From the analysis of the fatty acid spectrum of the individual lipid fractions of the cerebral white matter in cyanide induced encephalopathy would appear that essentially the phosphatidyl ethanolamine and plasmalogen fractions showed appreciable deviations from the control spectra. The observed changes in the cholesteryl ester composition did not correlate with the macrophage reaction, which is known to appear as late as 14 days after HCN intoxication when morphological signs of demyelination become apparent. Neither was there a correlation between the alterations of the phosphatide fatty acid composition and that of cholesteryl esters. It thus would appear that esterification of cholesterol in the myelin of rats occurring during early stages following cyanide intoxication constitutes one of the primary factors injuring the myelin sheath. The same conclusion seems to be applicable to the changes in fatty acid composition of the white matter phospholipids.  相似文献

Predictive testing of immunotoxicity associated with chemical compounds is complicated and cannot be accomplished with a single test. As most of the existing tests for immunotoxicity employ experimental animals, there is an increasing need for alternative tests in vitro. We have developed a new system for in vitro immunotoxicity testing, which employs changes in cytokine expression observed in vitro as an endpoint indicating potential for perturbation of the immune system in vivo. This system named "fluorescent cell chip" (FCC) is based on a number of genetically modified cell lines that regulate the expression of a transgene coding for fluorescent protein enhanced green fluorescent protein (EGFP) in a similar way as they regulate expression of IL-1beta, IL-2, IL-4, IFN-gamma, IL-10, TNF-alpha, and beta-actin. Morphological and functional features of selected cell lines expressing EGFP under the control of cytokine promotors were compared with maternal cell lines and this comparison showed that critical functional features of the maternal cell lines were preserved in EGFP expressing cells. Two chemicals with known immunotoxic activities, cyclosporine A and potassium tetrachloro-platinate(II), mediated compound-specific pattern of inhibition and activation of reporter gene expression. Thus, the "fluorescent cell chip" has demonstrated potential for application as a predictive screening test for immunomodulatory activities of chemicals. The major advantage of this approach is the possibility to apply this test in high throughput screening of high number of compounds for their well defined biological activity.  相似文献

Mast cells play an important role at the early stages of immunological response to bacterial infections and parasite infestations. One of the major mast cell proinflammatory mediators is TNF-alpha. Mast cells are considered the only cells capable of storing TNF-alpha in cytoplasmic granules and rapidly releasing it upon activation. To determine what pathway is utilized to direct TNF-alpha to cytoplasmic granules and what motifs are responsible for the sorting process, we constructed a fusion protein covering the full sequence of TNF-alpha, N-terminally fused to enhanced green fluorescent protein (EGFP). In rodent mast cells, such protein was sorted to secretory granules, and this process was inhibited by both brefeldin A and monensin. Considering the relationship between lysosomes and secretory granules and following TNF-alpha sequence analysis, it was determined whether TNF-alpha is sorted through the mannose-6-phosphate receptor (MPR)-dependent pathway. We observed that ammonium chloride and tunicamycin blocked TNF-alpha-EGFP fusion protein delivery to secretory granules. In situ mutagenesis experiments confirmed the necessity of N-linked glycosylation for efficient sorting of TNF-alpha into rodent mast cell granules. In this work we established that TNF-alpha travels from the ER to mast cell granules via a brefeldin A- and monensin-sensitive route, utilizing the MPR-dependent pathway, although this dependency does not seem to be absolute.  相似文献

Inflammatory bowel disease (IBD), which includes Crohn’s disease and ulcerative colitis, is characterized by chronic inflammation of the gastrointestinal tract. IBD has been associated with numerous symptoms and complications, with the most common being iron deficiency anemia (IDA). Iron deficiency in IBD is caused by inadequate intake, malabsorption (including duodenal involvement and surgical removal), and chronic blood loss by mucosal ulcerations. Therefore, an appropriate diet should be enforced. Iron deficiency and iron supplementation have been associated with alterations to gut microbiota. IBD-associated anemia, in particular iron deficiency anemia, is associated with a significant decrease in quality of life and with clinical symptoms such as chronic fatigue, headaches and dizziness, reduced exercise tolerance, pale skin, nails, conjunctiva, and fainting. However, despite these numerous adverse symptoms, IDA remains undertreated. The European Crohn’s and Colitis Organisation (ECCO) guidelines state that patients should be monitored for anemia. Adequate treatment, whether oral or intravenous, should be implemented while taking into consideration C-reactive protein values (CRP), hemoglobin levels, and therapeutic response. It should be stressed that every case of anemia in IBD patients should be treated. Intravenous iron formulations, which are more superior compared to the oral form, should be used. There is a need to increase awareness and implementation of international guidelines on iron supplementation in patients with IBD.  相似文献

The chlorination of iodide-bromide brines during exploitation of therapeutic pools at health resorts brings a hazard of generation of toxic trihalomethanes (THMs), especially bromide derivatives. The study was undertaken to establish the time changes of THM concentration in the pool brine and air in the pool hall. In the pool brine the concentration of THM increased from 40 mg/l at the beginning of the exploitation cycle up to 100 mg/l after 2 months of recirculating the pool water. In the pool hall air the concentration of THM varied from approximately 6 micrograms/m3 to 16.32 micrograms/m3. The values are the concentrations of total THMs per CHCl3, and fall within the range of limits currently admissible for drinking water (so in sport and recreational pools as well). However, they are from 2 to 5 times higher than the admissible level of THMs in indoor pools given in German standards DIN. An interesting finding was over 80% contribution of CHBr3 in the total THMs determined in the pool brine and its 55-59% contribution in the total THMs determined in the pool hall air.  相似文献
