Fibroblasts incorporated within collagen gels induce a cell-mediated contraction of the gel to form a three-dimensional, tissue-like structure by a mechanism thought to mimic wound contraction in vivo . In this study a gel contraction model was used to investigate the ability of fibroblasts derived from adult gingiva, adult skin and fetal skin to organise a collagen matrix. In addition the effects of interleukin-1β (IL-1β) on the contraction process was also investigated. Over the concentration range 5-50 U/ml, IL-1β induced a statistically significant inhibition of gel contraction in all fibroblast cell types ( P <0.05), although fetal fibroblasts appeared least responsive and gingival fibroblasts most responsive to the inhibitory effects of this cytokine. Comparison of gel contraction by the different fibroblast strains indicated that fetal and gingival fibroblasts shared similar contraction kinetics. For the adult skin fibroblasts, three of five strains studied showed significantly diminished levels of gel contraction compared to fetal and gingival cells. This apparent difference in fibroblast phenotype may, at least in part, explain the fetal-like wound healing pattern seen in the oral mucosa. 相似文献

Conjugate gaze is often defined as the equal angle rotation of the two eyes. For fixation at far distances, the optical axes are parallel and conjugacy is defined irrespective of the coordinate system. For nearby or finite fixation distances, the evaluation of conjugacy for many gaze postures depends on the coordinate system used to measure it. For example, if the eye is elevated or depressed and the eye is rotated about a vertical axis, the intersections of lines of sight with a tangent screen will describe either straight lines for arcs depending on whether the vertical axis is fixed with respect to the head or to the eye. Because of the horizontal separation of the two eyes, the binocular fixation of near targets at tertiary positions of gaze will require a vertical vergence component for head-referenced but not eye-referenced measurements. The vertical gaze alignment of three human subjects was measured as they viewed targets placed at secondary and tertiary eye positions at two different distances. Vertical vergence was either held open or closed-loop. The lines of sight were found lo intersect (i.e. vertical gaze was aligned) regardless of target position or viewing condition. 相似文献

WR2721 is a "prodrug" for a radioprotective thiol which has been proposed for adjunctive use as a free radical scavenger in cancer chemotherapy. When used adjunctively with oxygen radical generating chemotherapeutic agents in mice, however, WR2721 produces synergistic toxicity rather than attenuation of the toxic effects of such agents. The present paper discusses potential mechanisms for such synergistic toxicity. The pathway for glutathione synthesis appeared to be inactivated in mice treated with WR2721. The disulfide metabolite of WR2721 was a potent inactivator of gamma-glutamylcysteine synthetase, the rate-limiting enzyme in glutathione synthesis. The inactivation of the enzyme by this compound was similar to that reported for cystamine, a compound known to form a mixed disulfide with a cysteine residue near the glutamic acid binding site of the enzyme. Oxygen radicals not only inactivated the synthetase, as well, but hastened the oxidation of the free thiol metabolite of WR2721 to its corresponding disulfide. 相似文献

The development of chemotherapeutic approaches to cancer has been hampered by the toxicity of proposed agents for normal rapidly dividing cells. By using neocarzinostatin, a "pro-drug" which is activated by reduction by thiol compounds, adjunctively with 6-mercaptodopamine, a thiol-containing dopamine analogue, we have been able to enhance neocarzinostatin toxicity for cells of the neural crest tumor neuroblastoma. Thiol compounds that are not neurotransmitter analogues do not act synergistically with neocarzinostatin in this system. Since most normal rapidly dividing cells do not have surface dopamine receptors, we propose this approach for the selective targeting of toxicity for neuroblastoma cells. We further introduce cell-selective activation of prodrugs as a new chemotherapeutic strategy which demands further development. 相似文献

1. To compare the properties of the vestibulocollic reflex (VCR) with those of vestibular neurons projecting to the neck [vestibulocollic (VC) neurons], we have studied the behavior of the latter in the decerebrate cat. Neurons were identified by their antidromic responses to stimulation in C1-C2, but not C5. Responses to stimulation of vestibular and neck receptors were produced by rotation of the body and head in vertical planes. 2. We determined the plane of whole body (vestibular) or body with head counter-rotated (neck) rotation, which produced the maximal modulation of each neuron (response vector orientation). Neuron dynamics were then studied with sinusoidal (0.02-2 Hz) stimuli aligned with this orientation. 3. On the basis of dynamics and vector orientation, the neuron was assigned a vestibular input classification of otolith, vertical canal, otolith + canal, or spatial-temporal convergence (STC). 4. The properties of this sample of VC neurons are similar to those of a larger population of vestibular neurons whose projection was not identified. For example, the distributions of cells with different types of vestibular inputs were roughly the same; in particular, few cells showed STC responses. In addition, there was no evidence of significant convergence of like canals across the midline (e.g., right anterior + left anterior). 5. Also similar to the larger unidentified population, 80% of VC neurons tested for neck input received such an input.
