Copolymeric hydrogels constituting of vinylpyrrolidone and methacrylic or acrylic acid repeat units have been prepared and investigated for their ability to act as controlled release vehicles in ophthalmic drug delivery. The materials were synthesized by radical-induced polymerization in the presence of N,N'-methylenebisacrylamide crosslinker, and the influences of network composition and drug solubility upon the swelling properties, adhesion behavior, and drug release characteristics were studied. In vitro release experiments showed that some of these materials could be useful vehicles for the delivery of drugs such as pilocarpine or chloramphenicol, while in vivo studies, using the rabbit model, confirmed their high potential for the controlled ocular delivery of pilocarpine hydrochloride.  相似文献

PURPOSE: The survival of patients with local-regional adenocarcinoma of the esophagus or esophagogastric junction (EGJ) treated with preoperative chemoradiation is much better in patients with pathologic complete response than those with residual tumor. Some adenocarcinomas have mixed patterns, including signet-ring cell and mucinous histology, but the clinical significance of these subtypes is unknown. EXPERIMENTAL DESIGN: We studied 412 consecutive patients with esophageal or EGJ adenocarcinoma treated with chemoradiation followed by esophagectomy (193 patients) or surgery alone (219 patients). We evaluated signet-ring cell and mucinous histology in the resection and pretherapy biopsy specimens and compared clinicopathologic features with overall survival. RESULTS: The fraction of signet-ring cell and mucinous histology was similar in evaluated specimens of patients treated with preoperative chemoradiation or surgery alone (17% and 18%, respectively). The overall survival rate at 5 years of patients treated with preoperative chemoradiation was significantly better if residual signet-ring cell or mucinous histology was present in the esophagectomy specimen (63% versus 28%; P = 0.02). All 13 patients with acellular mucin pools and no residual carcinoma are still alive after an average follow-up time of 36 months. By contrast, in patients treated with surgery alone, overall survival rate was significantly worse if signet-ring cell or mucinous histology was present (14% versus 30%; P = 0.05). In multivariate analysis, overall survival was independently predicted by presence of signet-ring cell or mucinous histology (P = 0.04). CONCLUSIONS: Our study showed that patients with esophageal or EGJ adenocarcinoma who have signet-ring cell or mucinous histology benefited substantially from preoperative chemoradiation and esophagectomy.  相似文献

The present research is focused on the 3-Polker coins issued during 1619–1627 by Sigismund III Vasa, King of Poland. A major financial crisis took place at that time due to the 30-year War, which started in 1619. There are two theories among historians concerning the silver depreciation of these coins. The most common theory (generally accepted without proof) is that the later years of issue are depreciated below 60% Ag. The second theory is based on the medieval sources that indicate inflation during the years from 1621–1625, but the medieval source only refers to the inflation of the type of coins and does not mention the issuer. Therefore, in this study, we use modern investigation techniques and materials science methods to help historians elucidate the aforementioned aspects regarding the medieval period. The XRD investigation results are in good agreement with the SEM-EDX elemental analysis. The coins from 1619 and 1620 have high silver content, namely, 86.97% and 92.49%, which corresponds to good silver. The amount of Ag found in the coins from 1621–1625 issituated in the range of 63.2–74.6%. The silver titleis suddenly restored in 1626 at about 84.3% and is kept in a good range until the end of this decree under Sigismund III in 1627. In conclusion, the second theory was partly validated by our experimental results, certifying the currency depreciation during 1621–1625, but the silver title was not lower than 54.2%. Notably, even this depreciated silver title assures a good quality of the 3-Polker coins compared to similar coins issued in other countries that were copper–silver-plated. Therefore, the 3-Polker coins were preferably hoarded at that time.Small alterations in the mint mark’s design were observed in all the depreciated coins compared to the good ones. This might be a sign for an expert to identify the depreciated coins, a fact which requires supplementary investigations. The silver title’s restoration in 1626 also came with a complete change of the mintmark.  相似文献

