We cultivated kidney hematopoietic cells from clonal triploid ginbuna crucian carp, Carassius auratus langsdorfii. Proliferating cells from hematopoietic cell cultures were harvested and injected into tetraploid hybrids (clonal triploid ginbuna and goldfish hybrid) which possess three sets of chromosomes from a triploid clone and a haploid set of chromosomes from goldfish (Carassius auratus). After injection of cultured triploid cells (donor cells), blood samples were obtained from tetraploid hybrids (recipients) every other week. Blood cells stained with acridine orange were measured by flow cytometry to trace the injected donor cells by means of differences in DNA content. Although erythrocytes were not produced in donor cell cultures, such cultures maintained precursor cells capable of differentiation into erythrocytes in vivo. After 4-12 weeks of transplantation, mature erythrocytes derived from the donors were observed in the blood circulation of the recipient fish. These results indicated that the ginbuna hematopoietic cell culture system is an in vivo situation suitable for the study of hematopoietic control mechanisms.  相似文献

Kinematics of the scaphotrapezio-trapezoidal joint during wrist flexion/extension motion (FEM) and radial/ulnar deviation (RUD) was investigated using a 3-dimensional dynamic motion analysis system. The scaphoid/trapezoid motion was found to be a rotational motion obliquely oriented relative to the sagittal plane of the wrist and described in an ulnoflexion/radial extension motion plane in both FEM and RUD of the wrist. The axis of rotation of the scaphoid/trapezoid motion during both FEM and RUD wrist motions was essentially the same and runs through the radiopalmar aspect of the distal scaphoid and the waist of the capitate. Motion analysis also revealed that the trapezium-trapezoid and trapezoid-capitate joints are essentially immobile. Hence, the scaphotrapezio-trapezoidal motion is considered to be a single degree of freedom that is essentially the same in both FEM and RUD of the wrist.  相似文献

We investigated distal radioulnar joint (DRUJ) stability in different wrist positions and examined the relative contribution of each ligamentous component of the triangular fibrocartilage complex (TFCC) to DRUJ stability. We used nine fresh‐frozen cadavers. The humerus and ulna were fixed at 90° elbow flexion. The radiocarpal unit was translated relative to the ulna in dorsopalmar directions with the wrist in five positions. Displacement of the unit was measured by an electromagnetic tracking device. Magnitudes of displacement were compared between different wrist positions in various sectioning stages: ulnocarpal ligament (UCL) sectioning, radioulnar ligaments (RUL) sectioning, and extensor carpi ulnaris (ECU) floor sectioning. Wrist position and sectioning stage significantly influenced the displacement. In intact wrists, the displacement in wrist extension was significantly lower than that in neutral. However, after UCL sectioning, there were no longer any significant differences. After RUL sectioning, the displacement in radial deviation was significantly lower than that in neutral. Following ECU floor sectioning, there were no longer any significant differences. Thus, in intact wrists, DRUJ stability in wrist extension is likely due to tightening of the UCL. After complete RUL sectioning, DRUJ is stabilized in radial deviation due to tightening of the ECU floor. © 2014 Orthopaedic Research Society. Published by Wiley Periodicals, Inc. Evidence from a recent cadaveric study has revealed that the thickness of the distal interosseous membrane (DIOM) of the forearm shows substantial individual variations, and a thick fibrous tissue known as the distal oblique bundle (DOB) was found in 40 % specimens in the study. Another cadaveric study revealed that greater stability of the distal radioulnar joint (DRUJ) was obtained in a group with DOB following ulnar shortening than in a group without DOB. Thus, the presence of DOB influences the condition of DIOM and the ability of DIOM to provide stability to the compromised DRUJ. In the present study, we carried out a preoperative morphological evaluation of DIOM using ultrasound (US) and compared the US findings with intraoperative findings in order to investigate the efficacy and the meanings of this US evaluation. A total of 14 patients with various diseases or injuries of the distal forearm requiring corrective surgery were enrolled. The mean values of DIOM thickness as measured by US evaluation correlated well with those measured intraoperatively. The sensitivity of US evaluation in confirming presence of DOB was 80 % when the presence of DOB bundle was judged by DIOM thickness. In addition, 4 of 5 patients with chronic DRUJ instability showed no DOB. This study suggested that US can be a useful tool in evaluating DIOM and for detecting the presence of DOB and may provide helpful information regarding DRUJ instability.  相似文献

