Alcohol is an important risk factor for human oesophageal cancer. There is evidence from epidemiological studies that some
specific alcoholic drinks, e.g. Calvados apple brandy, are associated with a greater risk than others. Alcohol induces cytochrome
P450 2E1 (CYP2E1) and the hypothesis was tested that different alcoholic beverages, containing a variety of alcoholic compounds,
could differentially induce expression of cytochrome P450 enzymes. Twelve groups of five rats each were treated for 3 days
with different alcoholic beverages (ethanol alone, whisky, farm-produced or commercial Calvados brandy, beer, cider, wine)
adjusted to 4, 10 or 20% of ethanol in drinking water. Immunoblotting using a monoclonal antibody specific for rat CYP2E1
revealed a single protein band in liver microsomes. Densitometric quantitation of microsomal proteins demonstrated a significant
two-, three- and sixfold increase in band intensity after treatment with ethanol concentrations of 4, 10 and 20% respectively,
compared to control rats drinking water alone. There was a dose-dependent increase in liver microsomal metabolism of CYP2E1
substrates (para-nitrophenol and dimethylnitrosamine) in ethanol-treated rats. However, there were no significant differences
in the level of CYP2E1 protein or enzymatic activity between the different alcoholic beverages at the same ethanol concentration.
There was a slight increase in hepatic CYP1A-related enzymatic activities in the alcohol-treated rats compared to the controls,
but no difference between the treated groups either with dose of ethanol or type of beverage. These data show that induction
of CYP2E1 with acute alcohol treatment is predominantly determined by the ethanol content of the beverage.

对我国1987年流行的登革2型病毒43株包膜E蛋白基因的核苷酸序列进行了分析。结果表明登革2型病毒43株包膜E蛋白基因核苷酸序列含1485个核苷酸,编码495个氨基酸,并就其核苷酸序列及其相应的氨基酸序列与其它的登革2型病毒株进行了比较,发现核苷酸序列与我国1985年分离的登革2型病毒04株,新几内亚C株(NGC),牙买加株1409(JAM)和马来西亚当地流行株M1(登革出血热)、血2(登革休克综合征)、M3(登革热)同源性分别是95.8%、94.6%、97.5%、925%、92.7%和939%,氨基酸序列的同源性分别是94.3%、94.3%、96.0%、93.7%、93.7%和91.5%,推断出的氨基酸序列显示出12个保守的半胱氨酸残基和两个潜在的糖基化位点,分别位于Asn-67和Asn-153位。 相似文献

本文报告260例恶性肿瘤患者血清维生素A、E、C水平的测定结果。发现除慢性白血病外,蓁7种恶性肿瘤患者血清维生素A水平平均显著低于健康人(P<0.01);急性白血,肝癌、食管癌患者血清维生素E水平显著低于健康人(P<0.05);急性粒细胞白血病、急性单核细胞白血病、肝癌患者血维生素C水平显著低于健康人(P<0.01)。本研究工作为维生素A、E、C药物在肿瘤临床上的应用提供了参考。 相似文献

对两组大鼠分别喂饲核黄素缺乏(RD)膳和核黄素添加(R8,22mg/kg饲料)膳8周后,测定了两组大鼠的红细胞维生素E(RBLVe)、红细胞超氧化物歧化酶(SOD)和红细胞丙二醛(MDA)的水平。结果发现:RD组RBCVe水平(4.7173±0.7710mg/g蛋白质)显著低于RS组(5。3868±1.1537mg/g蛋白质,P<0.05)。而RD组的RBCSOD(7745.2±610.1u/g蛋白质)和MDA(0.6868±0.1372μg/g蛋白质)则分别显著低于和高于RS组(8268.5±301.0nu/g蛋白质,0.5548±0.0980,P<0.05)。研究提示,核黄素缺乏引起细胞膜脂质过氧化加重可能RBCVe消耗增加。 相似文献

As vitamin E enhances immune responses, it may reduce dietary ethanol (EtOH)-induced immune suppression, thereby favorably afffecting host disease resistance. The effects of dietary vitamin E at higher level in alcohol-fed female C57BL/6 mice was determined via in vitro cytokine production by splenocytes and thymocytes, and some other immune functions. A 15-fold increase of vitamin E (160IU/liter) in a liquid diet (National Council Research), with or without EtOH (4.5%, v/v), was fed to mice for 10 weeks. Vitamin E supplementation restored production of interleukin-2, -5, -6, -10, and inter-feron-γ by concanavalin A (Con A)-stimulated splenocytes and in terleukin-6 and tumor necrosis factor-a by lipopolysaccharide-stimulated splenocytes, which were suppressed by dietary EtOH. However, it had no effect on interleukin-4 secretion, which was also reduced by splenocytes from EtOH-fed mice. Vitamin E supplementation also restored EtOH-suppressed, mitogen-induced splenocyte proliferation, but not thymocyte proliferation, although it slightly increased production of immunoglobulin A and G by lipopolysaccha-ride-stimulated splenocytes, which were suppressed by dietary EtOH. Dietary vitamin E, furthermore, significantly increased interleu-kin-2 and -6 secretion by Con A-stimulated thymocytes, which were suppressed by dietary EtOH, although it had no effect on interleukin- 4 and interferon-γ production by Con A-stimulated thymocytes from EtOH-fed mice. These data suggest that dietary vitamin E supple-mentation can modulate dysregulation of cytokines initiated by dietary EtOH and restore immune dysfunctions induced by EtOH ingestion. 相似文献
