目的研究冠心病患者血液中单核细胞分化为巨噬细胞并荷脂后胆固醇流出的影响机制。方法分离冠心病患者(试验组)和正常人群(对照组)外周静脉血中单核细胞,并用佛波酯诱导为巨噬细胞,收集分离血清,检测各项血脂指标并备用。细胞用[3H]胆固醇处理24 h使其荷脂,然后分为四组:对照组+正常血清、对照组+高脂血清、试验组+正常血清及试验组+高脂血清。分别检测ABCA1、ABCG1表达及胆固醇流出率。结果与对照组比较,试验组单核/巨噬细胞胆固醇流出能力受损,油红O染色显示细胞内脂质蓄积增加,细胞内胆固醇流出减少,但ABCA1和ABCG1的表达改变不明显。与正常血清比较,试验组及对照组中高脂血清明显影响胆固醇流出能力,细胞内脂质蓄积增加,细胞内胆固醇流出减少,但不影响ABCA1和ABCG1的表达。结论冠心病患者巨噬细胞胆固醇流出能力受损,可能与动脉粥样硬化性血管疾病的发生相关。  相似文献

目的:探讨鼻内镜辅助RS泪道插管术治疗泪道探通无效的儿童先天性鼻泪管阻塞的有效性和安全性,指导临床应用。方法:回顾性系列病例研究。选取2016-09/2019-09在我院就诊的泪道探通无效的先天性鼻泪管阻塞患儿158例158眼作为研究对象,患儿均行鼻内镜辅助RS泪道插管术治疗。术后1、3、6mo定期随访。观察患儿性别、年龄、既往泪道探通次数、术前泪道有无分泌物、治疗效果、并发症、鼻泪管阻塞类型。对年龄、探通次数与手术有效率之间的关系做Spearman秩相关系数计算与分析,对不同类型鼻泪管阻塞的手术有效率采用Fisher确切概率法检验,对有无脓性分泌物的手术有效率做χ2检验。结果:总手术有效率为89.9%(142/158)。手术有效率随着年龄增长呈下降的趋势(rs=-1.000,P<0.01)。手术有效率随着泪道探通次数增多呈下降的趋势(rs=-1.000,P<0.01)。膜性鼻泪管阻塞手术有效率高于复杂性鼻泪管阻塞(P<0.05)。有无脓性分泌物两种症状的手术有效率无差异(P>0.05)。主要并发症有:假道16眼(10.1%)、泪道引流管提前脱出14眼(8.9%)、眼红、刺激性溢泪18眼(11.4%)、上下泪点对合黏连3眼(1.9%)、泪点肉芽肿1眼(0.6%)。结论:鼻内镜辅助RS泪道插管术治疗泪道探通无效的儿童先天性鼻泪管阻塞有较高有效率,较好的安全性。手术有效率随着年龄增长而下降,随着泪道探通次数增多而下降,膜性鼻泪管阻塞手术有效率高于复杂性鼻泪管阻塞。  相似文献

This retrospective multicenter cohort study aimed to compare the outcome of haploidentical hematopoietic stem cell transplantation (HID-HSCT) with matched sibling donor (MSD) and unrelated donor (URD) transplantation in severe aplastic anemia (SAA) patients 40 years of age and older. With a median follow-up time of 17.6 months, 85 consecutive patients were enrolled in the study, and the median patient age was 45 years (40, 58). The cumulative engraftment rates of neutrophil and platelet were 98.8 ± 0.0% and 92.9 ± 0.1%. The cumulative incidences of Grade 2-4 acute graft-versus-host disease (aGvHD) and chronic graft-versus-host disease (cGvHD) at 3 years were 14.1 ± 0.1% and 17.3 ± 0.2%. The 3-year estimated overall survival (OS) and failure-free survival (FFS) were 91.2 ± 3.2% and 89.7 ± 3.5%. In multivariate analysis, the only factor associated with inferior survival was an ECOG score ≥2. HID-HSCT was associated with a higher incidence of GvHD, but the difference of 3-year estimated OS between HID group and the other two cohorts was not significant (86.7 ± 6.4% for HID vs 92.1% ± 4.4% for MSD and 100% for URD, = .481). HID-HSCT might be a feasible alternative option for selected SAA patients aged 40 years and older without a matched donor.  相似文献

The hexagonal and monoclinic phase LaPO4 and LaPO4:Eu nanostructures have been controllably synthesized by a citrate-induced hydrothermal process at 100 °C. The crystal growth of LaPO4 nanostructures was investigated, and the phase transformation of nanostructured LaPO4 was systematically studied by varying the citrate concentration, pH value and reaction temperature. When 0.8 mmol of citrate was added into the reaction system, the hexagonal phase LaPO4 transformed into the monoclinic phase. High concentrations of citrate would lead to the formation of hexagonal phase LaPO4. The photoluminescence properties of the monoclinic phase LaPO4:Eu prepared using a citrate-induced process demonstrate that the electric dipole transition (5D07F2) is stronger than the magnetic dipole transition (5D07F1), which indicated that Eu3+ is in a site with no inversion center. The strongest emission peak of hexagonal phase LaPO4:Eu comes from 5D07F1. Furthermore, the citrate-induced hexagonal phase LaPO4:Eu has a stronger emission intensity than the hexagonal phase LaPO4:Eu prepared not using a citrate-induced process.

