目的:检测慢性乙型肝炎患者外周血T细胞表面KIR的表达情况。方法:采用三色流式细胞术检测慢性乙型肝炎患者外周血CD4^+T细胞和CD8^high T细胞表面KIR分子表达,并与正常外周血比较。结果:慢性乙型肝炎患者外周血中CD8^high T细胞KIR表达明显高于对照组CD8^high T细胞。正常外周血CD4^+T细胞几乎不表达KIR,慢性乙型肝炎患者外周血中CD4^+T细胞表达KIR。结论:慢性乙型肝炎患者T细胞表面KIR表达明显增加。  相似文献

To determine the daily energy requirement of elite synchronized swimmers during moderate-intensity training, the average daily energy expenditure measured by the doubly labeled water method, was calculated for nine female Japanese national team synchronized swimmers [four senior; mean (SD) 22.5 (1.0) years old, 52.2 (3.6) kg, and five junior; 17.6 (1.1) years old, 52.8 (2.3) kg]. Their total energy expenditure (TEE) was 11.5 (2.8) MJ · day−1 [2738 (672) kcal · day−1]. When compared with estimated energy requirements derived from “Recommended Dietary Allowances for the Japanese”, 12.1 (0.6) MJ · day−1 [2897 (139) kcal · day−1], there was no difference between mean actual and estimated energy requirements. However, there were considerable differences observed on an individual basis. Their energy intake, estimated from 7- day self-reported dietary records, was 8.9 (1.7) MJ · day−1 [2128 (395) kcal · day−1], which was significantly lower than their TEE (P < 0.05). Resting energy expenditure (REE), as determined by indirect calorimetry, was 5.2 (0.3) MJ · day−1 [1247 (75) kcal · day−1]. Their physical activity level (TEE/REE) was 2.18 (0.43). These results demonstrate that the TEE values of elite female synchronized swimmers are not dissimilar to those reported for athletes participating in other sports, especially competitive swimmers during moderate-intensity training. 目的 阐释川楝素的作用机制 ,为了解递质解释的基本过程提供线索。方法 以大鼠大脑皮层匀浆经密度梯度离心分离获得的突触体作为研究标本 ,分别施加 5 0mmol·L- 1KCl,1.5 μmol·L- 1卡西霉素或 7.5mmol·L- 14 氨基吡啶触发谷氨酸 (Glu)释放。通过检测由Glu氧化脱氢反应与NAD+ 生成的NADH荧光变化测定Glu释放量。结果 川楝素浓度、时间依赖地显著抑制由KCl诱发的Glu释放 ,并主要抑制钙依赖性释放 ;由卡西霉素直接提升胞内钙离子浓度而诱发的Glu释放也被川楝素明显抑制。结论川楝素抑制中枢突触Glu释放 ,该效应与其导致的递质释放机制中钙离子敏感性降低有关。  相似文献

Mutations that cause destabilization of the hemoglobin (Hb) tetramer are a rare cause of hemolytic anemia. In contrast to the hemolytic anemia caused by enzyme deficiencies, a dominant mode of inheritance characterizes the unstable Hbs. Hb Alesha [β67(E11)Val→Met; HBB: c.202G>A] is caused by a G>A mutation at codon 67 of the β-globin gene, resulting in a valine to methionine substitution at helix E11. This replacement disrupts the apolar bonds between valine and the heme group, producing an unstable Hb and severe hemolysis. We report this rare hemoglobinopathy in a Chinese girl with severe hemolytic anemia, splenomegaly and frequent requirement for red blood cell (RBC) transfusions.  相似文献

AIM:Both Hepatitis B virus(HBV)and Hepatitis C virus (HCV) are major causative agents of transfusion-associated and community-acquired hepatitis wordwide.Development of a HCV vaccine as well as more effective HBV vaccines is and urgent task.DNA immunization provides a promising approach to elicit protective humoral and cellular immune responses against viral infection.The aim of this study is to achieve immune responses against both HCV and HBV by DNA immunization with fusion constructs comprising various HCV E2 gene fragments fused to HBsAg gene of HBV.METHODS:C57BL/6 mice were immunized with plasmid DNA expressing five fragmets of HCV E2 fused to the gene for HBaAg respectively.After one primary and one boosting immunizatios,antibodies against HCV E2 and HBsAg weretested and subtyped in ELISA.Splenic cytokine expression of IFN-γ and IL-10 was analyzed using an RT-PCR assay Post-immune mouse antisera also were tested for their ability to capture HCV viruses in the serum of a hepatitis C patient in vitro.RESULTS:After immunization,antibodies against both HBsAg and HCV E2 were detected in mouse sera,with Ig2a being the dominant immunoglobulin sub-class.Highlevel expression of INF-γwas detected in cultured splenic cells Mouse antisera against three of the five nfusion constructs were able to capture HCV viruses in an in vitro assay.CONCLUSION:The results indicate that these fusion constructs could efficiently elicit humoral and Th1 dominant cellular immune responses against both HBV S and HCV E2 antigens in DNA-immunized mice.They thus could serve as candidates for a bivalent vaccine against HBV and HCV infection.In addition,the capacity of mouse antisera against three of the five fusion constructs to capture HCV viruses in vitro suggested that neutralizing epitopes may be present in other regions of E2 besides the hypervariable region1.  相似文献

Prenatal diagnosis of severe α-thalassemia (α-thal) diseases is usually performed by DNA analysis. To establish a simple and rapid method, we evaluated the reliability of cord blood hemoglobin (Hb) analysis using an automated capillary electrophoresis (CE) system. Our results demonstrated that analysis of fetal Hb using the Sebia CapillaryS 2 is an effective, accurate and simple alternative for prenatal diagnosis of Hb Bart's (γ4) disease.  相似文献

目的 探讨舒血宁对老年缺血性脑卒中患者临床疗效和生活质量的影响.方法 212例老年缺血性脑卒中患者随机分为治疗组和对照组各106例,两组均予常规治疗,治疗组加用舒血宁,对照组加用安慰剂,2周为一疗程.进行神经功能缺损评分,采用SS-QOL量表对患者进行生活质量问卷评价,检测血中内皮素1和降钙素基因相关肽.结果 治疗组总有效率为86.8%,明显高于对照组的76.5%(P<0.05);治疗后,治疗组SS-QOL量袁评分明显提高,内皮素1含量下降,降钙素基因相关肽含量上升,与对照组差异有显著性(P <0.05或P<0.01).结论 舒血宁结合常规治疗可明显改善老年缺血性脑卒中患者神经功能缺损评分,提高患者生活质量.  相似文献
