Experiments were carried out on isolated strips of guinea pig taenia coli. The smooth muscle was depolarized in a solution with high potassium concentration (120 mM KCl). The effect of papaverine (in concentrations of 10–5 to 3.10–5 g/ml) on the tone and off-response to a prolonged and strong hyperpolarizing current was investigated on the denervated muscle. Papaverine was found: 1) to abolish contractile responses to application of histamine, bradykinin, and acetylcholine; 2) to reduce the tone of the depolarized muscle and abolish the effect of an increase in the Ca++ concentration in the external medium on muscle tone; 3) to have no effect on the amplitude and velocity of the ascending phase of the off-response; 4) to accelerate the descending phase of the off-response. The following hypotheses are put forward to explain the result: 1) in the cell membrane there are chemically excitable calcium channels which are blocked by papaverine; 2) in the membrane there are calcium leakage channels responsible for the maintenance of tone and blocked by papaverine; 3) papaverine has negligible effect on electrically excitable calcium channels.A. The helix-coil transition in Phage T2 DNA in the presence of 6,4 · 10?3 mol/l Mn2+ is studied using light scattering and UV spectroscopy. The transition range is about 0,5°C. Near the temperature of the end of melting Tf the molecular weight Mw and the radius of gyration Rz of the complex are observed to decrease to about one half. At a temperature 0,1–0,25°C higher than Tf, Mw and Rz pass through a minimum, which implies that aggregation is preceded by unwinding of DNA strands. Thus, rise in temperature rather than Mn2+ -induced aggregation causes DNA + Mn2+ melting.  相似文献

Relapse of acute myeloid leukemia (AML) is thought to reflect the failure of current therapies to adequately target leukemia stem cells (LSCs), the rare, resistant cells presumed responsible for maintenance of the leukemia and typically enriched in the CD34(+)CD38(-) cell population. Despite the considerable research on LSCs over the past 2 decades, the clinical significance of these cells remains uncertain. However, if clinically relevant, it is expected that LSCs would be enriched in minimal residual disease and predictive of relapse. CD34(+) subpopulations from AML patients were analyzed by flow cytometry throughout treatment. Sorted cell populations were analyzed by fluorescence in situ hybridization for leukemia-specific cytogenetic abnormalities (when present) and by transplantation into immunodeficient mice to determine self-renewal capacity. Intermediate (int) levels of aldehyde dehydrogenase (ALDH) activity reliably distinguished leukemic CD34(+)CD38(-) cells capable of engrafting immunodeficient mice from residual normal hematopoietic stem cells that exhibited relatively higher ALDH activity. Minimal residual disease detected during complete remission was enriched for the CD34(+)CD38(-)ALDH(int) leukemic cells, and the presence of these cells after therapy highly correlated with subsequent clinical relapse. ALDH activity appears to distinguish normal from leukemic CD34(+)CD38(-) cells and identifies those AML cells associated with relapse.  相似文献
