Toll‐like receptor (TLR) 4 signalling is critical for innate immunoinflammatory response and widely triggers the development of various types of clinical diseases. MicroRNA‐7 (miR‐7) is well documented to play an important regulatory role in various biological events. However, the exact role of miR‐7 in TLR4 signalling pathway remains to be fully elucidated. In the present study, we found that miR‐7 expression in TLR4 signalling‐activated bone marrow‐derived macrophages (BMDMs) stimulated by LPS was dramatically increased. Importantly, miR‐7 deficiency significantly enhanced the production of related inflammatory cytokines including IL‐1β, IL‐6 and IL‐12, as well as TNF‐α, on LPS‐activated BMDMs, accompanied by elevated transduction of TLR4 signalling including Myd88‐dependent and Myd88‐independent pathways, whereas miR‐7 overexpression significantly decreased the transduction of TLR4 signalling and the production of related inflammatory cytokines. Mechanistically, we identified family with sequence similarity 177, member A (FAM177A) as a novel target molecule of miR‐7. Furthermore, down‐regulation of FAM177A using RNAi could impair the transduction of TLR4 signalling. Finally, down‐regulation of FAM177A also reversed the effect of miR‐7 deficiency on TLR4 signalling transduction and production of related inflammatory cytokines on BMDMs. Therefore, we provide the new evidence that miR‐7 acts as a novel negative fine‐tuner in regulating TLR4 signalling pathways by targeting FAM177A, which might throw light on the basal understanding on the regulatory mechanism of TLR4 signalling and benefit the development of therapeutic strategies against related clinical diseases.  相似文献

目的 提高病区静脉血标本采集质量。方法以儿童血液肿瘤2个病区34名护士为研究对象,针对静脉血标本退回率为0.95%(53/5 594)的问题,采取ADDIE模式对护士进行培训与静脉血标本采集操作督导。设定改进目标为血标本退回率降至0.49%。结果干预后血标本退回率为0.32%(37/11 527),显著低于干预前(P<0.05);护士血标本相关知识考核成绩显著高于培训前(P<0.05);护士对培训满意度为91.18%~100.00%。结论运用ADDIE模型进行培训与督导,可有效提高护士相关知识与技能,从而提高静脉血标本采集质量。  相似文献

Reticuloendotheliosis virus (REV) has a wide range of avian hosts leading to reticuloendotheliosis, and its characteristic of vertical transmission makes it to be one of the most important diseases in breeder avian populations. Up to date, reports on neoplastic disease caused by REV in breeding ducks are few. Here, spontaneously occurring neoplastic disease caused by REV in breeding Muscovy ducks was reported in Guangdong province, China. The most significant gross lesions of sick ducks were tumour‐bearing liver and enlarged spleen. Histopathological examination found proliferation of malignant lymphoreticular cells in the liver and reticuloendothelial cells in the spleen. REV strain, CH‐GD2019, was successfully isolated using DF‐1 cells, and the presence of REV was confirmed by PCR detection and transmission electron microscopy. The length of complete proviral genome is 8,238 nucleotides. Genetic and phylogenetic analyses revealed that CH‐GD2019 was closely related to chicken‐origin REV strains circulating in China. The results will provide a basic data for better understanding of REV in breeding ducks and suggest that REV from chickens may be a threat to ducks.  相似文献

目的 探讨慢性粒细胞白血病(CML)合并第二实体瘤病人的治疗方案和时机的选择。方法 系统分析1例CML合并宫颈腺癌病人的临床资料,并查阅参考相关文献。结果 对该病人初诊时CML(慢性期)予以伊马替尼治疗、手术联合化疗治疗宫颈腺癌,手术前后停用伊马替尼,术后4个月CML进展为急性淋巴细胞白血病(简称:急淋变),予以达沙替尼50 mg,每天2次,治疗1个月,获得主要分子学反应。结论 (1)CML合并第二实体瘤治疗时应权衡利弊,优先治疗危险度更高的疾病;(2)CML合并第二实体瘤治疗过程中,重点监测血象及骨髓象,评估病情,当CML出现疾病进展时,及时应用第二代酪氨酸激酶抑制剂;(3)CML合并宫颈腺癌的发生机制、以及宫颈腺癌是否影响CML进展尚不明确,有待进一步研究阐明。  相似文献

目的 探讨家庭支持护理模式对入住新生儿重症监护室(neonatal intensive care unit,NICU)的早产儿父母焦虑、抑郁情绪的影响。 方法 采用自身前后对照研究,选取我院NICU的192例早产儿及其父母,对其实施家庭支持护理模式。在早产儿父母参与家庭支持护理前和完成护理支持后的出院日采用焦虑自评量表(self-rating anxiety scale,SAS)和抑郁自评量表(self-rating depression scale,SDS)对其进行调查。 结果 接受家庭支持护理后,早产儿父母的焦虑、抑郁评分均低于实施家庭支持护理前(P<0.05)。 结论 家庭支持护理模式可以有效改善NICU早产儿父母的焦虑和抑郁心理状况,应将早产儿父母纳入NICU整体护理的关注范围。  相似文献

乳腺癌术后放疗可以降低局部复发率,且对于高危乳腺癌患者的长期生存有积极的影响。当前乳腺癌放射治疗的精准性主要依赖于不同的放疗技术、肿瘤的临床分期及病理类型。但随着乳腺癌研究的深入,要进一步提升乳腺癌术后放疗的精准性,乳腺癌本身的生物学特性应该得到重视。据相关文献报道,不同分子分型的乳腺癌其局部复发率有显著差异。因此乳腺癌的分子分型对放疗计划的制定有极为重要的指导意义。本文针对当前乳腺癌分子分型与肿瘤局部复发风险关系的相关研究以及乳腺癌分子分型指导放射治疗的临床进展作一综述。  相似文献
