Platelet adhesion is a critical step due to its hemostatic role in stopping bleeding after vascular damage. Short dystrophins are the most abundant dmd gene products in nonmuscle tissues, and in association with cytoskeleton proteins contribute to their intrinsic function; while utrophins are dystrophin-homologous related family proteins with structural and functional similarities. We previously demonstrated the presence of Dp71 isoforms, utrophins, and various dystrophin-associated proteins and their participation in cytoskeleton re-organization, filopodia and lamellipodia extension, and in centralizing cytoplasmic granules during the adhesion process of human platelets. To evaluate the morphologic changes and actin-based structures of mdx(3cv) platelets during the adhesion process, we compared the topographic distribution of Dp71d/Dp71Delta110(m) and dystrophin-associated protein in adhered platelets from dystrophic mdx(3cv) mouse. By confocal microscopy, we showed that absence of Dp71 isoforms in platelets from this animal model disrupted dystrophin-associated protein expression and distribution without modifying the platelet morphology displayed during the glass-adhesion process. By immunoprecipitation assays, we proved that up-regulated utrophins were associated with dystrophin-associated proteins to conform the dystrophin-associated protein complex corresponding to utrophins, which might compensate for Dp71 absence in mdx(3cv) platelets.  相似文献

The safety and antifungal activity of LY303366 (LY), a new broad-spectrum semisynthetic echinocandin, were studied against disseminated candidiasis in persistently neutropenic rabbits. In vitro time-kill assays demonstrated that LY has concentration-dependent fungicidal activity. The pharmacokinetics of LY in the plasma of nonneutropenic rabbits suggested a linear relationship between dose and area under the curve (AUC). The times spent above the MIC during the experimental dosing interval of 24 h were 4 h for LY at 0.1 mg/kg of body weight/day (LY0.1), 8 h for LY at 0.25 mg/kg/day (LY0.25), 12 h for LY at 0.5 mg/kg/day (LY0.5), and 20 h for LY at 1 mg/kg/day (LY1). Antifungal therapy was administered to infected rabbits for 10 days starting 24 h after the intravenous (i.v.) inoculation of 10(3) Candida albicans blastoconidia. Study groups consisted of untreated controls (UCs) and animals treated with amphotericin B (AmB; 1 mg/kg/day i.v.), fluconazole (FLU; 10 mg/kg/day i.v.), and LY0.1, LY0.25, LY0.5, or LY1 i.v. Rabbits treated with LY0.5, LY1, AmB, and FLU had similarly significant clearance of C. albicans from the liver, spleen, kidney, lung, vena cava, and brain in comparison to that for UCs. There was a dose-dependent clearance of C. albicans from tissues in response to LY. Among rabbits treated with LY0.1 there was a significant reduction of C. albicans only in the spleen. In animals treated with LY0.25 there was a significant reduction in all tissues but the brain. By comparison, LY0.5 and LY1 cleared all tissues, including the brain, of C. albicans. These in vivo findings were consistent with the results of in vitro time-kill assays. A dose-dependent effect of altered cell wall morphology was observed among UCs and animals treated with LY0.1, and LY0.25, with a progressive transition from hyphal structure to disrupted yeast forms. Serum creatinine levels were higher and serum potassium levels were lower in AmB-treated rabbits than in UCs and LY- and FLU-treated rabbits. LY0.5 and LY1 were well tolerated, displayed predictable pharmacokinetics in plasma, and had activities comparable to those of AmB and FLU in the treatment of disseminated candidiasis in persistently neutropenic rabbits.  相似文献

Porous silicon carbide is a promising material for ceramic receivers in next-generation concentrated solar power receivers. To investigate its tolerance to thermal shock, accelerated ageing of large coupons (50 × 50 × 5 mm) was conducted in a solar furnace to investigate the effects of thermal cycling up to 1000 °C, with gradients of up to 22 °C/mm. Non-destructive characterization by computed X-ray tomography and ultrasonic inspection could detect cracking from thermal stresses, and this informed the preparation of valid specimens for thermophysical characterization. The effect of thermal ageing on transient thermal properties, as a function of temperature, was investigated by using the light-flash method. The thermophysical properties were affected by increasing the severity of the ageing conditions; thermal diffusivity decreased by up to 10% and specific heat by up to 5%.  相似文献

A study is made on some significant aspects of the obstetric history and contraceptive practice in an urban health area by means of a random sample that selected 352 women of reproductive potential. They were interviewed by means of a questionnaire designed for that purpose. Most women in all age groups had a male partner, except for the 15-19 age group, and it seems that often marriage duration is not low. Pregnancy average per woman was high (3.04) and high rates for induced abortion per live birth (0.99) and of induced abortion per woman (1.44) were found, which is indicative of inappropriate contraceptive practice. The contraceptives most commonly used were intrauterine devices (74%) and oral tablets (51%); much less frequent is the use of condoms (5.8%), diaphragms (2.9%), periodic abstinence (2.5%) and breast feeding (2.5%). Results indicate an abuse of abortion as a family planning method, as well as a poor contraceptive practice.  相似文献
