合欢皮与山合欢皮氨基酸及微量元素的分析

目的 :探讨山东、湖北合欢皮与山合欢皮的内在质量。方法 :用氨基酸自动分析仪和原子吸收分光光度计对山东、湖北合欢皮与山合欢皮的氨基酸及部分微量元素进行分析。结果 :山东、湖北合欢皮与山合欢皮均含大量人体必需氨基酸和微量元素。结论 :为评价产地对合欢皮质量的影响以及合欢皮与山合欢皮的质量差异提供了依据     

二氧化硅对大鼠血浆中微量元素含量的影响

给大鼠经气管分别注入４、１６、６４ｍｇ石英粉尘生理盐水混悬液。在染尘后第７，１５和３０天解剖动物，测定血浆中Ｃｕ、Ｚｎ，Ｍｎ、Ｍｏ、Ｓｉ、Ｒｂ和Ｓｒ等７种微量元素的含量。实验结果表明，随着染尘剂量的增加，血浆中铜的含量明显增高，呈正相关，相关系数为０．９９９。而血浆锌的含量随着染尘剂量的增加而明显的降低，呈负相关，相关系数为－０．９９８。血浆中Ｍｏ、Ｍｎ、Ｓｉ、Ｒｂ和Ｓｒ含量与对照组比无明显差异。     

A review of Disrupted-In-Schizophrenia-1 (DISC1): neurodevelopment, cognition, and mental conditions.

Disrupted-In-Schizophrenia-1 (DISC1) is a promising candidate gene for schizophrenia (SZ) and bipolar disorder (BP), but its basic biology remains to be elucidated. Accumulating genetic evidence supports that DISC1 is associated with some aspects of cognitive functions relevant to SZ and BP. Here, we provide a summary of the current updates in biological studies of DISC1. Disrupted-In-Schizophrenia-1, preferentially expressed in the forebrain, has multiple isoforms with potential posttranslational modifications. Disrupted-In-Schizophrenia-1 protein occurs in multiple subcellular compartments, which include the centrosome, microtubule fractions, postsynaptic densities, actin cytoskeletal fractions, the mitochondria, and the nucleus. Recent studies have clarified that DISC1 mediates at least centrosome-dynein cascade and cyclic adenosine monophosphate (cAMP) signaling. Furthermore, both cytogenetic and cell biological studies consistently suggest that an overall loss of DISC1 function (either haploinsufficiency or dominant-negative, or both) may be associated with SZ and BP. On the basis of these findings, production of DISC1 genetically engineered mice is proposed as a promising animal model for SZ and BP. Several groups are currently generating DISC1 mice and starting to characterize them. In this review, the advantages and disadvantages of each animal model are discussed.     

东莞市正常儿童头发中微量元素含量参考值的研究

目的探讨东莞市区幼儿园3～7岁学龄前正常儿童头发中微量元素含量均值,以此作为东莞市区儿童头发中微量元素正常参考值.方法采取在群体自愿参加者中,选出经体检无重要脏器病变,营养一般,身高、体重在均值以上(称正常儿童).在枕后贴发根部剪0.5g头发检测其锌、铁、钙、铜、锰、铅元素含量.结果在1125例受检儿童中(称普查儿,占适龄儿95%).筛选出621例儿童,占总受检儿童的55.2%.这621例正常儿中,他们头发中锌、铁、钙、铜、锰、铅元素的含量分别为:93.473μg/g、29.710μg/g、779.50μg/g、9.773μg/g、3.222μg/g、9.251μg/g.结论检测的结果显示,2003年东莞市区正常儿童与2003年普查儿童相比,正常儿童发中微量元素含量明显升高,但差异无统计学意义.2003年东莞市正常儿童发中微量元素均值与广州市正常儿童相比,显著低于广州市正常儿童.由此可见,正常儿童发中微量元素含量均值受地域影响而有较大差异.东莞市区儿童发中微量元素含量应参照东莞市正常儿童发中微量元素含量均值较为合理.     

视网膜特异性表达人血管内皮生长因子 基因载体的构建

目的构建在视网膜组织特异性表达的人血管内皮生长因子（VEGF）₁₆₅基因。方法用聚合酶链反应(PCR)方法从BLAB/C鼠全基因组扩增能在视网膜组织特异性表达的rho启动子，经限制性内切酶纯化后克隆于质粒pcDNA^3.1+-VEGF₁₆₅中，建立重组质粒pcDNA^3.1+-rho-VEGF₁₆₅，通过限制性内切酶酶切分析及PCR鉴定筛选出正确重组质粒pcDNA^3.1+-rho-VEGF₁₆₅,由jetPEI介导转染人视网膜色素上皮细胞和人脐静脉内皮细胞，并通过免疫组织化学染色以及绘制细胞生长曲线检测在人视网膜色素上皮细胞和人脐静脉内皮细胞中VEGF蛋白的表达。结果在人视网膜色素上皮细胞中，重组质粒pcDNA^3.1+-rho-VEGF₁₆₅比质粒pcDNA^3.1+-rho-VEGF₁₆₅的VEGF蛋白表达强，在人脐静脉内皮细胞，两者的表达量无明显差别。结论pcDNA^3.1+-rho-VEGF₁₆₅载体的构建为进一步研究VEGF在视网膜新生血管形成中的致病机理提供基础材料，并为进一步建立视网膜特异性表达VEGF转基因鼠模型建立了基础。(中华眼底病杂志,2005,21:106-109)     

