ObjectiveTo asses the ability of 4 types of Mtb proteins-ESAT6, SCWP, MAN and Ag85 to serve as indicator for active tuberculosis among Malaysian population.MethodsSera from 90 individuals, 60 from confirmed pulmonary tuberculosis patients and 30 healthy PPD negative individuals were tested for presence of anti-IgG and anti IgA by ELISA assay.ResultMean concentration of IgG and IgA were higher in patients compared to healthy Positivity of the ELISA test were calculated, taking the cut off value at mean +2 SD of healthy sera. The sensitivity of the ELISA IgA assay for ESAT 6, SCWP, MAN and Ag85 were 81.1%, 83.3%, 11.7% and 53.3% respectively. The sensitivity of the ELISA IgG asay for ESAT 6, SCWP, MAN and Ag85 were 71.0%, 71.0%, 71.0% and 21.0% respectively.ConclusionDetectionof IgA against SCWP promised a good indicator for active tuberculosis infection among Malaysian. 相似文献

Recombinant Mycobacterium bovis bacille Calmette-Guèrin (rBCG) expressing the malarial epitopes F2R(II)EBA and (NANP)3 as well as two T cell epitopes of the M. tuberculosis ESAT-6 antigen, generated in favour of mycobacterium codon usage elicited specific immune response against these epitopes. Immunised Balb/c mice demonstrated an increase in almost all of the IgG subclasses against both malarial epitopes and enhanced splenocyte proliferative response against the malarial epitopes as well as selected peptides of ESAT-6. Furthermore, flow cytometric analyses showed elevated numbers of CD4+ lymphocytes expressing IFN-gamma and IL-2 against the ESAT-6 peptides, suggesting a specific Th1-mediated response. This study demonstrated that expressing malarial and TB epitopes in a single rBCG construct induced appropriate humoral and cellular immune response against immunogenic epitopes from both organisms. 相似文献

Botanical pentacyclic triterpenes possessed a broad range of pharmacological activities such as antioxidant, anti-tumor, anti-microbial and anti-inflammatory activities. It is believed that the mechanisms involved in these bioactivities are due to the modulation of immune system. Recently, the pharmacological validation on immunomodulatory of pentacyclic triterpenes derived from higher plants is very limited and several existence review papers related for this group of compound have not been focused for this activity. In this review, we have highlighted several studies on immunomodulatory potential of botanical pentacyclic triterpenes isolated from wide array of different species of medicinal plants and herbs based on various preclinical in vitro and animal models. This review also attempts to discuss on bioactivities of compouns related with their structure-activity relationship. Hence, the evaluation of pentacyclic triterpenes offers a great opportunity to discover adjuvants and novel therapeutic agents that presented beneficial immunomodulatory properties. 相似文献
