Involvement of ADAM10 in axonal outgrowth and myelination of the peripheral nerve

The disintegrin and metalloproteinase 10 (ADAM10) is a membrane‐anchored metalloproteinase with both proteolytic and disintegrin characteristics. Here, we investigate the expression, regulation, and functional role of ADAM10 in axonal outgrowth and myelination of the peripheral nerve. Expression pattern analysis of 11 ADAM family members in co‐cultures of rat dorsal root ganglia (DRG) neurons and Schwann cells (SCs) demonstrated the most pronounced mRNA expression for ADAM10. In further studies, ADAM10 was found to be consistently upregulated in DRG‐SC co‐cultures before the induction of myelination. Neurons as well as SCs widely expressed ADAM10 at the protein level. In neurons, the expression of ADAM10 was exclusively limited to the axons before the induction of myelination. Inhibition of ADAM10 activity by the hydroxamate‐based inhibitors GI254023X and GW280264X resulted in a significant decrease in the mean axonal length. These data suggest that ADAM10 represents a prerequisite for myelination, although its activity is not required during the process of myelination itself as demonstrated by expression analysis of myelin protein zero (P0) and Sudan black staining. Hence, during the process of myelin formation, ADAM10 is highly upregulated and appears to be critically involved in axonal outgrowth that is a requirement for myelination in the peripheral nerve. © 2009 Wiley‐Liss, Inc.     

石刻雕塑作业劳动卫生学调查及实验研究

测定了石雕作业粉尘浓度，几何平均浓度为８２．３５～３．６７ｍｇ／ｍ３。工人职业性健康检查结果，接尘工人的咽部充血、咳嗽、咯痰检出率高于对照组；肺功能低下的检出率也高于对照组。该厂自１９７２年以来发生尘肺患者１７例，其病情发展和晋期缓慢，发病工龄长。动物实验研究表明，石雕粉尘有轻微的致纤维化作用。     

女性压力性尿失禁的微创治疗

目的探讨女性压力性尿失禁的发病机制和微创治疗方法。方法女性压力性尿失禁患者25例,经临床、尿动力学检查或膀胱尿道造影确诊,均为稳定性膀胱,无膀胱出口梗阻。采用无张力阴道吊带术(TVT),将吊带无张力置于尿道中段。结果手术时间平均(40±5)min;术中出血量平均(43±10)mL;术后平均留置导尿1.5 d;术后平均住院3 d。23例患者术后尿失禁症状均消失,其中2例患者术后出现尿潴留,留置导尿1周后好转,尿潴留症状消失,尿失禁未复发;2例尿失禁症状显著改善。结论TVT等微创术式因简单、微创、疗效好、并发症少等,目前是女性压力性尿失禁外科手术治疗的发展方向。     

Reduced 293T cell susceptibility to acrolein due to aldose reductase-like-1 protein expression.

Acrolein is a highly reactive alpha,beta-unsaturated aldehyde produced endogenously during lipid peroxidation and naturally distributed pervasively in living environments, posing serious threats to human health if not properly metabolized. In this study, we report aldose reductase-like-1 (ARL-1) as a novel enzyme that catalyzes the reduction of acrolein and protects cells from their toxicity. Using purified ARL-1 protein, we determined its enzymatic activity in response to acrolein and defined its steady-state kinetics with K(m) and V(max) at 0.110 +/- 0.012 mM and 3122.0 +/- 64.7 nmol/mg protein/min, respectively. By introducing a functional Enhanced Green Fluorescent Protein (EGFP)/ARL-1 fusion protein into 293T cells, we demonstrated that plating efficiency in liquid culture and focus formation in soft agar increased by more than 60% (p < 0.05), compared to the vector control cells. More significantly, at a low dose of 5 microM acrolein, EGFP/ARL-1 expression enhanced both plating efficiency and focus formation by more than threefold, and the foci (in soft agar) of 293T cells expressing EGFP/ARL-1 were significantly larger than those of the vector control cells. At high concentrations of acrolein (25 and 50 microM), EGFP/ARL-1 protein prevented oncotic death of 293T cells induced by acrolein. In summary, our data demonstrated for the first time that the ARL-1 protein protects 293T cells from acrolein toxicity. Due to the high toxicity and wide distribution of acrolein, this finding is important to the understanding of its detoxification mechanisms.     