黄芪天麻对老年心脑血管疾病患者发中微量元素影响的研究

黄芪、天麻中含有较丰富地人体必需微量元素。而人体内的微量元素含量的多少与生长、发育、健康、衰老及心脑血管疾病的发生和防治等均有密切关系。我们检查了３９例健康老年人及９８例患有心脑血管疾病老年人头发中的６种微量元素，上述病人在服用黄芪、天麻前后头发中微量元素的变化。研究结果表明黄芪、天麻对调节体内微量元素的含量，预防和治疗心脑血管疾病有较好地效果。     

医院老年病房四季全膳食中十五种矿物质元素的测定及评价

对我院老年病房四季１１２份全膳食中的１５种元素（钾、钠、钙、磷、镁、锌、铜、铁、锰、硒、铝、钴、铬、钼、镍）进行了实测。结果表明：钾的平均日摄入量为１８０９＋３８ｍｇ，接近ＲＤＡ值的下限；钠的平均日摄入量为５９７２±１４６ｍｇ远高于１１００～３３００ｍｇ的供给标准。钙、磷、镁的日摄入量已达到了ＲＤＡ值的标准。全膳食中锌、铜的含量分别为１１．６８±０．１９ｍｇ和１．７２±０．１０ｒｎｇ，略低于ＲＤＡ值（１５ｍｇ及２～３ｍｇ）的标准，锰含量为３．４１±０．０９ｍｇ硒为９８±２．５μｇ，都处于安全与适宜摄入范围（２．５～５．０ｍｇ和５０～２００μ９）。钴、铬、钼、镍均为人体必需的微量元素、目前我国尚未建立有关营养供给量标准。该项研究结果可为今后确定其推荐供给量标准提供必要的基础资料．     

A Drosophila melanogaster hobo-white+ vector mediates low frequency gene transfer in D. virilis with full interspecific white+ complementation

Transformation of a Drosophila virilis white mutant host strain was attempted using a hobo vector containing the D. melanogaster mini-white⁺ cassette (H[w⁺, hawN]) and an unmodified or heat shock regulated hobo transposase helper. Two transformant lines were recovered with the unmodified helper (HFL1), one containing only the white⁺ marked vector, and a sibling line containing the vector as well as an HFL1 helper integration. An approximate total transformation frequency of 1% is deduced. A high frequency of wing and eye morphology mutants were also observed, suggesting that hobo may have mobilized a related element in D. virilis. The data reaffirms a relatively low transformation vector activity for the hobo transposon in D. virilis; however, nearly full interspecific expression of the white⁺ marker supports its possible function in other species as well.     

Response of V beta 8.1+ T cell clones to self Mls-1a: implications for the origin of autoreactive T cells.

Clonal deletion and anergy are two major mechanisms of self-tolerance. However, the molecular mechanisms underlying clonal deletion and anergy, as well as the threshold of TCR affinity/avidity required for these processes, are not known. Expression of the V beta 8.1 TCR correlates with the reactivity of the T cells to the minor lymphocyte stimulating locus-1a (Mls-1a) and T cells expressing this TCR are deleted in the thymus of Mls-1a mice. Similarly, in TCR V beta 8.1 transgenic mice, the number of CD4+CD8-T cells is reduced in Mls-1a mice. However, small numbers of CD4+CD8-T cells remain in the periphery of adult Mls-1a transgenic mice. We have generated T cell clones from TCR V beta 8.1 transgenic mice by stimulation of lymph node T cells with C57BL/6 alloantigens. Interestingly, CD4+CD8-V beta 8.1+ clones isolated from the transgenic mice of Mls-1a background responded to the self-antigen Mls-1a, to which they did not respond in primary assay. Reactive patterns of the clones were compared with clones derived from Mls-1b mice. Proliferation and cytokine production of the clones from Mls-1a mice to the self-antigen Mls-1a were generally reduced when compared with clones from Mls-1b mice. More importantly, T cell clones from Mls-1a mice required more Mls-1a antigen for their activation, and were more susceptible to the inhibitory effects of anti-CD4 antibody on the proliferative responses to Mls-1a than those from Mls-1b mice. These results suggest that the T cell receptor on clones derived from Mls-1a mice have functional but reduced affinity/avidity for self-antigen Mls-1a.     

心脏病患者血清七种元素含量与血压及生化指标的关系

我们检测了心脏病患者血清微量元素Ｚｎ、Ｃｕ、Ｆｅ和常量元素Ｋ、Ｎａ、Ｃａ、Ｍｇ的含量，并与血压及生化指标作相关分析。结果发现心脏病患者Ｋ和Ｃａ／Ｍｇ显著低于对照组，而Ｎａ、Ｍｇ、Ｚｎ、Ｆｅ和Ｎａ／Ｋ、Ｚｎ／Ｃｕ则显著高于对照组。Ｋ、Ｃａ／Ｍｇ与血压呈负相关，而Ｎａ、Ｚｎ、Ｆｅ、Ｚｎ／Ｃｕ和Ｎａ／Ｋ与血压呈正相关；Ｆｅ与红细胞压积（Ｈｃｔ）呈正相关；Ｚｎ／Ｃｕ与Ｈｃｔ、甘油三酯、极低密度脂蛋白均呈正相关，而与高密度脂蛋白呈负相关。
