目的研究TPO基因修饰的基质细胞对CD34+造血细胞向巨核系细胞定向增殖分化的影响.方法将TPO cDNA构建到pBabe/pura中,通过"乒乓转染法"获得产高滴度逆转录病毒载体的包装细胞;利用所获的病毒上清转染基质细胞HFCL,用Northern blot检测细胞中TPO基因的表达,并利用TPO依赖株检测上清中的TPO活性;以未转基因的HFCL为对照,将CD34+造血细胞在TPO基因修饰的HFCL支持下扩增,用流式细胞仪检测扩增细胞中表型为CD34+CD41+和表型为CD41-CD61+细胞的比例.结果基质细胞HFCL能够有效表达外源性TPO基因,在HFCL/TPO培养上清中有0 75 ng/ml水平的TPO活性;并且在HFCL/TPO支持下,CD34+造血细胞经过7 d扩增后,CD34+CD41+细胞的比例为(13.7±2.0)%,HFCL为(6.5±1.8)%(P<0.01);CD41+CD61+细胞的比例为(11.9±0.7)%,略高于HFCL的(10.6±1.5)%(P>0.05).结论基质细胞能够有效表达外源性TPO基因和其蛋白,而且TPO基因修饰的基质细胞能显著促进CD34+CD41+巨核祖细胞的扩增,但对较成熟的CD41+CD61+巨核细胞影响不明显.  相似文献

目的 探索促血小板生成素(Thrombopoietin,TPO)分子间相互作用及其存在位置。方法 构建了含有不同TPO区段的酵母双杂交系统融合表达质粒,转化酵母菌SFY526,分析β-关乳糖苷酶活性以推测TPO分子间的相互作用。结果 全长TPO-pGBT9与全长TPO-pGAD424、全长TPO-pGAD424与TPO(N)-pGBT9与TPO(N)-pGAD424之间有相互作用,全长TPO-pGBT9与TPO(C)-pGAD424、TPO(N)-pGBT9与TPO(C)-pGAD424、TPO(C)-pGBT9与TPO(C)-pGAD424之间无相互作用。结论 自然状态下TPO在N端结构区可能存在相互作用,C端结构域不参与此过程。  相似文献

BACKGROUND: The cytokine network in peripheral blood progenitor cell (PBPC) grafts may affect hematopoietic reconstitution or the risk of postransplant relapse of malignant disorders through effects on normal progenitor cells or contaminating malignant cells. Whether thrombopoietin (TPO), SCF, and platelet-secreted mediators are parts of this network was investigated. STUDY DESIGN AND METHODS: Peripheral blood and PBPC plasma samples were collected consecutively from patients with malignant disorders who underwent PBPC harvest. Blood samples were collected immediately before and after apheresis. Patients underwent mobilization by chemotherapy plus G-CSF, except for one patient who received only G-CSF. Plasma levels were also determined for healthy controls. RESULTS: PBPC grafts had greater levels of platelet-secreted platelet factor 4 (PF4), beta-thromboglobulin, and platelet-derived growth factor isoform AB, as compared with venous levels in patients and controls. Although platelet and PF4 levels in autografts were significantly correlated, the graft:blood ratio was higher for PF4 than for platelets. In both the patients' blood and the autografts, TPO levels were increased from the levels in normal controls. Blood and graft levels of SCF were within the normal range. CONCLUSION: The cytokine network of PBPC autografts includes increased levels of TPO and several platelet-derived mediators.  相似文献

Background

Idiopathic pleuroparenchymal fibroelastosis (IPPFE) is a rare type of interstitial pneumonia characterized by fibroelastosis. Patients with IPPFE as well as idiopathic interstitial pneumonia often have autoimmune diseases, which sometimes coincide with hypothyroidism (HypoT). However, there have been no reports on the association between IPPFE and HypoT. The purpose of this study was to evaluate the correlation between IPPFE and HypoT. We also examined the pathological features of the thyroid glands from autopsied cases.

Methods

Thirteen patients diagnosed with IPPFE from among 255 consecutive cases of idiopathic interstitial pneumonia were included in this study; pertinent data were obtained from our hospital’s clinical library. We examined the prevalence of HypoT and compared the clinical, radiological, and pathological features between the patients with and those without HypoT. Histological analyses of the lungs and thyroid glands were performed in 4 and 3 cases, respectively.

Results

HypoT was identified in 7 of 13 patients (53.8%). Sex, body mass index, survival time, and laboratory test results were not significantly different between patients with and those without HypoT. Radiological and pathological lung findings were similar between both groups of patients. Thyroid gland histology demonstrated perifollicular or interlobular fibrosis without inflammation in all three cases, including a euthyroid case.

