Purpose. Due to the importance of drug-polymer interactions in, inter alia, drug loading/release, supramolecular assemblies and DNA delivery for gene therapy, the aim of this study was therefore to establish the mechanism of interaction between a model polymer (Polyacrylic acid, PAA) and a model drug (procaine HCl).
Methods. This was performed by studying the effect of salt (KCl) concentration on their heat released values using Isothermal Titration Microcalorimetry (ITM). The integrated released heat data were computer fitted to a one class binding model and the thermodynamic parameters (Kobs, H, and N) were determined.
Results. As the KC1 concentration was increased, Kobs decreased thus establishing the salt dependence of the interaction. The linear variation of Gobs with Sobs indicated that their interaction was entropically driven. The stoichiometry of the interaction was calculated to be one procaine molecule per monomer of PAA. Dissection of the total observed free energy at each KC1 concentration indicated that the contribution of the non-electrostatic attractions to the interaction of PAA with procaine HC1 was greater than those of the electrostatic attractions.
Conclusions. We have shown that the interaction between PAA and procaine HC1 is dependent upon the presence of counterions (monovalent ions) and is mainly entropically driven. The calculated stoichiometry indicated that one procaine HC1 molecule neutralised one carboxylic acid group on PAA. Although electrostatic interactions were necessary for initiating complex formation, the non-electrostatic forces were dominant in stabilising the PAA-procaine HC1 complex. 相似文献

In a few years, oxaliplatin became a standard treatment of colorectal cancer not only in metastatic situation but also in the adjuvant setting. In the others gastro-intestinal malignancies such as oesophageal, stomach, pancreatic, liver and biliary tract cancers, the effectiveness of the current chemotherapies is unfortunately most often disappointing. Several teams thus decided to evaluate oxaliplatin, essentially in polychemotherapy regimens and sometimes in combination with radiotherapy. Many clinical trials are in progress, others will begin soon. Iran is one of 27 countries that are likely to face increasing water shortage crises between now and 2025 unless action is taken to reduce currently high-per-capita urban water consumption. Accordingly, consumption control in the mega-city of Tehran will be an invaluable achievement. A study of Tehran water consumers has determined that household consumers are responsible for 70 percent of the total consumption. Keeping that figure in mind, the authors set out to assess rates of consumption by water fixtures, with an emphasis on household users, and to examine the effects of installing subcounters and reducers. They selected an apartment complex in which it was possible to install water subcounters for each unit. The first step was to evaluate resident attitudes. Block 3, which had 10 units, was selected to cooperate with the project. The second step was to install counters for all 10 units to determine consumption by different fixtures. (The counters were installed in kitchens, in bathrooms, on toilets, on washing machines, and on flash tanks). In the next step, data entry forms for fixture consumption were completed for a period of 10 days. Then single-handle faucets and reducers were installed, and the outcomes were logged for a period of 10 days. Counter readings were performed by the volunteer residents or by educated personnel every 24 hours, and the total volume of inside consumption was compared with the consumption registered by a base counter placed outside each unit. In the course of the project, the consumption per capita was calculated every 24 hours to yield a real and unbiased model that is applicable to city of Tehran. The results showed a total reduction in water consumption of about 20 percent. Thus, with suitable planning and application of cultural and technical methods, it is possible to optimize consumption in Tehran in the near future. 相似文献

AIM:To evaluate the contribution of the G-197A polymorphism in the interleukin-17(IL-17)promoter region to gastric cancer risk in an Iranian population.METHODS:We performed a case control study using samples from 161 individuals with gastric cancer and171 healthy controls.For each individual,the G-197A genotype was determined by restriction fragment length polymorphism analysis of polymerase chain reaction-amplified fragments.Statistical analyses were performed to determine whether any demographic or behavioral factors,infection with Helicobacter pylori(H.pylori),or a particular G-197A genotype was associated with gastric cancer risk.RESULTS:We found that the G-197A genotype wassignificantly associated with increased gastric cancer risk(P=0.001).Patients who were homozygous(AA)at position-197 were 2.9 times more likely to develop disease(95%CI:1.56-5.4;P=0.001).Furthermore,logistic regression analysis revealed that the presence of a single A allele increased the risk of gastric cancer up to 1.7-fold(95%CI:1.26-2.369;P=0.001).This association was observed for early stage gastric adenocarcinomas only,and was not linked to H.pylori infection.CONCLUSION:These results suggest that carrying one or more G-197A polymorphisms at position-197 in the IL-17 promoter region significantly increases gastric cancer risk in this patient population. 相似文献