Chondrodysplastic dwarfism in Japanese brown cattle is an autosomal recessive disorder characterized by short limbs. Previously, we mapped the locus responsible for the disease on the distal end of bovine chromosome 6. Here, we narrowed the critical region to approximately 2 cM by using linkage analysis, constructed a BAC and YAC contig covering this region, and identified a gene, LIMBIN (LBN), that possessed disease-specific mutations in the affected calves. One mutation was a single nucleotide substitution leading to an activation of a cryptic splicing donor site and the other was a one-base deletion resulting in a frameshift mutation. Strong expression of the Lbn gene was observed in limb buds of developing mouse embryos and in proliferating chondrocytes and bone-forming osteoblasts in long bones. These findings indicate that LBN is responsible for bovine chondrodysplastic dwarfism and has a critical role in a skeletal development.  相似文献

PURPOSE: The purpose of this study was to obtain qualitative and quantitative information regarding in vivo 3-dimensional (3D) kinematics of the midcarpal joint during wrist radioulnar deviation (RUD). METHODS: We studied the in vivo kinematics of the midcarpal joint during wrist RUD in the right wrists of 10 volunteers by using a technology without radioactive exposure. The magnetic resonance images were acquired during RUD. The capitate was registered with the scaphoid, the lunate, and the triquetrum by using a volume registration technique. Animations of the relative motions of the midcarpal joint were created and accurate estimates of the relative orientations of the bones and axes of rotation (AORs) of each motion were obtained. RESULTS: The scaphoid, lunate, and triquetrum motions relative to the capitate during RUD were found to be similar, describing a rotational motion around the axis obliquely penetrating the head of the capitate in almost a radial extension/ulnoflexion plane of motion of the wrist. The AORs of the scaphoid, the lunate, and the triquetrum were located closely in space. In the axial plane the AORs of the scaphoid, lunate, and triquetrum formed a radially and palmarly opening angle of 43 degrees +/- 7 degrees, 41 degrees +/- 11 degrees, and 42 degrees +/- 14 degrees with the wrist flexion/extension axis, respectively. CONCLUSIONS: This study reports the in vivo 3D measurements of midcarpal motion relative to the capitate. Isolated midcarpal motion during RUD could be approximated to be a rotation in a plane of a radiodorsal/ulnopalmar rotation of the wrist, which may coincide with a motion plane of one of the most essential human wrist motions, known as the dart-throwing motion.  相似文献

PURPOSE: To obtain anatomic and kinematic information regarding the relative motion of the triquetrum-hamate (TqH) joint. METHODS: In this anatomic study the contact surface constraints of the TqH joint that affect TqH motion were investigated by passively simulating TqH motion according to the kinematic data. Two fresh and 28 embalmed cadaver wrists were dissected. In the kinematic study we studied the in vivo 3-dimensional (3D) kinematics of the TqH joint during radioulnar deviation (RUD) and wrist flexion and extension motion (FEM) in 5 healthy wrists using a magnetic resonance image (MRI)-based markerless bone registration algorithm. Animations of the relative motion of the TqH joint were created and accurate estimates of the relative positions and orientations of the bones and axes of rotation of TqH motion during RUD and FEM were obtained. RESULTS: The anatomic study revealed that the contact surface constraints of the TqH joint include primarily the oval convex surface of the hamate. In the kinematic study TqH motion was likely to be not helicoidal but rotational around an oval convex surface of the hamate. In RUD the triquetral movement was rotation in an ulnoflexion-radial extension plane of the wrist. In FEM it was rotation in an almost flexion-extension plane of the wrist. The axes of rotation of the TqH joint in all wrist motions always ran distal to the TqH joint. CONCLUSIONS: Typical motion of the TqH joint in functional range of motion is not a helicoidal motion on the saddle, but rather a rotational motion on an oval, whose axes of rotation are located on the distal side of the joint.  相似文献

Early, wide excision of heterotopic ossification in the medial elbow   总被引:1,自引:0,他引:1  
To prevent the occurrence of secondary contracture during maturation of heterotopic ossification of the elbow, early, wide excision of heterotopic ossification was performed on 9 consecutive patients with a stiff elbow caused by the mass. The average duration between initial injury and operation was 7.7 months. The thick, elastic mass on the posterior oblique ligament of the medial collateral ligament, which included mature and immature heterotopic ossification, was resected widely with the posterior oblique ligament, the posteromedial joint capsule, and the surrounding scar tissue. After an average follow-up of 52.0 months, the total arc of motion improved from a preoperative mean of 37.3 degrees to a postoperative mean of 112.8 degrees. No patient complained of instability or pain on movement. Despite recurrence of spotty heterotopic ossification at the medial elbow in 5 cases, at final follow-up, a sufficient range of motion was obtained in all patients.  相似文献