Conclusions

Although we only analyzed a small number of IPPFE cases, HypoT was prevalent among all of them. Characteristic fibrosis in the thyroid gland was observed even in a euthyroid case. Therefore, patients with IPPFE may potentially have thyroid gland dysfunction through a common pathogenesis in both organs.  相似文献

This study is a meta-analysis of available literature examining the effect of an observer on cognitive task performance. Of the 210 identified relevant articles, 62 met inclusion criteria yielding a final sample with 4405 individuals (2496 observed cases, 1909 not observed). The overall effect size was significant (d?=??0.24), i.e., the presence of an observer was associated with poorer performance. However results were moderated by the effect size calculation method, cognitive domain, visibility of the observer, and number of observers. Attention, learning/memory, and delayed recall tasks were most adversely impacted by the presence of an observer.  相似文献

Objectives

To determine the degree of conversion (DC) over 48 h post-curing of resin mixtures containing trimethylbenzoyl-diphenylphosphine oxide (TPO) initiator cured by a polywave or a monowave LED light-curing unit (LCU).

Methods

In resin mixtures based on equal weight percent (wt%) of BisGMA and TEGDMA the following initiators were added: 0.2 wt% camphorquinone (CQ) + 0.8 wt% ethyl-4-dimethylaminobenzoate (EDMAB) (Group 1); 1 wt% TPO (Group 2) and 0.1 wt% CQ + 0.4 wt% EDMAB + 0.5 wt% TPO (Group 3). Half of the samples in each group (n = 5) were cured using a polywave (bluephase® G2, Ivoclar Vivadent) or a monowave LED LCU (bluephase®, Ivoclar Vivadent). The DC was measured using micro-Raman spectroscopy within 5 min and then 1, 3, 6, 24 and 48 h post-irradiation. The data were analysed using general linear model and two-way ANOVA for the factors ‘time’, ‘material’, ‘surface’ and ‘LCU’ at α = 0.05.

Results

The initial DC values obtained upon light curing remained similar over a 48 h period. bluephase® G2 produced the highest DC in Group 2 followed by Group 3, and Group 1. bluephase® resulted in the highest DC in Group 1, followed by Group 2 and Group 3 (p < 0.05).

Conclusions

Unfilled resin materials containing both TPO– and CQ–amine initiators are effectively cured using bluephase® G2. Resin mixture with the same wt% of initiators is better cured when TPO is the only initiator, compared to CQ–amine only or combined TPO and CQ–amine system. After initial light cure, no additional conversion of uncured monomers was detected in an unfilled resin material over 48 h at 37 °C.  相似文献

Objective

To evaluate the effect of photo-initiator on the degree of conversion (DC) and elution of Bis-GMA and HEMA for 8 one-step adhesive formulations.

Methods

We used Scotchbond Universal (‘SBU-CQ/amine_4.0’, 3M ESPE), containing about 2 wt% camphorquinone (CQ) and 2 wt% ethyl-4-dimethylamino benzoate (EDMAB), an experimental ‘SBU-TPO_2.1’ version, containing 2.1 wt% diphenyl(2,4,6-trimethylbenzoyl)phosphine oxide (TPO), and 6 experimental LUB adhesives (Kuraray Noritake), namely ‘LUB-CQ/amine_0.7’, ‘LUB-CQ/amine_1.4’, ‘LUB-CQ/amine_4.0’, ‘LUB-TPO_0.35’, ‘LUB-TPO_0.7’ and ‘LUB-TPO_2.0’, respectively containing 0.35 wt% CQ and 0.35 wt% EDMAB, 0.7 wt% CQ and 0.7 wt% EDMAB, 2.0 wt% CQ and 2.0 wt% EDMAB, 0.35 wt% TPO, 0.7 wt% TPO, and 2.0 wt% TPO. DC was measured using micro-Raman spectroscopy. Additional specimens were immersed in ethanol for 24 h to determine the elution of Bis-GMA and HEMA using HPLC.

Results

DC of the respective SBU and LUB adhesives was alike at high photo-initiator concentration. At low concentration, TPO was significantly more efficient than CQ/amine (LUB adhesives only). A statistically significant positive photo-initiator concentration effect on DC was noted for both CQ/amine and TPO (LUB adhesives only). A statistically significant inverse photo-initiator concentration effect on HEMA elution was noted for both the CQ/amine- and TPO-containing LUB adhesives. A significantly strong correlation was found between DC and Bis-GMA elution (R2 = 0.744, p = 0.026), and between DC and HEMA elution (R2 = 0.913, p = 0.002) for the LUB adhesives.

Significance

The photo-initiator kind and concentration affect DC and the Bis-GMA/HEMA elution. TPO can be used as an alternative photo-initiator for CQ/amine.  相似文献

目的 探讨TPO蛋白在甲状腺乳头状癌与结节性甲状腺肿伴乳头状增生鉴别诊断中的作用。方法 用免疫组化EnVision法测定50例甲状腺乳头状癌及50例结节性甲状腺肿伴乳头状增生的TPO表达情况。结果 50例甲状腺乳头状癌中有48例TPO(-),2例(+);50例结节性甲状腺肿伴乳头状增生中TPO均(+)。TPO在甲状腺乳头状癌与良性乳头状增生鉴别诊断中的评价指标:敏感度为96%,特异度为100%,准确度为98%,阳性预测值为100%,阴性预测值为96.15%。结论 TPO蛋白可作为鉴别甲状腺乳头状癌与良性乳头状增生的重要参考标记物。  相似文献