电视胸腔镜心血管外科手术150例

目的 总结、分析150例电视胸腔镜心血管外科手术的临床结果。方法 应用电视胸腔镜手术技术对动脉导管未闭、房间隔缺损、室间隔缺损、三尖辨关闭不全、冠状动脉-肺动脉瘘、心包积液、原发性长Q-T综合征、雷诺病，心包囊肿等进行治疗。结果 动脉导管未闭均成功在胸腔镜下钳闭；房缺、室缺在胸腔镜体外循环下进行补片修补，冠状动脉瘘胸腔镜下修补；心包积液进行心包活检与开窗引流；长Q-T综合征进行胸腔镜下左T2、T3胸交感神经节切除，均无严重并发症，术后患者恢复良好。结论 电视胸腔镜心血管外科手术可取得满意的临床治疗结果。     

99Tcm标记MIBI与MDP骨常规及延迟显像对比研究

目的探讨99Tcm-MIBI与99Tcm-MDP骨常规及其延迟显像对骨良恶性病变的诊断、鉴别诊断及疗效评估的临床应用价值及影响因素. 方法68例临床拟诊骨病变患者分别进行99Tcm-MDP和 99Tcm-MIBI骨显像;MIBI呈阳性者均行延迟显像;最终均行手术治疗及病理检查;其中12例于术前曾行化疗和/或放疗,治疗前后均行上述2项显像. 结果①99Tcm-MIBI显像76.3%(61/80病灶)恶性肿瘤病灶肉眼可见中、高度MIBI浓聚;而良性病灶仅有27.3%(6/22)的轻、中度浓聚;恶性病灶与对侧正常组织计数比值(L/C)为3.16±1.36明显高于良性病灶(1.35±0.56),P<0.01;延迟显像有利于良性病灶假阳性的鉴别.②99Tcm-MDP显像上述所有病灶均肉眼可见MDP浓聚,但恶性病灶L/C(3.66±1.27)与良性病灶L/C(2.97±1.03)比较差异无显著性(P>0.05).③化疗及放疗可以较明显抑制99Tcm-MIBI摄取(但放疗却可增加99Tcm-MDP摄取),因此99Tcm-MIBI摄取程度与99Tcm-MDP比较能较好反应治疗效果. 结论 99Tcm-MIBI骨常规及延迟显像对鉴别诊断良恶性骨病和评估疗效有较好的应用价值,若与99Tcm-MDP显像联合应用可提供更多有价值的信息.     

国内丙型肝炎病毒感染与肝细胞癌关系的19个病例对照研究的 Meta 分析

本文对国内１９个有关丙型肝炎病毒（ＨＣＶ）感染与肝细胞癌（ＨＣＣ）关系的病例对照研究进行Ｍｅｔａ分析。经齐性检验表明，１９个研究中有１７个研究结果是一致的。对此１７个研究结果加权合并，累积ＨＣＣ病例１５７７例，ＨＣＶ感染率为２３．０％，累积对照１９８０例，ＨＣＶ感染率为４．０％；合并ＯＲ值为６．７。结果表明，国内ＨＣＶ感染与ＨＣＣ关系密切，ＨＣＶ感染是ＨＣＣ发生的一个危险因素。     

妇女月经周期睡眠行为变化

目的 观察在月经周期中,妇女睡眠活动的变化。方法 本工作以问卷填表方式调查在月经周期中妇女睡眠行为的变化,应用多道睡眠脑电图观察在卵泡期和黄体期睡眠脑电波形的变化。结果 与黄体期比较,在卵泡期妇女多梦、记忆清楚,易醒、再入睡困难,白天精力体力差,自 感觉没睡好,总睡眠时间无显著差异,总睡眠分值低;与黄体期比较,卵泡期快波睡眠时间延长,慢波2期和4期睡眠时间缩短。结论 在月经周期中,妇女睡眠行为的变化系由体内雌激素浓度变化所致。