BACKGROUND Pancreatic cancer has a poor prognosis;40%–50% of patients have liver metastases at the time of initial diagnosis and only 15%–20% undergo surgical resection. Irreversible electroporation(IRE) is a new, non-thermal local ablation method for solid tumors, which can induce cell membrane permeabilization,resulting in unrecoverable nanoscale perforation and apoptotic cell death without damaging the structural components of tissues.CASE SUMMARY We report the case of a 66-year-old female patient with liver metastasis from pancreatic cancer with a pathological diagnosis of poorly differentiated adenocarcinoma. Carbohydrate antigen 19-9 was elevated to 420.3 U/m L.Computed tomography showed a pancreas mass of 2.7 cm × 2.5 cm and single liver metastasis of 1.4 cm × 1.1 cm in the S6 area. The patient underwent IRE and arterial infusion chemotherapy and received tegafur. The therapeutic effect of the combination treatment has been evaluated as complete response. To date, the patient has survived for > 12 mo and is receiving tegafur as maintenance therapy(at the time this case report was written).CONCLUSION IRE plus arterial infusion chemotherapy and tegafur may be synergistic,providing a reference for treating liver metastasis from pancreatic cancer.  相似文献

BACKGROUND Solid pseudopapillary tumor(SPT) of the pancreas is a rare pancreatic tumor and 10% to 15% of cases are associated with metastasis. Cryoablation is a new method that can induce tumor necrosis, and treatment of tumors by cryoablation can cause anti-tumor immune responses.CASE SUMMARY A 16-year-old woman with SPT of the pancreas developed liver metastases 5.3 years after complete resection of the primary pancreatic tumor. She was admitted with chief complaints of abdominal pain in the upper abdomen and a weight loss of approximately 5 kg over 4 mo. Carbohydrate antigen(CA) 125,carcinoembryonic antigen, and CA 199 were normal. An abdominal computed tomography scan found multiple nodules in the right lobe of the liver that measured approximately 13.5 cm × 10.8 cm × 21.4 cm. Immunohistochemical staining results showed that CD10 and CD56 were positive, and the patient was diagnosed with SPT of the pancreas with liver metastasis. The patient underwent percutaneous cryoablation and interventional embolization. During the 5-year follow-up, the patient remained disease-free after cryoablation, with relatively normal immune function.CONCLUSION Herein, we for the first time report the treatment of liver metastasis from SPT of the pancreas using cryoablation plus interventional embolization, which could be a promising alternative therapy for pancreatic SPT liver metastasis.  相似文献

目的探讨哺乳动物不育系20样激酶1(MST1)及其DNA甲基化在载脂蛋白E基因敲除(Apo E-/-)小鼠肾损伤中的作用及意义。方法实验动物分为2组:(1)Apo E-/-组(n=10):雄性Apo E-/-鼠饲以高蛋氨酸饮食;(2)对照组(n=10):雄性C57BL/6J鼠饲以高蛋氨酸饮食。喂养14周后,全自动生物化学分析仪测定小鼠血清肌酐和尿素氮水平;PAS染色及透射电子显微镜观察小鼠肾脏组织损伤情况;实时定量PCR及Western blot分别检测小鼠肾脏中MST1 mRNA和蛋白表达水平。巢式甲基化特异性PCR(n MS-PCR)检测小鼠肾脏MST1 DNA甲基化水平。结果与对照组比较,Apo E-/-组血清肌酐和尿素氮分别升高了1.1倍和1.6倍(P0.01);PAS染色和透射电镜结果显示,Apo E-/-组较对照组肾脏明显损伤;Apo E-/-小鼠肾脏MST1表达分别与血清肌酐和尿素氮水平呈正相关(R2=0.7571,P=0.0012;R2=0.7342,P=0.0015);n MS-PCR结果显示,Apo E-/-组肾脏中MST1 DNA甲基化水平较对照组显著降低(P0.01)。结论 MST1表达上调可能在Apo E-/-小鼠肾损伤中发挥重要作用,而MST1启动子区DNA低甲基化改变可能是MST1表达上调的重要机制。  相似文献
