①目的 探讨骨感染病人肿瘤坏死因子α(TNF α)、白细胞介素 6 (IL 6 )和C反应蛋白 (CRP)水平的变化及意义。②方法 采用双抗体夹心ELISA法测定 30例骨感染病人及 2 5例正常人的血清TNF α、IL 6水平 ,采用散射比浊法测定CRP水平。③结果 骨感染病人血清中TNF α、IL 6及CRP含量均处于高水平状态 ,与健康人比较 ,差异有显著性 (t=2 .2 6~ 10 .6 7,P <0 .0 5、0 .0 1)。④结论 TNF α、IL 6及CRP是预测骨感染病人病情危重的重要指标之一 ,对临床有一定指导意义 相似文献

Previous studies from this laboratory indicated a role for epidermal growth factor (EGF) in androgen-dependent male reproductive tract differentiation of the fetal mouse. Expression of an EGF-like protein during Wolffian duct differentiation was indicated from the determinations by radioimmunoassay (RIA) and radioreceptor assay. To further characterize the protein and to assess its role in male sexual differentiation, expression of the protein has been analyzed by Western blot assay and its tissue-specific cellular expression has been determined by immunocytochemical assay in the present study. Western blot analysis of the 18-day fetal male reproductive tract detected an immunoreactive band of the predicted 6-kDa size. Immunocytochemical analysis also detected EGF-specific immunostaining in the Wolffian duct derivatives. At day 18 of gestation, the staining was localized predominantly in the epithelial nuclei of the Wolffian duct derivatives whereas at days 14 and 16 of gestation, the staining was equally distributed in the mesenchymal and epithelial sites of the Wolffian duct derivative. The intensity of the staining increased with progression of differentiation during the 14th–18th days of gestation. Prenatal exposure to the antiandrogen flutamide significantly reduced the immunostaining of the duct. Thus, a role for EGF in Wolffian duct differentiation is indicated. 相似文献

Summary The metabolic potency of recombinant human insulin-like growth factor II was studied in anaesthetized adult rats by obtaining dose-response curves for the hypoglycaemic action and for the stimulation of glucose metabolism during euglycaemic clamping. Compared to insulin, about 50 times higher doses of insulin-like growth factor II were required to result in identical in vivo responses, with half-maximally effective serum concentrations for the stimulation of glucose disposal during clamp studies of about 0.8 and 50 pmol/ml, respectively. A similar difference in potency was observed for the dose-dependent stimulatory actions on glucose metabolism in individual target tissues. Half-maximally effective serum concentrations in the range of 0.8 to 3.0 pmol/ml for insulin and of 40 to 70 pmol/ml for insulin-like growth factor II were seen to be required for 2-deoxyglucose uptake, glycogen formation in skeletal muscle and lipogenesis in epididymal fat. Maximal responses were identical with both peptides. These data suggest that in vivo acute metabolic actions of insulin-like growth factor II on carbohydrate metabolism occurred through insulin receptors. 相似文献

The density of substance P (SP)-, calcitonin gene-related peptide (CGRP)- and vasoactive intestinal polypeptide (VIP)-immunoreactive
(ir) nerve endings was quantitatively evaluated in intact and inflamed gastrocnemius-soleus muscle of the rat. In persistently
inflamed muscle (12 days after a single injection of Freund’s adjuvant into the muscle), the density of SP-ir fibres was significantly
increased. CGRP- and VIP-ir fibres displayed an insignificant increase in density. The density of fibres ir for nerve growth
factor (NGF) and for growth-associated protein 43 (GAP-43/B-50), a marker for axonal sprouting, regeneration and synaptic
reorganisation, increased significantly in persistently inflamed muscle. The data are consistent with the established contribution
of NGF on the expression of SP and GAP-43 in afferent neurones under the influence of a persistent inflammation.

The effects of administrating recombinant human granulocyte colony-stimulating factor (rhG-CSF) and passively transferring immune serum on infection with an attenuated variant of Plasmodium berghei XAT (Pb XAT), in severe combined immunodeficiency (SCID) mice were examined. In immune competent (C.B-17) mice, the attenuated parasite infection was inevitably self-resolving and degenerating forms inside erythrocytes appeared, coinciding with the drop in parasitaemia, whereas SCID mice were unable to control parasite growth and all the mice died. Continuous administration with rhG-CSF caused neutrophilic granulocytosis in both SCID and C.B-17 mice. The effect of rhG-CSF on the infection in C.B-17 mice was to suppress the course of the parasitaemia at an early phase whereas it had no effect in SCID mice. When immune serum was transferred on the day of infection, the prepatent period was prolonged two days in both SCID and C.B-17 mice. When administration with rhG-CSF was combined with transfer of immune serum, SCID mice showed four days delay in patency and degenerating parasites were seen during the course of parasitaemia, although the infection was ultimately fatal. C.B-17 mice similarly treated showed a seven day delay in the onset of the patent parasitaemia which was of a lesser magnitude and shorter in duration compared with control mice. On the other hand, when C.B-17 mice were splenectomized three weeks before infection and then treated with rhG-CSF and immune serum, no degenerating parasites were seen during the infection and all mice died with high parasitaemias. These results show that antibody-dependent neutrophil-mediated parasite killing may occur in the spleen of mice infected with P. berghei XAT. 相似文献

