DNA vaccination has caught the attention of many for triggering humoral as well as cellular immune responses. And delivering DNA into the antigen presenting cells (APCs) in order to induce efficient immunoresponse has become the backbone of this field. It has been confirmed that Saccharomyces cerevisiae, though non-pathogenic, is being engulfed by the dendritic cells and macrophages and delivers not only proteins, but also DNA materials (already confirmed in vitro). In this research, S. cerevisiae is used to deliver green fluorescent protein (GFP) reporter gene controlled under cytomegalovirus (CMV) promoter in living organism (mice). The recombinant yeast, transfected with the plasmid containing the GFP gene, was heat killed and orally administered to mice. After 60 h of yeast administration, mice were sacrificed and intestine was separated, washed and frozen in liquid nitrogen. Tissues were cut at the size of 10 μm using Cryostat machine, and GFP expression was successfully detected under a fluorescence microscope. After 45 days Western blot was able to detect GFP antibody in the blood of mice. These results imply that S. cerevisiae, being non-pathogenic, cheap, and easy to culture could be a good candidate to deliver DNA materials to the immune cells for vaccination.  相似文献

The sequences of approximately 34 kb from the 3' end of the varicella-zoster virus (VZV) Oka vaccine strain and the previously sequenced Dumas strain were compared. Sequence differences were noted in the coding sequences of several VZV open reading frames (ORFs), including ORFs 48, 51, 52, 55, 56, 58, 59, 60, 62, 64, and 68. Tests based on differences in the ORF62 gene and in the ORF64 poly-A region successfully distinguished the Oka vaccine strain from its wild-type parent and from other Japanese and US clinical isolates. Mosquito control using insecticides has been the most successful intervention known to reduce malaria prevalence and/or incidence. However, over time success has been hampered due to the development of resistance by mosquitoes against chemical insecticides recommended for public health use. Development of effective botanical mosquitocidal compounds however can be potential alternatives tool in malaria vector control. Thus, the present study aimed at investigating the mosquitocidal effect of “Birbira” (Mellitia ferruginea) seed extract against the primary malaria vector, Anopheles arabiensis from Ethiopia. The mosquitocidal activity of M. ferruginea was assessed following WHO susceptibility test procedure. Methanol extract of M. ferruginea seeds was evaluated against third & fourth instar larvae, pupae and, non-blood fed 3–5 days old laboratory strains and field populations of A. arabiensis under laboratory condition. Mortality was then recorded after 24 h exposure. The seed extract of M. ferruginea showed high mosquitocidal activity against larvae, pupae and adult stages of both the laboratory strain and field population of A. arabiensis. The LC50 values for larvae and pupae population from a laboratory strains was, respectively, 14.7 and 41.33 mg/L. While the LC50 values for the larvae and a pupa of the field population were, respectively, 30.88 and 74.54 mg/L. In addition, the LC99 and LD99 values were also identified. The findings of this study indicated that, the extract could be applied on mosquito breeding sites. The plant extract could be also used for indoor residual sprays after conducting the persistency test.  相似文献

Ethiopian wolves, Canis simensis, live in large multi-male family packs, where males are philopatric and do not disperse. Within a pack, mating and breeding is largely monopolized by the dominant male and female, although extra-pack copulations are common, and subordinate males may sire pups in neighboring packs. Regardless of paternity, all males in a pack help rear the pups. We non-invasively studied patterns in fecal testosterone and glucocorticoid metabolite concentrations using radioimmunoassays of fecal samples collected from nine wild male Ethiopian wolves between August 2007 and February 2008. We tested the predictions of the Challenge Hypothesis, namely that fecal testosterone metabolite concentrations would be higher during the annual mating season, which is the portion of the reproductive cycle when mating and increased aggression typically occur, and lower when there were pups in the pack for which to care. Contrary to the predictions of the Challenge Hypothesis, we did not detect patterns in fecal testosterone metabolite concentrations associated with reproductive stage during our study period. Similarly, we found no patterns associated with reproductive stage in male fecal glucocorticoid metabolite concentrations. Dominant males had higher average fecal testosterone and glucocorticoid metabolite concentrations than did subordinates, which may be related to higher rates of aggression and mate guarding in dominant males of group-living canids, a pattern also reported in African wild dogs, Lycaon pictus.  相似文献
