对不同工艺和条件制备的 7个类金刚石薄膜 (Diamondlikecarbon ,DLC)试样 ,经X光电子能谱 (XPS)碳相成分分析后 ,分别进行了血小板黏附实验、黏附血小板的形貌观察、分类计数和形态指数计算 ,并通过灰色关联分析 ,研究了碳相成分对血小板黏附量、黏附血小板的形态指数的影响。结果显示 :来自全方位离子注入离子束增强沉积工艺的DLC ,其血小板黏附量和形态指数明显小于等离子体化学气相沉积工艺制备的样品 ;在DLC的 5种碳相成分中 ,DLC碳相与血小板黏附量和形态指数的 (负 )关联度远大于其它碳相成分 ,除此之外只有C H和C O碳相与血小板形态指数的 (正 )关联度较大。表明 :(1)DLC碳相对血小板黏附的影响远较其它碳相成分为大 ,增加DLC碳相的含量是优化DLC血液相容性的关键所在 ;(2 )C H和C O碳相对黏附血小板的变形有促进作用 ,须从工艺上抑制其产生或尽可能降低其含量 ;(3)采用全方位离子注入离子束增强沉积工艺有助于改善DLC的血液相容性。这些结论对设计与改进DLC的制备工艺具有重要的指导意义。  相似文献

目的:探讨过表达miR-126对人肺癌95D细胞体外生长的影响.方法:利用PCR技术成功构建pcDNA3.1(-)-pri-miR-126载体(命名为p-miR-126),将p-miR-126重组质粒体外瞬时转染人肺癌95D细胞,TaqMan探针法检测miR-126的相对表达水平,CCK-8(cell counting kit-8)法和划痕法分别检测95D细胞的增殖、迁移能力.Western Blot检测95D细胞中蛋白AKT和磷酸化AKT(phosphorylation AKT,p-AKT)的表达情况.结果:成功构建miR-126真核表达载体,且该载体可上调miR-126表达(P<0.05),抑制95D细胞的生长和迁移能力(P<0.05).Western Blot结果表明细胞中磷酸化AKT水平显著降低(P<0.05).结论:通过上调miR-126的表达水平可有效抑制95D细胞的体外生长,其作用机制有可能与AKT信号通路的变化有关,但其具体机制仍需后续深入研究.  相似文献

Background:Although existing mycological tests (bronchoalveolar lavage [BAL] galactomannan [GM], serum GM, serum (1,3)-β-D-glucan [BDG], and fungal culture) are widely used for diagnosing invasive pulmonary aspergillosis (IPA) in non-hematological patients with respiratory diseases, their clinical utility in this large population is actually unclear. We aimed to resolve this clinical uncertainty by evaluating the diagnostic accuracy and utility of existing tests and explore the efficacy of novel sputum-based Aspergillus assays.Methods:Existing tests were assessed in a prospective and consecutive cohort of patients with respiratory diseases in West China Hospital between 2016 and 2019 while novel sputum assays (especially sputum GM and Aspergillus-specific lateral-flow device [LFD]) in a case-controlled subcohort. IPA was defined according to the modified European Organization for Research and Treatment of Cancer/Mycoses Study Group criteria. Sensitivity and specificity were computed for each test and receiver operating characteristic (ROC) curve analysis was performed.Results:The entire cohort included 3530 admissions (proven/probable IPA = 66, no IPA = 3464) and the subcohort included 127 admissions (proven/probable IPA = 38, no IPA = 89). Sensitivity of BAL GM (≥1.0 optical density index [ODI]: 86% [24/28]) was substantially higher than that of serum GM (≥0.5 ODI: 38% [39/102]) (χ2 = 19.83, P< 0.001), serum BDG (≥70 pg/mL: 33% [31/95]) (χ2 = 24.65, P < 0.001), and fungal culture (33% [84/253]) (χ2 = 29.38, P < 0.001). Specificity varied between BAL GM (≥1.0 ODI: 94% [377/402]), serum GM (≥0.5 ODI: 95% [2130/2248]), BDG (89% [1878/2106]), and culture (98% [4936/5055]). Sputum GM (≥2.0 ODI) had similar sensitivity (84% [32/38]) (Fisher''s exact P = 1.000) to and slightly lower specificity (87% [77/89]) (χ2 = 5.52, P = 0.019) than BAL GM (≥1.0 ODI). Area under the ROC curve values were comparable between sputum GM (0.883 [0.812–0.953]) and BAL GM (0.901 [0.824–0.977]) (P = 0.734). Sputum LFD had similar specificity (91% [81/89]) (χ2 = 0.89, P = 0.345) to and lower sensitivity (63% [24/38]) (χ2 = 4.14, P = 0.042) than BAL GM (≥1.0 ODI), but significantly higher sensitivity than serum GM (≥0.5 ODI) (χ2 = 6.95, P = 0.008), BDG (χ2 = 10.43, P = 0.001), and fungal culture (χ2 = 12.70, P < 0.001).Conclusions:Serum GM, serum BDG, and fungal culture lack sufficient sensitivity for diagnosing IPA in respiratory patients. Sputum GM and LFD assays hold promise as rapid, sensitive, and non-invasive alternatives to the BAL GM test.  相似文献