目的 :研究低氧对体外培养的口腔鳞癌细胞系血管内皮生长因子 (vascularendothelialgrowthfactor ,VEGF)和明胶酶 A(matrixmetalloproteinase 2 ,MMP 2 )的影响。 方法 :分别用酶联免疫吸附实验 (ELISA)和半定量逆转录聚合酶链反应 (RT PCR)测定了低氧处理不同时间段时口腔鳞癌细胞系TSCCa和GNM细胞的细胞中VEGF和MMP 2的活性和mRNA表达情况。结果 :低氧处理 4h时 ,VEGF和MMP 2的活性便显著增加 ,8h时达到最高 ,GNM细胞中VEGF和MMP 2分别增加 2倍和 2 .5倍 ;而TSCCa细胞中VEGF和MMP 2增加的更为明显 ,分别增加了 6倍和 4倍。RT PCR结果显示GNM细胞VEGF和MMP 2mRNA表达水平均较TSCCa细胞高 (P <0 .0 5 ) ,低氧处理时在TSCCa细胞中VEGF和MMP 2增加的尤为明显。结论 :低氧可通过调节口腔鳞癌细胞VEGF和MMP 2的活性和mRNA的表达在口腔鳞癌血管形成中起重要作用 相似文献

目的探讨早产儿血清表皮生长因子水平与脑发育的关系。方法依据入选标准观察对象为自2005年8月起连续收治入本院新生儿病房、出生24h内的胎龄为34周±3d的“正常”早产适于胎龄儿30例。采用酶联免疫吸附测定(ELISA)法,分别测定30例早产儿生后24h内和出生第8天空腹股静脉血清表皮生长因子(EGF)水平,分别为t0和t1值,计算血清EGF浓度变化的速率(Δt)。同时,在新生儿出生后6周(纠正胎龄接近40周)和纠正1月龄时进行NBNA评分测定。并将以上实验室数据与新生儿行为神经测定(NBNA)评分值及差值、早产儿出生后的头围增长值及增长速率进行Pearson相关分析。如果P值<0.05,则认为差有统计学意义。结果30例早产儿观察对象t0为(234.71±58.07)pg/ml,t1为(347.81±87.49)pg/ml,Δt为(16.16±9.94)(12.14、15.20)pg/ml/d;Pearson相关分析显示2次NBNA评分结果及差值、出生后6周和纠正1月龄时的头围增长值/增长速率与t0值无相关性(P>0.05);2次NBNA评分结果及差值、出生后6周和纠正1月龄时的头围增长值/增长速率与t1和Δt之间有相关性(P<0.05)。结论研究结果表明,早产儿出生后血清EGF浓度呈上升趋势,提示早产儿出生后体内EGF合成和释放增加,可能促进脏器成熟和组织修复;更重要的是,早产儿出生1周时高水平血清EGF浓度和/或较快上升速率者,其出生后头围增长速度也快,可能有较好的脑发育水平、远期预后良好。 相似文献

The treatment times for the fabrication of Extensive Amalgam Restorations (EAR) as recorded in a longitudinal clinical trial at the Dental School in Nijmegen were analyzed. 269 EAR's with different retention methods were made on molars by three operators. A mean total treatment time of 60 min was found. The factor operator had a significant influence on all treatment phases resulting in a 26% difference in total treatment time between operators. Also significant influences were found in some treatment phases for the factor retention, side location of the tooth and extension of the restoration. The results of this study with regard to treatment times must be considered from the perspective of the quality of the care provided. 相似文献

The effect of human recombinant TNF on the growth of T. musculi has been investigated. When added to parasites cultured in vitro, TNF inhibited their growth. In the presence of thioglycollate-elicited peritoneal exudate cells, the opposite effect was seen and TNF enhanced the growth of trypanosomes in vitro. Similarly, administration of TNF in vivo during the course of infection led to a net increase in the parasite population. It is suggested that TNF exerts a direct antitrypanosomal effect while simultaneously promoting the growth of the parasite through an indirect effect mediated via the host's cells, possibly the macrophages. 相似文献
