人血清载脂蛋白E火箭电泳测定法

Apo E was purified from the very low density lipoprotein (VLDL) of normal and hypertriglyceridemic subjects by Sephadex G-75, heparin-Sepharose 4B affinity chromatography, and preparative SDS polyacrylamide gel electrophoresis. The antisera were produced by injecting apo E into goats. With apo E antisera, Rocket immuno-electrophoretic method was developed to quantitate apo E levels in normal and hypertriglyceridemic human serum. The detection limit was 5.5 ng apo E. The coefficients of variation of intra- and interassay were 2.6% and 5.2%, respectively. The serum apo E level of 50 normal lipodemic subjects in Chengdu area was 4.7 +/- 1.2 mg/dl.

The serum apoE levels in 238 healthy adults in Chengdu are aware determined by radioimmunodiffusion (RID) assay. The average apoE level was 3.98 +/- 0.98 mg/dl (chi +/- s). The result was basically identical with other reports. There is no difference in sex, and the level has a tendency to increase with age. One hundred and sixty-one hyperlipidemic subjects (including types IIa, IIb, IV and V were compared with the normals. Their serum apoE levels increased by different percentages depending on the type of hyperlipidemia. The percentage of increasing was especially higher in types V and IIb. The content of apoE in serum positively correlated with serum triglyceride (r = 0.65, P < 0.01) and with serum total cholesterol (r = 0.50, P < 0.01).

成都地区238例健康成人及161例高脂血症患者血清载脂蛋白E含量测定

作者采用本室研制的载脂蛋白E(apoE)免疫扩散测定试剂盒测得成都地区238例正常人血清apoE含量呈正态分布,均值为3.98±0.98mg/dl,且apoE含量有随年龄增加的趋势,而与性别无关,161例高脂血症患者血清apoE含量,除Ⅱa型外,均明显增加,并以Ⅴ型及Ⅱb型高脂血症增加最多。血清apoE含量与血清甘油三酯及胆固醇含量呈正相关(r分别为0.65及0.50,P<0.01)。     

高糖及高脂膳食对血浆载脂蛋白AⅠ、AⅡ、B100、CⅠ、CⅡ、CⅢ及E的影响

作者对高糖及高脂膳食对正常人(10人)及内源性高甘油三酯血症者(10人)血浆载脂蛋白(apo)AⅠ、AⅡ、B100、CⅠ、CⅡ、CⅢ及E含量的影响进行了观察。高糖膳食中糖(主要为淀粉)、脂肪及蛋白质分别占总热量的80％、5％及15％;高脂膳食为40％、45％及15％;正常膳食为60％、25％及15％。结果表明,摄取高糖膳食后,apoAⅠ、B100、CⅠ、CⅡ、CⅢ及E轻度下降,apoAⅡ增加。摄取高脂膳食后,血浆apoB100、AⅠ、CⅡ及CⅢ轻度增加,apoCⅠ及E轻度下降,而apoAⅡ无改变。膳食引起的血浆apo的这些改变可能在血脂代谢中发挥调节作用。     

人血清载脂蛋白E火箭电泳测定法

本文报道一种简便、灵敏、特异的人血清载脂蛋白E(apoE)的火箭免疫电泳测定法。本法用自制的羊抗人apoE血清及1.5％的琼脂糖凝胶制成抗体凝胶板,血清apoE含量在1.1～8.9mg/dl范围内,在一定条件下电泳,其火箭峰高与抗原浓度成比例。最低检测量为5.5ng,板内及板间变异系数分别为2.6％和5.2％;回收率为94.0±3.9％(n=6);用此法测得成都地区血脂正常的成年人血清apoE的含量为4.7±1.2mg/dl(n=50)。     

人血清载脂蛋白E酶联免疫测定法的研究

本文报道一种简便、灵敏、特异的人血清载脂蛋白E(apoE)的双抗体夹心酶联免疫测定法。采用羊抗人apoE—IgG 包被的聚苯乙烯板及过碘酸钠法交联的辣根过氧化物酶—羊抗人 apoE IgG交联物。载脂蛋白CⅠ、CⅡ、CⅢ、AⅠ、B及人血清白蛋白不干扰测定。最低检测量为 11ng,测定范围0.45～29mg/dl,板内及板间重复的CV分别为7.4％和6.7～14.9％;回收率为108±5.5％(n=8);测得成都地区血脂正常的成年人血清apoE含量为3.57±1.24mg/dl(n=42)。