目的:观察中药熏蒸联合推拿和针灸治疗中风的临床疗效。方法:选取2018年11月至2020年7月南通大学第二附属医院收治的中风患者100例作为研究对象,按照入院顺序进行编号,按编号单双数不同分为对照组和观察组,每组50例。对照组给予推拿+针灸治疗,观察组则在对照组基础上给予中药熏蒸治疗。比较2组患者临床疗效、红细胞压积(HCT)、纤维蛋白原(FBG)、血浆黏度(CP)、免疫学(lgA,lgG,lgM)、神经缺损(NDF)、运动功能(FMA)、临床痉挛指数(CSI)及不良反应。结果:治疗后,观察组临床有效率为96.00%,对照组为84.00%,差异有统计学意义(P<0.05);2组FBG、CP、HCT、IgA、IgM、IgG水平均显著降低,且观察组显著低于对照组(P<0.05);2组NDF、CSI评分均明显降低,且观察组明显低于对照组(P<0.05);2组FMA评分上升,且观察组显著高于对照组(P<0.05);对照组与观察组的不良反应发生率分别为10.00%和12.00%,差异无统计学意义(P>0.05)。结论:改良中药熏蒸法联合推拿、针灸可改善中风患者血循环状态和免疫调节,促进神经及运动功能恢复,疗效确切。  相似文献

The emergence of graphene-based polymer composite fibers provides a new opportunity to study the high-performance and functional chemical fibers. In this work, we have developed an efficient and convenient method with polydopamine (PDA) to functionalize and reduce graphene oxide (GO) simultaneously, and the modified graphene nanosheets can obtain uniform dispersion and strong interfacial bonding in nylon 6 (PA6). Furthermore, the reinforced PA6 composite fibers were prepared through mixing PDA-rGO into the PA6 polymer matrix and then melt spinning. The functional modification was characterized by surface analysis and structural testing including SEM, TEM, FTIR, and Raman. When the addition amount of the modified GO was 0.15 wt%, the tensile strength and Young’s modulus of the composite fiber reached 310.4 MPa and 462.3 MPa, respectively. The results showed a meaningful reinforcement with an effect compared to the pure nylon 6 fiber. Moreover, the composite fiber also exhibited an improved crystallinity and thermal stability, as measured by DSC and TGA.  相似文献

Acanthopanax senticosus (Rupr. et Maxim.) Harms (AS), a traditional herbal medicine, has been widely used to treat ischemic heart disease. However, the underlying cellular mechanisms of its benefits to cardiac function remain unclear. The present study examined the effects of total flavones from AS (TFAS) on L‐type Ca2+ channel currents (ICa‐L) using the whole cell patch‐clamp technique and on intracellular calcium ([Ca2+]i) handling and cell contractility in rat ventricular myocytes with the aid of a video‐based edge‐detection system. Exposure to TFAS resulted in a concentration‐ and voltage‐dependent blockade of ICa‐L, with the half‐maximal inhibitory concentration (IC50) of 283.12 µg/mL and the maximal inhibitory effect of 36.49 ± 1.95%. Moreover, TFAS not only increased the maximum current in the current–voltage relationship but also shifted the activation and inactivation curves of ICa‐L toward the hyperpolarizing direction. Meanwhile, TFAS significantly reduced amplitudes of myocyte shortening and [Ca2+]i with an increase in the time to 10% of the peak (Tp) and a decrease in the time to 10% of the baseline (Tr). Thus, the cardioprotective effects of TFAS may be attributed mainly to the attenuation of [Ca2+]i through the direct inhibition of ICa‐L in rat ventricular myocytes and consequent negative effect on myocardial contractility. 目的:研究11-羰基-β-乳香酸的抗癌作用,并探讨其作用机制。方法:用MTT法和生长曲线法检测11-羰基-β-乳香酸对人结肠癌HCT-116细胞增殖的抑制作用,计算半数有效抑制浓度(IC50)值;用Hoechst33258荧光染色法观察经药物处理后细胞核的形态变化;用流式细胞仪测定药物处理后HCT-116的细胞周期和细胞凋亡率。结果:结果提示11-羰基-β-乳香酸浓度为15~50μg/ml时,对HCT-116细胞的48h抑制率为8.7%~90.16%,48h的IC50为27.36±0.14μg/ml;生长曲线结果提示11-羰基-β-乳香酸对HCT-116细胞生长的抑制作用呈明显的时效和量效关系;30μg/ml的药物作用48h后,Hoechst33258染色结果显示细胞核出现细胞核固缩,染色质不均一或边集呈致密强荧光,凋亡小体等明显的凋亡形态学变化;流式仪检测结果提示11-羰基-β-乳香酸可阻滞细胞周期并诱导细胞凋亡。结论:11-羰基-β-乳香酸在体外对结肠癌具有较强的抗癌作用,能够阻断HCT-116细胞周期,诱导细胞凋亡。  相似文献
