Tissue‐engineered dermal substitutes represent a promising approach to improve wound healing and provide more sufficient regeneration, compared with current clinical standards on care of large wounds, early excision, and grafting of autografts. However, inadequate regenerative capacity, impaired regeneration/degradation profile, and high cost of current commercial tissue‐engineered dermal regeneration templates hinder their utilization, and the development of an efficient and cost‐effective tissue‐engineered dermal substitute remains a challenge. Inspired from our previously reported data on a pullulan/gelatin scaffold, here we present a new generation of a porous pullulan/gelatin scaffold (PG2) served as a dermal substitute with enhanced chemical and structural characteristics. PG2 shows excellent biocompatibility (viability, migration, and proliferation), assessed by in vitro incorporation of human dermal fibroblasts in comparison with the Integra® dermal regeneration template (Control). When applied on a mouse full‐thickness excisional wound, PG2 shows rapid scaffold degradation, more granulation tissue, more collagen deposition, and more cellularity in comparison with Control at 20 days post surgery. The faster degradation is likely due to the enhanced recruitment of inflammatory macrophages to the scaffold from the wound bed, and that leads to earlier maturation of granulation tissue with less myofibroblastic cells. Collectively, our data reveal PG2's characteristics as an applicable dermal substitute with excellent dermal regeneration, which may attenuate scar formation. 相似文献

[目的]观察载骨髓间充质干细胞(Bone marrow mesenchymal stem cells,MSCs)明胶微球磷酸钙骨水泥(Calcium phosphate cement,CPC)的理化性质,探究新型CPC的制备。[方法]制备载MSCs的明胶微球后,分别以0%(A),2.5%(B),5%(C),10%(D)的质量比(w/w)与CPC复合。测定其抗压强度、初凝时间、孔隙率和大孔率等理化性质,进一步检测MSCs在CPC中的成长情况。[结果]在各组CPC中MSCs均生长良好,且与空白对照组比较,随着明胶微球比例的增大,各组CPC的抗压强度逐步降低,初凝时间延长,孔隙率增大,大孔率增高(P<0.05)。[结论]载MSCs的明胶微球与CPC的质量比为5%时,CPC的理化性质最为理想,细胞生长良好,此结果为下一步体内实验奠定了坚实的基础。 相似文献

Objectives: Artificial colloids, frequently used to prevent hemorrhagic shock in children, impair blood coagulation. To determine the impact of acute isovolaemic hemodilution with artificial colloids on clot formation, we conducted an experimental study in a pediatric animal model. Methods: Fifteen piglets underwent hemorrhage by withdrawing 40 ml·kg?1 of blood volume in steps of 10 ml·kg?1 each within 1 hour. After each withdrawal, the blood loss was randomly compensated by administering 4% gelatin (GEL) or hydroxylethyl starch 130/0.42 (HES) in a ratio of 1 : 1, or isotonic crystalloid solution (ICS) in a ratio of 1 : 4 for isovolaemic hemodilution. Quality of clot formation and platelet function was measured using Thrombelastometry (ROTEM®) and Multiple electrode impedance aggregometry (Multiplate®) after 10, 20, and 40 ml·kg?1 blood replacement. Results: Moderate hemodilution (10–20 ml·kg?1 blood replacement) caused no significant differences among groups (e.g. INTEM®‐MCF after 20 ml·kg?1 blood replacement (ICS vs GEL vs HES, P > 0.05). Profound hemodilution with 40 ml·kg?1 blood replacement showed a significant difference between ICS and both colloids (P < 0.05), but no significant differences between GEL and HES. Conclusions: Impairment of clot formation by moderate isovolaemic hemodilution did not significantly differ between ICS, GEL, and HES. Profound hemodilution of more than 50% of the estimated blood volume with GEL and HES caused significant impairment of clot formation in comparison to ICS and has to be considered when using high amounts of these synthetic colloids. 相似文献

目的 研究粉尘螨变应原明胶微球经口服免疫小白鼠后的脱敏疗效。 方法 以粉尘螨变应原明胶微球 (含粉尘螨变应原 2× 10 -5mg )口服免疫小白鼠 ,以ELISA法检测 3 5d内血清特异性IgG水平变化。并以腹腔注射相同剂量的粉尘螨变应原浸液的小白鼠为对照实验 ,以口服不含粉尘螨变应原的空白明胶微球的小白鼠为空白实验。 结果口服粉尘螨变应原微球的小白鼠实验组可产生高水平的血清抗体 ,在给药后第 14d抗体最高值为 2 .45× 10 5Au/ml(Ar bitraryunitspermilliliter) ,高于腹腔注射相同剂量的粉尘螨变应原浸液小白鼠的最高抗体水平 (4 .2 7× 10 4Au/ml)。结论 在给予相同剂量粉尘螨变应原的前提下 ,明胶微球口服制剂的免疫效果优于变应原浸液腹腔注射剂。 相似文献

The treatment of the hypertrophic obstructive cardiomyopathy (HOCM) by percutaneous transluminal septal myocardial ablation (PTSMA) with ethanol injection has greatly improved in the last years. This report describes the case of a patient with symptomatic drug-refractory HOCM who underwent an unsuccessful attempt to thrombose the septal artery by PTSMA with alcohol. It has therefore been decided to use small absorbant gelatin sponge (AGS) particles, obtaining immediate thrombotic occlusion of the artery and excellent hemodynamic results with immediate and permanent disappearance of the gradient. The patient progressed satisfactorily and displayed lower than average creatine kinase levels in comparison to the rest of our PTMSA patients. After 2 years of follow-up he still remains asymptomatic and without any gradient. These results suggest that PTSMA with AGS could be a valuable alternative treatment of HOCM. 相似文献

目的 探讨不同介入栓塞方案治疗肝血管瘤的疗效及安全性.方法 收集2010年8月-2013年4月在济宁市第一人民医院介入放射科接受治疗的肝血管瘤患者的临床资料.入组患者根据栓塞方法不同分为明胶海绵微粒+平阳霉素碘化油乳剂组(观察组)和单纯的平阳霉素碘化油乳剂组(对照组),比较两组患者的治疗效果、术后并发症、下床时间、住院时间以及治疗前后总胆汁酸(total bile acid,TBA)的变化.结果 本研究共纳入研究对象60例,其中观察组和对照组各30例.观察组患者治疗有效率和显效率均优于对照组(x^2=6.734,P=0.034);两组并发症发生率差异无统计学意义(x^2=0.098,P=0.75),观察组下床时间(t=19.39,P<0.01)、住院时间(t=7.257,P<0.01)均显著少于对照组;治疗前两组患者TBA水平差异无统计学意义(t=0.073,P=0.94),治疗后3d观察组TBA水平低于对照组(t=3.195,P<0.01).两组患者的谷丙、谷草转氨酶、白蛋白、总胆红素、凝血酶原时间等指标均无统计学差异(P均>0.05).结论 明胶海绵微粒加平阳霉素碘化油乳剂相对于单纯平阳霉素碘化油乳剂治疗肝血管瘤,不仅有效、安全,而且患者术后恢复速度更快. 相似文献
