人冠状病毒72-62和73-50干燥毒种,用人胚器官和原代人胚肾细胞合管培养未能引起细胞病变,而将病毒液接种到人胚器官细胞或人胚肺传代细胞第一代就能引起明显的细胞病变。新传代病毒经电镜及中和试验鉴定确属人冠状病毒。实验结果证明人胚肺传代细胞比原代人胚肾细胞敏感,为进一步研究人冠状病毒提供了一株敏感的细胞。  相似文献

目的:选择能与整合素α3受体结合的小分子多肽cNGQGEQc-L作为靶向载体,将羧基荧光素(FAM)与cNGQGEQc-L连接构建荧光分子探针,通过荧光成像探讨荧光多肽分子探针用于肺腺癌显像的可行性。方法:利用倒置荧光显微镜观察FAM-cNGQGEQc-L与肺腺癌A549细胞结合部位,流式细胞仪检测荧光多肽与A549细胞的竞争抑制实验,观察FAM-cNGQGEQc-L随浓度的增加与肺腺癌A549细胞结合能力的变化情况。通过小动物活体成像仪,观察荧光多肽在荷瘤裸鼠体内的生物分布特点。结果:倒置荧光显微镜显示荧光多肽cNGQGEQc-L能与A549细胞结合,结合部位在细胞膜和细胞质中。流式细胞仪测试结果证明荧光多肽与A549细胞的结合具有特异性和饱和性,当FAM-cNGQGEQc-L浓度为0.125 mmol/L时,荧光多肽与A549细胞的结合趋近饱和。荷瘤裸鼠活体成像显示移植瘤能够摄取荧光多肽,且荧光多肽通过泌尿系统和胆道系统排泄。结论:体外、体内荧光实验结果显示,荧光多肽分子探针FAM-cNGQGEQc-L可与肺腺癌A549细胞、肺癌移植瘤结合,能够特异性靶向肺腺癌。  相似文献

目的:系统评价缺血性脑卒中中医康复治疗指南的质量,为指南的制定规范提供借鉴。方法:计算机检索数据库:PubMed、国家知识基础设施数据库、中国学术期刊数据库、中文科技期刊数据库、中国生物医学文献数据库,检索指南相关网站:卫生保健规范研究所(NICE)、国际网指南(GIN)数据库,梅斯医学数据库,中华中医药学会标准化委员会网站(http://www.cacm.org.cn/category/zyzn/bzh/)、中国中西医结合学会网站(http://www.caim.org.cn/),检索时限为建库至2021年9月,收集缺血性脑卒中中医康复治疗指南。应用研发与评价指南评估Ⅱ(AGREE Ⅱ)工具对纳入指南进行质量评价,并汇总中等质量及以上的指南推荐意见。结果:共纳入13部指南,AGREE Ⅱ的平均得分分别为范围和目的65.67%、参与人员43.02%、严谨性43.64%、清晰性63.82%、应用性19.87%和独立性32.05%。13部指南中7部指南为中等质量,6部指南为低质量。本研究得到23条中医康复治疗的推荐意见,其中2条为强推荐。结论:指南总体质量不高,未来指南的制定应该从指南应用性、严谨性和独立性方面规范,同时重视开展高质量的临床研究,为指南的制定提供证据支持。  相似文献

目的 观察伤痛酊擦剂治疗急性闭合性软组织损伤的临床疗效。方法  2 0 0例急性闭合性软组织损伤患者随机分为 2组 ,治疗组 1 5 0例应用伤痛酊擦剂治疗 ,对照组 5 0例应用正红花油治疗。 2组均治疗 2周后观察疗效。结果 治疗组临床治愈率 49.3% ,总显效率 90 .0 % ,对照组分别为 2 8.0 %、5 2 .0 % ,2组比较均有非常显著性差异 (P <0 .0 1 )。结论 伤痛酊擦剂治疗急性闭合性软组织损伤疗效确切。  相似文献

Magnesium hydride is regarded as one of the most ideal candidates for hydrogen storage, but its relatively high operating temperatures and slow kinetics always hinder its commercial applications. Herein, we first fabricated hard-carbon-sphere-wrapped Ni (Ni/HCS) via a mild chemical method; subsequently, the as-prepared additive was introduced to fabricate an Mg–Ni/HCS composite by using hydriding combustion synthesis. Hard carbon spheres (HCS) effectively inhibited the agglomeration of hydride particles during hydrogen storage cycling; they could also provide active sites to promote the nucleation of Mg-based hydrides. During the hydriding combustion synthesis procedure, in situ-formed Mg2NiH4 could induce the absorption of MgH2, thus triggering its hydrogen properties. Remarkable enhancement in hydrogen absorption properties of the composite was found. The composite absorbed 6.0 wt% H2 within 5 min at 275 °C; moreover, even at 75 °C, it could still absorb 3.5 wt% H2. Furthermore, it delivered a high reversible hydrogen absorption capacity of 6.2 wt% and excellent rate capability at 350 °C. It was also demonstrated that the composite could release 6.2 wt% H2 at 350 °C within 5 min. A rather low activation energy value (65.9 kJ mol−1) for the dehydrogenation of MgH2 was calculated as compared to that for commercial MgH2 (133.5 kJ mol−1).

