(P<0.01).结论 血中D-D和 hs-CRP与青年急性进展型脑梗死的发生、发展及预后有密切关系,可作为评估的指标之一,对于脑梗死的进展预测和干预处理有一定临床价值. Abstract: Objective To study the correlation between the levels of D-Dimer(D-D),high sensitive C-reactive protein(hs-CRP) in serum of the young patients with acute progressive cerebral infarction before and after the proceeding and treatment. Methods Levels of hs-CRP, D-D were detected respectively in serum of the 20 young patients with acute progressive cerebral infarction, the 30 young patients with acute non- progressive cerebral infarction and 50 healthy people sampled as control group and correlation were analysed before and after the proceeding and treatment. Results Compared with healthy person and young patients with non-progressive cerebral infarction, the levels of D-D and hs-CRP in serum of young patients with progressive cerebral infarction were significantly higher (P<0.01). The level of D-D was positively correlated with that of hs-CRP,The levels of D-D and hs-CRP in serum of patients with cerebral infarction were significantly lower than before after having been treated and intervented effectively. Conclusions The levels of D-D,hs-CRP are closely related to the existence and development of the young patients with cerebral infarction. The levels of D-D and hs-CRP can be used as one of the assessment indexes of the young patient's conditions and is helpful for clinic forecast and intervention of acute progressive cerebral infarction.  相似文献

纤粘连蛋白诱导卵巢癌细胞基质金属蛋白酶-2基因表达   总被引:9,自引:0,他引:9  
目的 研究细胞外基质蛋白对肿瘤细胞基质金属蛋白酶 (MMP)基因表达的影响及机理。方法 以纤粘连蛋白处理SKOV3卵巢癌细胞后 ,用明胶 酶谱法和反转录PCR观察MMP分泌及基因表达。以MMP 2启动子转染细胞 ,通过测定萤火虫酶活性 ,观察纤粘连蛋白对MMP 2启动子活性的影响。细胞p5 3含量采用Westernblot分析。结果  5 μg/mL纤粘连蛋白刺激MMP 2分泌 ,但不影响MMP 9分泌 ,这一作用随纤粘连蛋白浓度增加而增强。经 10 μg/mL纤粘连蛋白刺激 1h后 ,细胞内MMP 2mRNA量显著增加 ;作用时间延长 ,诱导作用减弱。纤粘连蛋白可诱导转染细胞中萤火虫酶活性增加 ,AP 1功能抑制剂姜黄素 (5 0 μmol/L)不能阻断这一作用。经纤粘连蛋白处理后 ,细胞内p5 3含量明显增加。结论 纤粘连蛋白可刺激卵巢癌细胞中MMP 2分泌 ,诱导MMP 2启动子活性增强及mRNA含量增加 ,这一作用可能与p5 3蛋白转录因子活性有关 ,而与AP 1通路无关。  相似文献

Effects of genistein on invasion and matrix metalloproteinase activities were investigated in HT1080 human sarcoma cells, lnvasion of HTI080 cells through reconstituted basement membnme was inhibited when the cells were treated with 100μmol/L and 200μmol/L genistein; At the same concentrations,genistein not only suppressed latent forms of matrix metalloprotinese-2 and-9 (MMP-2 mad MMP-9) to convert into active forms, but also increase dramatically the tissue inhibitor of metalloproteinase (TIMP-1 ) mRNA contents and reverse the imbalance of MMPs and TIMPs. However, expressions of MMP-2 and MMP-9 were not sigrdficantly affected. Suppression of MMP activation and increase of TMP-1 expression will decrease matrix degradation by MMPs, and consequently inhibit invasions of the cells. These results emphasized the existence of the imbalance between MMPs and TIMPs in tumor invasion mad metastasis formation, The value of genistein as a drug for antiinvasion and anti-metastasis chemotherapy was suggested.  相似文献

Trisomy 21, also named Down syndrome was the most frequent autosomal aneuploidy and the most common cause of mental retardation. Fifty percent patients had congenital heart malformation. Objective To study the therapeutic effect,safety and risk of fractional resurfacing with ablative laser in the treatment of superficial scar.Methods 88 cases of superficial scar,including 66 cases of acne scar,12 cases of burn scar and 10 cases of other scar,were treated.All the patients were treated with Pixel(Er:YAG 2940 nm,≥3 times),or Encore(Ultrapulse CO2 10600 nm,≥2 times),or a combination of Pixel and Encore(≥3 times).Results The therapeutic effect of treatment was graded as medium when the scar was improved >25%,as good when the scar wag improved >50%.The effective result Was achieved in 80% of the patients.Good effect was achieved in 50% of the patients.There were 244 treatments of Pixel and 86 treatments of Encore.Persistent hyperpigmentation was happened in one case with Encore treatment.which relieved four months later.No other complication was happened.Conclusions The fractional resurfacing with ablative laser is an effective method for surperficial scar,especially acne scar.It is mini-invasive with low risk.The key mechanism is the ability of enhancing the tissue remodeling induced by heat effect of laser.  相似文献
