BACKGROUND AND PURPOSE
The serine and cysteine peptidase inhibitor, BbCI, isolated from Bauhinia bauhinioides seeds, is similar to the classical plant Kunitz inhibitor, STI, but lacks disulphide bridges and methionine residues. BbCI blocks activity of the serine peptidases, elastase (Kiapp 5.3 nM) and cathepsin G (Kiapp 160.0 nM), and the cysteine peptidase cathepsin L (Kiapp 0.2 nM). These three peptidases play important roles in the inflammatory process.EXPERIMENTAL APPROACH
We measured the effects of BbCI on paw oedema and on leucocyte accumulation in pleurisy, both induced by carrageenan. Leucocyte–endothelial cell interactions in scrotal microvasculature in Wistar rats were investigated using intravital microscopy. Cytokine levels in pleural exudate and serum were measured by elisa.KEY RESULTS
Pretreatment of the animals with BbCI (2.5 mg·kg−1), 30 min before carrageenan-induced inflammation, effectively reduced paw oedema and bradykinin release, neutrophil migration into the pleural cavity. The number of rolling, adhered and migrated leucocytes at the spermatic fascia microcirculation following carrageenan injection into the scrotum were reduced by BbCI pretreatment. Furthermore, levels of the rat chemokine cytokine-induced neutrophil chemo-attractant-1 were significantly reduced in both pleural exudates and serum from animals pretreated with BbCI. Levels of interleukin-1β or tumour necrosis factor-α, however, did not change.CONCLUSIONS AND IMPLICATIONS
Taken together, our data suggest that the anti-inflammatory properties of BbCI may be useful in investigations of other pathological processes in which human neutrophil elastase, cathepsin G and cathepsin L play important roles. 相似文献3.

AIM: To validate accuracy of a novel chair-side test for matrix metalloproteinase (MMP)-8 as compared to enzyme-linked immunosorbent assay (ELISA) in Periodontal health and disease.
METHODS: Gingival crevicular fluid was collected from 150 subjects, Group 1 (healthy) - 50 subjects, Group 2 (gingivitis) - 50 subjects and Group 3 (chronic periodontitis) - 50 subjects. A chair-side test strip was indigenously prepared using polyclonal antibodies (principle of immunochromatography) to detect the MMP-8 levels. The detection accuracy (sensitivity and specificity) of the MMP-8 levels by chair-side test kit were compared with ELISA at baseline and 3 mo after scaling and root planing among the study population.
RESULTS: The novel chair side test detected MMP-8 levels in accordance with ELISA which at baseline were higher in Group 2 and Group 3 as compared to controls (P < 0.05), and these enzyme levels decreased after therapy (P < 0.05). The chair-side test could differentiate healthy, gingivitis and periodontitis. The detection accuracy of the chair-side test strip were on par with ELISA (sensitivity 92.9% and specificity of 100%) which were statistically significant (P < 0.05). A desire to arouse interest about periodontal health and maintenance in the Indian population provided a strong rationale for us to develop our chair-side test strips to suit our economy. Moreover, this was the first ever effort to develop and validate a chair-side test strip to detect MMP-8 levels in the Indian population. This test can be used on a large scale in private dental practice for the early detection of disease, tapping the sites at risk for disease, alongside helps in patient education and motivation for maintenance.
CONCLUSION: This study shows that the novel chair side test kit detects MMP-8 levels a biomarker of periodontal disease progression accurately making it a good chair side diagnostic tool. Further, it is cost effective and time saving which can make it applicable in private dental practice on a large scale for the early detection of periodontal disease. 相似文献

Efficacy of lycopene in the treatment of gingivitis: a randomised, placebo-controlled clinical trial

PURPOSE: The aim of the present study was to compare the effect of systemically administered lycopene (LycoRed) as a monotherapy and as an adjunct to scaling and root planing in gingivitis patients. MATERIALS AND METHODS: Twenty systemically healthy patients showing clinical signs of gingivitis were involved in a randomised, double-blind, parallel, split-mouth study. The subjects were randomly distributed between the two treatment groups: experimental group (n = 10), 8 mg lycopene/day for 2 weeks; and controls (n = 10), placebo for 2 weeks. Quadrant allocation within each group was randomised with two quadrants treated with oral prophylaxis (OP) and two quadrants not receiving any form of treatment (non-OP). Bleeding index (SBI) and non-invasive measures of plaque (PI) and gingivitis (GI) were assessed at baseline, 1 and 2 weeks. Salivary uric acid levels were also measured. RESULTS: All the treatment groups demonstrated statistically significant reductions in the GI, SBI and PI. Treatment with OP-lycopene resulted in a statistically significant decrease in GI when compared with OP-placebo (p < 0.05) and non-OP-placebo (p < 0.01). Treatment with non-OP-lycopene resulted in a statistically significant decrease in GI when compared with non-OP-placebo (p < 0.01). The OP-lycopene group showed a statistically significant reduction in SBI values when compared with the non-OP-lycopene group (p < 0.05) and the non-OP-placebo group (p < 0.001). There was a strong negative correlation between the salivary uric acid levels and the percentage reduction in GI at 1 and 2 weeks in the OP-lycopene group (r = -0.852 and -0.802 respectively) and in the non-OP-lycopene group (r = -0.640 and -0.580 respectively). CONCLUSIONS: The results presented in this study suggest that lycopene shows great promise as a treatment modality in gingivitis. The possibility of obtaining an additive effect by combining routine oral prophylaxis with lycopene is also an exciting possibility, which deserves further study. 相似文献
