The actinobacterium strain ABH26 closely related to Saccharothrix xinjiangensis, isolated from an Algerian Saharan soil sample, exhibited highly antagonist activity against Gram-positive bacteria, yeasts and filamentous fungi. Its ability to produce antimicrobial compounds was investigated using several solid culture media. The highest antimicrobial activity was obtained on Bennett medium. The antibiotics secreted by strain ABH26 on Bennett medium were extracted by methanol and purified by reverse-phase HPLC using a C18 column. The chemical structures of the compounds were determined after spectroscopic (1H NMR, 13C NMR, 1H-1H COSY and 1H-13C HMBC spectra), and spectrometric (mass spectrum) analyses. Two new cyanogriside antibiotics named cyanogriside I (1) and cyanogriside J (2), were characterized along with three known caerulomycins, caerulomycin A (3), caerulomycin F (4) and caerulomycinonitrile (5). This is the first report of cyanogrisides and caerulomycins production by a member of the Saccharothrix genus. The minimum inhibitory concentrations (MIC) of these antibiotics were determined against pathogenic microorganisms. 相似文献

Changes in the distribution of the iron-binding protein lactotransferrin have recently been described in the central nervous
system during a variety of neurodegenerative disorders. To investigate whether lactotransferrin is associated with the neuropathological
changes that characterize Parkinson’s disease, we analyzed the distribution of this protein in the mesencephalon of neurologically
normal individuals and patients affected with Parkinson’s disease using quantitative immunohistochemical methods. High levels
of lactotransferrin were observed in a large population of neurons in the substantia nigra of control cases. Lactotransferrin-positive
neurons were severely affected by the neurodegenerative process that occurs in Parkinson’s disease as indicated by a severe
decrease in the number of immunolabeled neurons in all of these cases. Quantitative analysis also demonstrated higher immunolabeling
levels of lactotransferrin in the surviving neurons in the substantia nigra and ventral tegmental area of Parkinson’s disease
cases compared to control cases. These results suggest that lactotransferrin may participate actively in the mechanism of
neuronal degeneration in Parkinson’s disease.

The authors describe their experience with methyl tertiary butyl ether (MTBE) in a larger series of patients than previously reported in order to acquaint physicians with both its effectiveness for dissolution of common bile duct calculi and the limitations of its use. Ten patients with 13 biliary calculi underwent percutaneous stone dissolution treatment with the experimental cholesterol solvent, MTBE. Three stones completely dissolved within 30 minutes, seven were reduced in size, and three were visibly unaffected. All stones not completely dissolved were easily extracted by means of a stone basket except for one in a patient taken to surgery. Although MTBE perfusion is an effective technique for management of biliary calculi, practitioners should be aware that its use is quite time consuming and its odor difficult to control. 相似文献

Detailed analyses of the neuropathologic changes in the cerebralcortex of elderly individuals and Alzheimer's disease patientshave demonstrated that certain components of the neocorticaland hippocampal circuits are likely to be selectively vulnerable.Based on the distribution of neurofibrillary tangles (NFTs)and senile plaques, it has been proposed that a global cortico-corticaldisconnection leads to the loss of integrated functions observedin Alzheimer's disease. In order to investigate the distributionof lesions associated with aging as well as with the earliestsymptoms of senile dementia, we performed a quantitative neuropathologicavaluation of a large series of elderly patients representingthe entire autopsy population for the year 1989 from a geriatrichospital. Among the 145 cases quantitatively assessed, therewere 102 nondemented patients, 33 patients presenting clinicallywith globally intact intellectual function but early signs ofimpairment of specific cognitive functions, and 10 cases withsenile dementia of the Alzheimer type. All of the cases hadNFTs in layer II of the entorhinal cortex, regardless of theirclinical diagnosis, and most cases had some NFTs in the CA1field of the hippocampus. Severe pathologic changes within theinferior temporal neocortex were observed only in the dementedcases. The extent of amyloid deposition was not correlated withthe clinical diagnosis and seemed to be present in the neocorticalareas earlier than in the hippocampal formation. Also, severalcases contained NFTs without amyloid deposition, but amyloidnever occurred without NFTs. These results suggests that involvementof certain structures within the hippocampal formation is aconsistent feature of aging. Thus, involvement of the hippocampalformation may be a necessary, but not sufficient, conditionfor the clinical expression of dementia, which is likely tobe more closely related to the progressive degeneration of selectneuronal populations in the neocortex. 相似文献

