EXPERIMENTAL STUDY ON THE CYTOCIDAL EFFECT OF AT-1840 AND PARVOVIRUS H-1 AGAINST STOMACH CANCER CELLS

We had given evidences of cytocidal effect in vitro of either parvovirus H-I(PV H-1) or AT-1840 (lycobetaine) on gastric cancer cells. They act on different phase of the cell cycle of the gastric cancer cells, with PV H-1 on the late stage of S phase and AT-1840 on M phase. Combined use of these drugs gives better killing effect than the individual drug alone. In gastric cancer-bearing nude mice (ascites and solid tumor forms), both AT-1840 and PV H-1 increased the survival peroids and decreased the size of solid tumors. Depsite these are the experimental study which may not to be the same in human being. However, the study has layed down a solid foundation for our further exploration of these two substances as anti-gastric cancer drugs in the future.     

RADIOIMMUNOLOCALIZATION OF XENOGRAFTED HUMAN GASTRIC ADENOCARCINOMA WITH ~(131)I-LABELED MONOCLONAL ANTIBODY RWS_(4) IN NUDE MICE

The monoclonal antibody (MAb) RWS4 specific to membrane-associated antigen of human gastric adenocarcinoma was purified by protein A-Sepharose 4B affinity chromatography and labeled with 131I by chloramine-T method. 131-RWS,, was injected (65 μCi/10μg/0.2 ml, intraperitoneally) into the stomach cancer-bearing nude mice (solid tumor about 1 cm in diameter), and its biodistribution was studied by SPECT and gamma-counter over a peroid of 7 days. A clear image of transplanted tumor was observed on the 4th day, and the image became more clear on the 6th day. After SPECT scanning, the animals were killed on the 3rd to 7th day separately and radioactivity was detected in various organs. The ratios of T/NT were calculated. The results were shown as follows: tumor/blood, was 3.41±0.29 on the 6th day and the tumor/other organs (liver, spleen, stomach, lung, heart, kidney and brain etc.) were>3. The specificity of the 131I-RWS4 was 7.74±0.65.

循环免疫复合物测定在系统性红斑狼疮诊断及治疗观察中的地位

T亚群细胞与胃癌患者病情的相关

石蒜碱内铵盐和细小病毒H-1对胃癌细胞杀伤作用的实验研究

体外三种胃癌细胞株的染料排斥试验及~3H—胸腺嘧啶掺入试验均证实石蒜碱内铵盐(AT-1840)及细小病毒H-1(PVH-1)对胃癌细胞具有直接杀伤作用,药物浓度与杀伤强度呈正比,最低有效浓度分别为0.1μg/孔和1×10~4/孔。流式细胞仪检测证实AT-2840作用于G2/M增殖期胃癌细胞。PVH-1作用于S期胃癌细胞。PVH-1感染肿瘤细胞后即掺入细胞核内增殖,细胞膜绒毛变性、丢失,其后细胞肿胀、崩解。两药最低有效剂量联用,对胃癌细胞杀伤作用起协同增强效应,抗肿瘤作用可超过任一单独应用最大剂量时的作用强度。AT-1840使实体瘤重量明显减少,PVH-1和AT-1840明显延长带胃癌裸小鼠(腹水型及实体型)的生存期。本研究首次证实AT-1840和PVH-1均为低浓度有效的胃癌细胞直接杀伤因子,且具有协同增强作用。     

抗幽门螺旋菌单克隆抗体的制备和临床应用

用幽门螺旋菌（HP）109免疫Balb/c小鼠，取其脾细胞和小鼠骨髓瘤细胞SP2／0融合，得到3株分泌高效价抗HP单克隆抗体（HP－MAbs）的杂交瘤细胞株SCH7、SCH9和．SCH10。3株HP－MAbs主要与HP58000菌体蛋白条带反应。HP－MAbsSCH9免疫过氧化物酶染色，与18株HP反应均阳性，与空肠弯曲细菌、结肠弯曲细菌及13种常见肠道标准菌株反应均阴性。用该方法检测121例胃粘膜活检成本，阳性率高于分离培养和／或涂片Gram染色法（P＜0．05）。结果表明，采用本方法制备的HP－MAbs敏感、特异，可应用于临床诊断HP感染。     

核素标记抗人胃癌单克隆抗体(RS_4)的定位诊断研究

ABC免疫酶染色法证实单克隆抗体RS4对胃癌细胞有阳性反应。除对结肠癌、小细胞肺癌及舌瘤有弱阳性交叉反应外,对肝、肺癌等各种细胞株均无反应,免疫组化表明:RS4对各类胃癌的阳性率为70％(42/60),其高分化腺的阳性率达100％,与其他非胃癌恶     

人体抑制性单核细胞引起体外植物血凝素淋转低下的实验证据及其临床意义

植物血凝素(PHA)皮试及体外淋转试验是检测各种疾病患者细胞免疫功能最常见的指标之一。通常认为淋转低下代表了T细胞功能缺陷或数量不足。本文则报告由人体抑制性单核细胞引起免疫低反应性的结果。
