Activation of hepatic guanylate cyclase by nitrosyl-heme complexes: Comparison of unpurified and partially purified enzyme

The objective of this study was to evaluate the effects of several agents on activation of both unpurified and partially purified hepatic soluble guanylate cyclase by performed NO (nitric oxide or nitrosyl)-heme complexes. Guanylate cyclase was activated by NO complexes of the heme compounds, hematin, hemoglobin, myoglobin, catalase and cytochrome c, and also by the reaction product of NO and ferredoxin, a non-heme, iron sulfur electron transfer protein. NO-lipoxygenase, which contains non-heme iron, did not activate guanylate cyclase. NO-heme complexes activated unpurified enzyme almost equally well in the presence of either Mg²⁺ or Mn²⁺. However, activation of purified (350- to 750-fold) guanylate cyclase was markedly greater with Mg²⁺ than with Mn²⁺. At concentrations that did not alter basal enzymatic activity, Ca²⁺ markedly inhibited guanylate cyclase activation in the presence of Mg²⁺ but not of Mn²⁺. Hemoproteins inhibited activation of unpurified and purified enzyme by NO-heme complexes, and increasing the concentrations of the latter overcame the inhibition. Gel filtration studies indicated that uncomplexed and NO-complexed hematin bind to common or adjacent sites on guanylate cyclase. Whereas dl-dithiothreitol enhanced activation, ferricyanide, cystine, o-iodosobenzoic acid and ethacrynic acid inhibited activation of guanylate cyclase by NO-heme complexes. The data indicate that the effects of these diverse agents on guanylate cyclase activation by preformed NO-heme are similar to their effects on enzyme activation by NO and nitroso compounds, both of which readily form NO-heme complexes. Therefore, the effects of these diverse agents may be on guanylate cyclase rather than on NO-heme formation.     

Single Aortotomy for Multiple Aortocoronary Saphenous Vein Bypass Grafts

A new technique for the proximal anastomoses of saphenous vein grafts in coronary artery bypass operations is described. This technique has several advantages over existing methods and has been shown to be safe and satisfactory in 389 consecutive patients operated on between May, 1978, and May, 1981.     

Arteriosclerosis related deaths in severe neurosis

1,482 previous in-patients with the diagnosis of neurosis from three hospitals in Birmingham, U.K., were followed up after a mean of 10.9 years. 91% of the sample were traced and 139 patients were found to have died. When those causes of death from the International Classification of Diseases indicating arteriosclerosis were summed, there was a highly significant increase of death from arteriosclerosis in the sample compared with expected mortality. Death from accident, poisoning and violence was also highly significantly increased, especially self-inflicted death. There was also an increase in deaths from respiratory causes.     

Physiological responsivity of normal and sexually dysfunctional women during erotic stimulus exposure.

The purpose of this investigation was to determine how sexually dysfunctional and normal women differ in their physiological responsivity during, and subjective responsivity shortly after, exposure to an erotic stimulus. Utilizing analysis of covariance, subjects were equated on physiological basal measures as well as other relevant demographic and sexual activity measures. Normal subjects showed increases in diastolic blood pressure and vaginal capillary engorgement, but the groups did not differ on electrodermal activity, systolic blood pressure, heart rate, and subjective ratings of sexual or anxiety arousal. There were significant positive correlations for the combined groups between vaginal capillary engorgement and Sexual Arousability Inventory scores, awareness of physiological changes during sexual activity, day in menstrual cycle, and frequency of intercourse.     

A controlled study of inpatient vs. outpatient treatment of delinquent drug abusing adolescents: One year results

Seventy-four delinquent drug abusing adolescents were randomly assigned to inpatient or outpatient treatment. One year results on several measures of social functioning as well as MMPI scores are analyzed. At one year, no significant differences in outcome are noted on these measures. Implications of these findings are discussed as well as possible changes which may occur at a subsequent, two year, follow-up.     

Nasotracheal intubation in diphtheria

Nasotracheal intubation can be used effectively for the relief of upper airway obstruction in dephtheria. It has many advantages over tracheostomy; it avoids an operation and it eases anxiety of the parents. Of 57 patients so treated, 50 recovered without injury to the larynx in any patient.     

A danger of haloperidol therapy in children.

Attention is directed to the potential danger of therapy with haloperidol in children. Its value in reducing the excessive movements of Sydenham chorea must be weighed against the usual tendency of this disorder to remit spontaneously and the occasional tendency of the drug to cause castastropic adverse reaction     

Traumatic dislocation of the hip with ipsilateral femoral shaft fracture: a method of treatment

Two cases of posterior fracture-dislocation of the hip associated with an ipsilateral femoral shaft fracture are presented with a technique of reduction and stabilization which allows for closed treatment of both injuries. This approach is advantageous in the severely injured patient.