We evaluated the immunohistochemical expression of cadherins and CD44
variants in 20 endometriomas, 20 cystadenomas, 20 borderline ovarian
tumours as well as 20 ovarian carcinomas, and the serological and cystic
fluid concentrations of soluble E-cadherin and soluble CD44 standard
(sCD44sdt) in 20 endometriomas, 20 cystadenomas, six borderline and 11
carcinomas of the ovary. In endometriomas, immunostaining of E- and
N-cadherin was negative (20 and 30% respectively). CD44 H, v3 and v6
immunostaining were detected in 63, 10 and 40% respectively. A difference
in immunostaining for E-cadherin was found between endometriomas and
cystadenomas (P < 0.001) and for N- cadherin between endometriomas and
carcinomas (P < 0.001). A difference in CD44H immunostaining was
observed between endometriomas and cystadenomas (P < 0.035) but not with
borderline ovarian tumours and carcinomas. No difference in serum
concentrations of soluble E- cadherins and CD44 standard was found between
the four groups of tumours. Cystic fluid concentrations of E-cadherin were
lower in endometriomas than in borderline tumours and ovarian carcinomas (P
< 0.001). High concentrations of soluble CD44 standard cystic fluid were
found in endometriomas than in other ovarian cysts. Endometriomas and
borderline tumours share alterations of cadherins and CD44 isoforms which
may help in the understanding of the aggressive and invasive potentials of
endometriotic cells.

This study examined genital asymmetry in a large sample of men. The
probands were 6544 non-delinquent men who were interviewed by the Kinsey
Institute for Research in Sex, Gender and Reproduction from 1938 to 1963.
The measures were four indicators of penile and scrotal asymmetry, along
with self-reported handedness, from Kinsey's interview protocol. Most men
reported some degree of lateral asymmetry in their flaccid penis and in
their testicles; less asymmetry was reported for their erect penis. The
asymmetry typically occurred in the left direction, and this pattern
occurred in both right- and nonright- handers. However, this 'leftward'
pattern was significantly less pronounced in nonright-handers. The results
are discussed in relation to previous findings of genital asymmetry in men,
the possible relationship of genital asymmetry to functional cerebral
asymmetry, and recent data suggesting genital asymmetry may predict
patterns of cognitive performance and genital/sexual organ cancers.

Betz cells are giant motoneurons located in layer Vb of the primate primary motor cortex. We conducted stereological analyses of Betz cells and neighboring pyramidal cells from the brains of six neurologically normal elderly humans to determine their volume, total number, and spatial distribution, and to relate these data to functional localization. The distribution of cellular volumes exhibits a bimodal pattern, delineating two different subpopulations. Betz cell volumes follow a mediolateral gradient, the largest Betz cells being located on the most medial part of the motor cortex. Additionally, the shape of Betz cells varies between the rostral and caudal parts of the primary motor cortex, supporting the notion that there are anatomically distinct zones in primary motor cortex. The total number of Betz cells per hemisphere accounts for about one-tenth of the total number of pyramidal cells in layer Vb. Analysis of spatial distribution using Voronoi tessellation revealed maximal clustering of Betz cells in a zone situated two-thirds from the midline along the mediolateral axis of the primary motor cortex. These data suggest that Betz cells have a discrete subregional distribution that may correspond to certain aspects of the functional parcellation of area 4. These results may offer a histological correlate of functional imaging studies and are relevant in the context of neurodegenerative diseases such as amyotrophic lateral sclerosis, progressive supranuclear palsy, and Guamanian amyotrophic lateral sclerosis/Parkinsonism-dementia, and in studies of normal brain aging. 相似文献

Immunoreactive-vasopressin, -oxytocin, -dynorphin, -dynorphin-(1-8), -alpha-neo-endorphin and -[Met]enkephalin were, in each case, present in greater concentrations in dorsal as compared to ventral, and lumbo-sacral as compared to cervico-thoracic, spinal cord. These differences were significantly more pronounced for vasopressin and oxytocin than for the other peptides. Lesions of the hypothalamic paraventricular nucleus depleted levels of immunoreactive-vasopressin and -oxytocin throughout the cord whereas levels of the opioid peptides therein were unaffected. In contrast, destruction of either the supraoptic or suprachiasmatic nucleus failed to change the content of immunoreactive-vasopressin, -oxytocin or any of the opioid peptides in the cord. Dehydration for 3 days depressed levels of immunoreactive-vasopressin, -oxytocin and -dynorphin in the neurointermediate lobe of the pituitary. In distinction, the levels of these were not modified in the spinal cord. Further, treatment with the synthetic corticosteroid, dexamethasone, elevated levels of immunoreactive-vasopressin, -oxytocin and -dynorphin in the neurointermediate pituitary whereas these were unaffected in the spinal cord. It is concluded that vasopressin and oxytocin in the spinal cord are predominantly derived from the paraventricular nucleus, localized in dorsal lumbo-sacral regions of the cord and insensitive to endocrinological manipulations. These pools may, thus, be modulated differently from their counterparts in the neurohypophysis and have a differing role, possibly in the control of the primary processing, autonomic or motor junctions. Further, there is no evidence from these or our prior studies for a close interrelationship of spinal cord vasopressin with dynorphin-related peptides (or oxytocin with [Met]enkephalin), likewise in contrast to the neurohypophysis.(ABSTRACT TRUNCATED AT 250 WORDS) 相似文献
