【目的】了解常见的皮肤肿瘤组织环氧合酶-2(COX-2)和凋亡相关基因BCL-2表达的情况及其临床意义。初步探讨COX-2与BCL-2在皮肤肿瘤中过表达对肿瘤形成和发展的作用。【方法】免疫组织化学法检测脂溢性角化病(SK)15例,Bowen’s病(BD)15例,基底细胞上皮瘤(BCE)20例,鳞癌(SCC)20例及正常组织5例COX-2和BCL-2蛋白表达。【结果】各标本的肿瘤组织均有COX-2的表达,总体上的阳性率无显著性差异(x^2=0.148,P〉0.05),染色强度有显著性差异(Hc=90.70,P〈0.05),周围正常组织未见COX-2的表达。SCC、BD表达范围弥漫,以SCC表达强度最为显著;细胞分化高的SCC标本其COX-2的表达较细胞分化低者明显。BCE、SK呈灶性表达,强度不一;正常组织仅于表皮基底层有强度很弱的阳性表达。在SCC、BCE、BD。COX-2表达阳性者BCL-2表达的阳性率较COX-2表达阴性者高(P〈0.05)。【结论】皮肤肿瘤存在COX-2的过表达;检测SCC组织COX-2的表达可预测其病变的恶性程度;COX-2表达的模式较阳性与否、表达的强度对病变的提示更有意义。BCL-2的表达与COX-2的过表达有关,COX-2可能是通过激活BCL-2发挥抗凋亡作用而促进皮肤肿瘤形成及发展。 相似文献

目的:了解表皮肿瘤组织环氧合酶-2(COX-2)及血管内皮生长因子(VEGF)表达的情况及其临床意义.方法:免疫组织化学法检测脂溢性角化病(SK)15例,皮肤原位癌(BD)15例,基底细胞上皮瘤(BCE)20例,鳞癌(SCC)20例及正常组织5例COX-2和VEGF的表达.结果:所检测的各种组织标本均有COX-2的表达,分别为SCC 95.0%,BD 73.3%,SK 46.6%,BCE 68.0%,表达强度以SCC最为显著.VEGF在SCC(85.0%),BD(60.0%),SK(53.3%)的表达阳性率较高,而在正常组织和BCE(13.3%)中基本不表达.SCC COX-2表达阳性者其VEGF表达的阳性率较COX-2表达阴性者高(P<0.05).结论:表皮肿瘤存在COX-2的过表达;VEGF表达与否与组织细胞的性质无关;VEGF的表达与COX-2的过表达有关,COX-2可能与VEGF发挥协同作用,促进SCC的形成及发展. 相似文献

目的研究p62基因在黑素瘤A375细胞生长增殖及细胞周期调控等方面的作用。方法取对数生长期的A375细胞,用干扰p62基因表达的siRNA慢病毒载体转染细胞;MTT法检测转染及正常A375细胞的平均OD值;流式细胞术检测两组的细胞周期分布。结果转染组的平均OD值较正常组小(P0.05);转染组的G0/G1期比例(0.73±0.01)较正常组(0.64±0.02)高(P=0.002);转染组的凋亡率(0.10±0.02)较正常组(0.03±0.01)高(P=0.004)。结论干扰p62表达后,细胞周期阻滞于G0/G1期,p62在维持细胞周期和肿瘤细胞凋亡中起着重要的作用。 相似文献

目的:了解门诊痤疮患者严重程度的相关因素及导致出现痤疮瘢痕的危险因素。方法:对2331例门诊痤疮患者进行问卷调查。结果:14~19岁、居住在农村、初中文化程度、油性皮肤、伴发脂溢性皮炎、每天睡眠时间少于8h的患者痤疮病情更严重。男性、有瘢痕家族史、痤疮程度严重的患者出现痤疮瘢痕的概率比较大。结论:对于具备重度痤疮高危因素的患者,应及早干预,给予积极的治疗。由于重度痤疮出现瘢痕的概率更高,因此早期、及时的治疗对于避免出现瘢痕也有一定帮助。 相似文献

Objective To explore the role of mouse dermis-derived mesenchymal stem cells (mdMSC) on skin repair. Methods mdMSC and human dermal fibroblasts were isolated and identified. Human dermal fibroblasts were cultured alone or eoeultured with mdMSC in Transwell chambers with the density ratio of human dermal fibroblasts to mdMSC being 2/5, 1/1, and 2/1. On day 4 and 8 of culture, the expression levels of hydroxyproline and transforming growth factor-beta (TGF-beta) 1 were measured in the supematant of monoculture and coculture by alkaline hydrolysis and ELISA respectively. Results The level of hydro-xyproline was significantly higher in the supematants of coculture system with a density ratio of 2/5 and 1/1 than that in monoculture supematants of human dermal fibroblasts on day 8 (both P < 0.05). Elevated level of TGF-betal was observed in all coculture supematants on day 8 (all P < 0.01) and in the supernatants of coculture system with a density ratio of 1/1 on day 4 (P < 0.05). There was no significant correlation between the expression level of TGF-betal and hydroxyproline in the coculture supernatants (r = 0.108, P > 0.05). Conclusion In vitro coculture with mdMSC can increase the production of hydroxyproline and TGF-betal by fibroblasts, which may be a mechanism underlying the facilitation of skin repair by mdMSC. 相似文献

目的 了解痤疮患者目前的治疗状况和就医行为特点。方法 对 2 3 3 1例门诊痤疮患者进行问卷式流行病学调查。结果 在近 1年内接受过治疗的痤疮患者占所调查痤疮患者总数的 5 2 .6% ;痤疮程度重、女性患者和广州患者更愿意接受治疗 ;患者对治疗满意的占 43 % ,Ⅲ级和Ⅳ级痤疮患者更容易对治疗不满意 ,而在医院接受治疗、采用维持治疗和同时配合面膜治疗的患者一般对疗效满意。结论 要对痤疮患者进行健康教育 ,及早、正确、足够疗程的治疗和维持治疗对于提高疗效和患者的满意度非常关键 相似文献

目的 观察评价果酸[甘醇酸(glycolic acid,GA)]治疗痤疮及痤疮瘢痕的临床疗效,探讨果酸治疗的适应证、方法及安全性.方法 34例痤疮患者,应用浓度为20%、35%、50%、70%的羟基乙酸,进行化学性剥脱治疗,每3周1次,共治疗5次,通过比较治疗前后皮损计数进行疗效观察,观察项目包括:治疗前后毛孔大小、油脂分泌情况、皮肤光滑度、白皙度、瘢痕大小及深浅度,GA治疗对工作生活的影响及疗效满意程度等,并做出疗效评价.结果 34例患者均按计划完成治疗.痊愈4例,显效15例,有效12例,无效3例,总有效率91.18%.治疗后73.53%(25/34)患者毛孔 改善,55.88%(19/34)患者油脂分泌减少,82.35%(28/34)患者皮肤光滑度提高,61.77%(21/34)患者肤色均匀白皙,67.65%(23/34)瘢痕改善.结论 GA适合用于治疗轻、中度痤疮,并能改善毛孔粗大,减少油脂分泌,提高皮肤光滑度,使肤色均匀白皙,减轻痤疮瘢痕,安全且无严重不良反应,患者易于接受,并得较好的评价. 相似文献