Two new types of N-doped ZrO2 photocatalysts ZON and AZON have been synthesized using ethylenediamine as the nitrogen source by a facile and low-cost sol–gel method. The N-doped ZrO2 samples have been characterized using various techniques including X-ray diffraction (XRD), UV-Vis spectroscopy, Fourier transform infrared spectroscopy (FTIR), X-ray photoelectron spectroscopy (XPS), photoluminescence spectroscopy (PL) and N2 adsorption–desorption tests. The XRD analysis shows that the crystallinity of ZON samples calcined at 400–600 °C can be indexed to monoclinic ZrO2; while the AZON samples calcined at 400–550 °C only show amorphous diffraction patterns. The UV-Vis response of both N-doped ZrO2 samples can be extended to the visible light regime. The high resolution XPS spectra indicate that N element has been doped in the lattice of ZrO2. Visible-light photocatalytic reactions using the N-doped ZrO2 photocatalysts (i.e. ZON, AZON) calcined at 450 °C show the highest hydrogen production rate (2.12 mmol g−1 h−1) and best methylene orange degradation performance due to substitutional N-doping of the ZrO2. The novel N-doped ZrO2 materials are demonstrated to be very promising photocatalysts with enhanced visible-light photocatalytic activity. Our results provide useful insights into the development of novel photocatalytic materials for hydrogen production and degradation of organic wastes by narrowing the wide bandgap of semiconductors with high photocatalytic activity under UV-Vis light.

Objectives:

To evaluate the relationship between alpha epithelial sodium channel (alpha-ENaC) T663A polymorphism and the risk of essential hypertension.

Methods:

This meta-analysis was conducted between November 2014 and February 2015 in Shanghai Medical Instrumentation College, Shanghai, China. We collected all published available case-control data (N=12) identified through PubMed, Web of Science, Scopus, and Chinese National Knowledge Infrastructure (CNKI) up to December 2014. The pooled odds ratio (OR) with 95% confidence interval (CI) was calculated using the fixed- or random-effect model.

Results:

Although subgroup analysis showed that alpha-ENaC T663A polymorphism was associated with essential hypertension in North American individuals (OR=1.55, 95% CI=1.22-1.98, p=0.0003), our meta-analysis results did not confirm such association overall (OR=1.03, 95% CI=0.92-1.15, p=0.62). The lack of association was further confirmed by the non-superiority test (p<0.0001).

Conclusion:

Alpha-ENaC T663A polymorphism might not be a risk factor for essential hypertension.Hypertension is one of the most important risk factor for cardiovascular disease.1-3 However, its etiology in the vast majority of cases (~90%) is unknown, and thus the essential hypertension term is employed to describe such situations.4 Nowadays, essential hypertension is considered as a multifactorial disease resulting from the interplay of many genetic, environmental, and behavioral factors.5 Among them, sodium has long been deemed as one of the pivotal environmental factors due to its direct regulatory effect on blood pressure.5,6 The amiloride-sensitive epithelial sodium channel (ENaC) lies in the collecting duct of the kidney, and regulates sodium reabsorption. This channel is composed of 3 homologous subunits: alpha, beta, and gamma. Several studies have reported that mutations in beta-ENaC or gamma-ENaC can result in constitutive sodium reabsorption, thus leading to the development of an autosomal-dominant Mendelian hypertensive disorder, Liddle syndrome.7-11 Therefore, some “milder” mutations or functional polymorphisms were assumed to play some etiological roles in essential hypertension. Following this hypothesis, recent studies have reported some potential associated variants.12 Among them, T663A polymorphism in the alpha-ENaC gene has attracted some attention due to its reported ability to influence the channel activity.13,14 The A allele of T663A polymorphism could reduce the surface expression of ENaC, and the T allele of T663A polymorphism could increase ENaC activity.13,14 Accordingly, Ambrosius et al15 observed that the A allele of T663A polymorphism was associated with being normotensive in Blacks and Caucasians. However, a similar study performed in a Japanese population indicated that the A allele of T663A polymorphism was enriched in essential hypertensive patients,16 and Wang et al17 found that there was a lack of association between the T663A polymorphism and essential hypertension in 2 ethnic groups in China. Therefore, there is still a controversy over the association between T663A polymorphism in the alpha-ENaC gene and essential hypertension. Periacetabular osteotomy (PAO) is a surgical procedure to correct acetabular orientation in developmental dysplasia of the hip (DDH). It changes the position of the acetabulum to increase femoral head coverage and distribute the contact pressure over the cartilage surface. The success of PAO depends significantly on the surgeon's experience. Using computed tomography data from patients with DDH, we developed a 3D finite element (FE) model to investigate the optimal position of the acetabulum following PAO. A virtual PAO was performed with the acetabulum rotated in increments from the original center edge (CE) angle. Contact area, contact pressure, and Von Mises stress in the femoral and pelvic cartilage were analyzed. Five dysplastic hips from four patients were modeled. Contact area, contact pressure, and Von Mises stress in the cartilage all varied according to the change of CE angle through virtual PAO. An optimal position could be achieved for the acetabulum that maximizes the contact area while minimizing the contact pressure and von Mises stress in the pelvic and femoral cartilage. The optimal position of the acetabulum was patient dependent and did not always correspond to what would be considered a “normal” CE angle. We demonstrated for the first time the interrelation of correction angle, contact area, and contact pressure between the pelvic and femoral cartilage in PAO surgery. © 2012 Orthopaedic Research Society. 目的 通过系统评价来评估化痰通腑法治疗急性脑梗死的临床疗效及安全性.方法 检索8个常用中英文数据库,应用RevMan5.4.1软件进行Meta分析,并进行偏倚风险评估.结果 干预组vs对照组:NIHSS的评分较低,BI评分、临床有效率较高.但在中医证候积分上,疗程小于14 d无显著差异,大于14 d疗效较好[MD疗程<14 d=-3.12,95%CI[-7.30,1.05],P=0.14;MD疗程>14 d=-5.77,95%CI(-6.53,-5.01),P<0.00001].结论 化痰通腑法相对常规西医可降低NHISS评分和中医证候积分,提高BI评分和临床有效率,但考虑相关临床研究及样本量较少,仍需开展更多大规模、高质量的研究来进一步验证.  相似文献