Driven by the modern lifestyle, the consumers are interested in healthier and balanced diets, including both probiotics and natural antioxidants. The beneficial health effects of probiotics are mainly due to their capacity of modulating the human intestinal microbiota, although achieving at least a 6 log of viable cells at the targeted site is still challenging. Therefore, in this study, an attempt to improve the bioaccessibility of Lacticaseibacillus casei (L. casei) 431® through a co-microencapsulation by complex coacervation and freeze-drying, using an extract from Cornelian cherry and two biopolymeric matrices, whey protein isolates and casein (WPI-CN) and inulin (WPI-I), was studied. The powders showed a comparable anthocyanin content of around 32.00 mg cyanidin-3-rutinoside (C3R)/g dry matter (DM) and a microbial load of about 10 Log CFU/g DM. A high stability of the lactic acid bacteria was assessed throughout 90 days of storage, whereas the anthocyanins’ degradation during storage followed a first order kinetic model, allowing the estimation of a half-time of 66.88 ± 1.67 days for WPI-CN and 83.60 ± 2.46 days for WPI-I. The in vitro digestion showed a high stability of anthocyanins in the simulated gastric juice, whereas the release in the simulated intestinal juice was favored in the variant with inulin (up to 38%). The use of casein permitted to obtain finer spherosomes, with smaller diameters, whereas a double encapsulation was obvious in both variants, thus explaining the high resistance in the gastric environment. The anti-proliferative effect against the human colon cancer cell line (HT-29) was also demonstrated. No cytotoxicity has been found for the concentrations between 1 and 25 μg/mL for the WPI-I variant, whereas a cell proliferation effect was observed at low concentrations of 1–5 μg/mL.  相似文献

Green route is an economic, facile and eco-friendly method, employed for the synthesis of various types of nanoparticles, having it as a starting point biological entity, especially as a plant extract. The present study aims to obtain silver nanoparticles (AgNPs) starting from an ethanolic extract of Populi gemmae (Pg), by adjusting the reaction parameters. The morphological and structural characterization exhibited that both the reaction temperature and the concentration of metal salt, contributes to the obtaining of Pg-AgNPs with adjustable size and shape. The newly synthesized nanoparticles exhibited a good antibacterial activity on Gram-positive bacteria as well as antifungal activity. The in vitro antiproliferative activity of Pg-AgNPs was assessed on two different cancer cell lines (breast cancer cells—MCF7 and lung carcinoma epithelial cells—A549). Results have shown that the green-synthetized Pg-AgNPs_S2 (obtained at 60 °C, using AgNO3 of 5 M) induced a substantial decrease in tumor cell viability in a dose-dependent manner with an IC50 ranging from 5.03 to 5.07 µg/mL on A549 cell line and 3.24 to 4.93 µg/mL on MCF7 cell line.  相似文献

Amphiphilic molecules AY 9944 and chlorpromazine (CPZ) inhibited DNA synthesis in Concanavalin A-stimulated lymphocytes in a dose-dependent manner. While AY 9944 strongly decreased 7-dehydrocholesterol conversion to cholesterol, CPZ did not significantly affect this reaction. Moreover, the inhibitory effect of AY 9944 and CPZ on DNA synthesis took place in the presence of cholesterol in the culture medium. These findings suggest that the mechanism of inhibition of DNA synthesis by AY 9944 or CPZ is not related to endogenous cholesterol synthesis or exogenous cholesterol supply. Results are discussed in relation to the amphiphilic properties of AY 9944 and CPZ and to the interaction of these drugs with membranes or other intracellular targets such as calmodulin.  相似文献

The paper presents a CNC component manufacturing process using the WAAM process. The study depicts all the execution steps of a component from the CAD drawing, deposition procedure (technological parameters, times, layers, etc.), examination, and economic calculation. The manufacturing of this component using WAAM is more advantageous given the fact that the execution time and delivery are significantly shorter, mainly when a single piece is required and also when discussing the raw material used, usually expensive titanium alloys. For example, for Ti-6AI-V used in the aircraft industry, for which the material price is about 90 Euro/kg, the costs for obtaining a given component using the WAAM process will be about 497 Euro/piece compared to 1657 Euro/piece when using another manufacturing process, as it is shown in this paper. In conclusion, additive manufacturing can easily become a feasible solution for several industrial applications when it replaces a classic manufacturing process of a single component or replacement products, even simple-shaped.  相似文献

Microbial infections associated with skin diseases are frequently investigated since they impact on the progress of pathology and healing. The present work proposes the development of freeze-dried, glutaraldehyde cross-linked, and non-cross-linked biocomposite dressings with a porous structure, which may assist the reepithelization process through the presence of collagen and carboxymethylcellulose, along with a therapeutic antimicrobial effect, due to silver nanoparticles (AgNPs) addition. Phisyco-chemical characterization revealed the porous morphology of the obtained freeze-dried composites, the presence of high crystalline silver nanoparticles with truncated triangular and polyhedral morphologies, as well as the characteristic absorption bands of collagen, silver, and carboxymethylcellulose. In vitro tests also assessed the stability, functionality, and the degradability rate of the obtained wound-dressings. Antimicrobial assay performed on Gram-negative (Escherichia coli), Gram-positive (Staphyloccocus aureus) bacteria, and yeast (Candida albicans) models demonstrated that composite wound dressings based on collagen, carboxymethylcellulose, and AgNPs are suitable for skin lesions because they prevent the risk of infection and have prospective wound healing capacity. Moreover, the cell toxicity studies proved that the obtained materials can be used in long time treatments, with no cytotoxic effects.  相似文献
