抗高血压药物2-亚氨基-3-(β-羟基苯乙基)-噻唑烷衍生物的合成

2-亚氨基-3-(β-羟基苯乙基)噻唑烷(H_(322))(Ⅰ)是驱虫药四咪唑的中间体。在生产中发现操作工人血压偏低,高血压患者血压下降。为此进行动物试验,发现具有明显的降压活性,并进行了临床观察。据报道,剂量120～150mg/天,分三次口服,疗效为60～70％,但作用时间短,疗效也不够满意。为此进行结构改造并观察其结构与降压作用关系。

2-Imino-3-(β-hydroxyphenethyl)thiazolidine showed hypotensive activity. In order to find more active derivatives, a series of compounds has been synthesized.In preliminary screening tests, Compounds (Ⅱ-2) and (Ⅳ-1) showed considerable hypotensive effect.
