Trypanosoma rangeli is a hemoflagellate parasite of man, domestic and wild animals in Central and South America. The genus Rhodnius is particularly susceptible to infection by T. rangeli and transmission by salivary inoculation has been demonstrated in 12 of 14 nominal species of naturally and experimentally infected insects. This report describes the molecular characterization of 37 strains of T. rangeli isolated from vertebrate and invertebrate hosts. Strains were analyzed by hybridization with kinetoplast DNA (kDNA) probes, polymerase chain reaction (PCR) amplification of kDNA minicircles and random amplification polymorphic DNA (RAPD). Strains isolated from Rhodnius prolixus present KP1, KP2 and KP3 minicircle amplification products but strains isolated from R. colombiensis or Panstrongylus megistus present amplification products derived only from KP2 and KP3 minicircles. The two T. rangeli groups defined as KP1(+) and KP1(-) present a high genetic divergence as they have probably been co-evolutioned with different adaptive radiated lines of the genus Rhodnius in Latin-America. The data obtained from insects with intestinal and salivary glands infections confirm that each Rhodnius species select the sub-population of T. rangeli KP1(+) or KP1(-) which is susceptible to transmit it by salivary inoculation to the vertebrate host.  相似文献

Background  

Antigen specificity and IgG subclass could be significant in the natural history of Chagas' disease. The relationship between the different stages of human Chagas' disease and the profiles of total IgG and its subclasses were thus analysed here; they were directed against a crude T. cruzi extract and three recombinant antigens: the T. cruzi kinetoplastid membrane protein-11 (rKMP-11), an internal fragment of the T. cruzi HSP-70 protein192-433, and the entire Trypanosoma rangeli HSP-70 protein.  相似文献

Despite clinical importance of leishmaniasis, an infectious disease that affects 12 thousand million people in 88 countries, the treatment is still unsatisfactory due to its limited efficacy, cost expensive and undesirable side effects. Aiming to develop new antileishmanial lead compounds, we used a rational approach to synthesize a new set of sulfonamide 4-methoxychalcone derivatives (3a-3i) and evaluate the sulfonamide and methoxy moieties as promising adding-groups to chalcones. For that purpose we tested this new set against Leishmania braziliensis promastigotes and intracellular amastigotes and determined its cell toxicity profile. Interestingly all compounds presented a concentration-dependent antileishmanial profile and the benzylamino derivative (3i) showed a biological activity better than pentamidine. None of these compounds affected Trypanosoma cruzi epimastigotes, which suggests a specific antileishmanial profile. The structure-activity analysis of these sulfonamide 4-methoxychalcone derivatives pointed the molecular volume, the HOMO density concentrated in the chalcone moiety and the conformational structure of the compounds as important structural and stereoelectronic features for the antileishmanial activity. In addition, these compounds also fulfilled Lipinski rule of 5 and presented druglikeness similar to antileishmanial drugs. Altogether these results point the sulfonamide 4-methoxychalcone derivatives as potential lead compounds for designing new candidates for leishmaniasis treatment.  相似文献

Intracellular multiplication of Trypanosoma rangeli was evaluated in vitro using experimental infection of Vero cells, murine macrophages, and promonocytes with T. rangeli Choachi, Macias, and SC-58 clone B1 strains. Our results revealed a low infectivity of all T. rangeli strains to these cell lines. Macrophages showed the highest infection rate; however, intracellular forms were no longer observed 48 h post infection Despite the observation of intracellular parasites up to 144 h post infection, the infection rates of Vero cells and J774G.8 promonocytes with these parasite strains were always below 5%. Pre-incubation of parasites with normal mouse serum increased the initial infectivity but not the time course of the infection. Under our experimental conditions, we did not observe any evidence of intracellular multiplication of T. rangeli within these cell lines.  相似文献

A case of spontaneous rupture of a nonparasitic liver cyst, documented by serial computed tomography scans, is reported. The patient was closely followed clinically after the rupture of the cyst and was treated conservatively, without surgical intervention. No peritonitis developed. This unusual complication of nonparasitic simple liver cysts may not always require surgery.  相似文献

Four Leishmania sp. samples were isolated from autochthonous human cases of American cutaneous leishmaniasis (ACL) in Santa Catarina State, southern Brazil. These strains were characterized using indirect immunofluorescence with a panel of Leishmania-specific monoclonal antibodies (MAbs), and by PCR amplification and hybridization assay of the mini-exon gene with group specific probes. The results obtained with the MAbs were in agreement with the genetic marker. Two isolates (MHOM/BR/89/JSC89-H1 and MHOM/BR/89/JSC89-H2) were identified as L. (Leishmania) amazonensis and two (MHOM/BR/96/LSC96-H3 and MHOM/BR/97/LSC97-H4) as L. (Viannia) braziliensis. The southernmost autochthonous cases of ACL in Brazil are due to two different Leishmania sp. species, confirming the spreading of ACL on the American continent.  相似文献

Two stocks and nine clones of trypanosomes of the subgenusSchizotrypanum were analyzed based on their electrophoretic pattern ofEcoR1 digestion products of kinetoplast DNA (k-DNA) minicricles (schizodeme) and isoenzymes (zymodeme). The trypanosome stocks were isolated fromPhyllostomus hastatus bats collected in different locations in Minas Gerais State, Brazil. The k-DNA pattern showed major variations between stocks and microvariations among the clones. Furthermore, in one of several cloning attempts, two different populations could be isolated. Among the six enzymes studied, glucose phosphate isomerase presented different patterns for stocks and clones. No similarity was found among bat trypanosomes andTrypanosoma (Schizotrypanum) cruzi standard zymodemes. This shows that bat trypanosomes are distinct fromT. (S.) cruzi.  相似文献
